Cytoplasmic RNA was isolated from varicella-zoster virus (VZV)-infected cells. By oligo(dT)-cellulose chromatography, the RNA was separated into polyadenylated, poly (A)+, and nonpolyadenylated, poly (A)∼, fractions. RNA blot hybridization was employed to detect and map VZV transcripts. As VZV infection cannot be coordinated, cytoplasmic RNA was isolated from VZV-infected cells when the cells showed extensive cytopathology. Therefore, while the VZV transcripts represented heterogeneous temporal classes, it may be assumed that late VZV RNA predominated. At least 41, and as many as 67 (depending on DNA probe overlap), VZV polyadenylated transcripts have been identified. Two modes of assembly of murine cytomegalovirus (MCMV) were observed in cultured mouse embryo fïbroblasts, generating two morphologically different types of viral particles: monocapsid virions and multicapsid virions. The assembly of nucleocapsids appeared to be the same for both types of morphogenesis. Three successive stages of intranuclear capsid formation could be distinguished: capsids with electron-lucent cores, coreless capsids, and capsids with dense cores. Some of the capsids were enveloped at the inner nuclear membrane to form monocapsid virions, which were first detectable in the perinuclear cisterna. Other capsids left the nucleus via nuclear pores and usually entered cytoplasmic capsid aggregates that received an envelope by budding into extended cytoplasmic vacuoles, thereby forming multicapsid virions. Since the formation of multicapsid virions is not restricted to cell culture conditions and also occurs in vivo in immunosuppressed mice, multicapsid virions may play a role in the pathogenesis of cytomegalovirus infection

The host range of the inscet virus Autographa californica nuclear polyhedrosis virus (AcMNPV) was examined. AcMNPV could not initiate a productive infection in frog, turtle, trout, or codling moth cell lines. After exposure to AcMNPV, neither viral DNA nor RNA synthesis could be detected in these cell lines as assayed by nucleic acid probe hybridization. Entry of AcMNPV nucleocapsids into the cytoplasm and viral DNA into the nucleus, however, was as efficient in the nonpermissive cell lines as it was in a permissive insect cell line. The data suggest that the block in AcMNPV infection in these nonpermissive cell lines is at a stage subsequent to viral DNA entry into the nucleus.

摘要:
DNA of the human PLC/PRF/5 hepatoma cell line can induce the appearance of colonies on soft agar after transfection of BK-BK cells (BALB/c mouse kidney cells partially transformed by BK virus). Blot hybridization analyses indicate that the transformants contained the hepatitis B virus DNA sequence. This sequence disappeared during serial passage of transformants. The amplification and rearrangement of the integrated BK virus genome appears to be specifically associated with transformation.

The RNAs of two independently isolated strains of lactate dehydrogenase-elevating virus (LDV), which differ antigenically and in neurovirulence for C58 mice, were isolated and Tl RNase fingerprinted. Of about 30 unique Tl oligonucleotides, 27 seemed to be common for both strains of LDV, whereas 2 or 3 oligonucleotides were unique for each strain. In other physical and biological properties, such as virion density, molecular weights of their structural proteins, interaction with mouse anti-LDV IgG, and replication in primary cultures of peritoneal macrophages from various mouse strains, the two strains of LDV were indistinguishable. The Tl RNase patterns and the affinity of LDV RNA for oligo(dT) indicated that it contained poly-A.
