Electron Microscopic Studies on the Crystallization of Adenovirus Hexon Capsomers |

Highly purified soluble adenovirus type 1 hexons were crystallized by dialyzing against 0.5 M acetate buffer, pH 4.5, for 3 days. Crystals of tetrahedral shape were formed after storage at 4° within 1–10 days and ranged in size from 0.01 to 0.1 mm. In the early phase of crystallization, diffusely dispersed hexons were observed by electron microscopy. Later, continuous monolayers of two-dimensional hexon paracrystalline arrays developed. Direct analysis of the electron micrographs and their optical diffraction patterns showed that the structure of the dense two-dimensional crystalline arrays of hexon capsomers was characterized by slightly skew and irregular hexagonal packing. Rhinovirus type 14 (RV14) genomic RNA, but not mengovirus or coxsackievirus B3 genomic RNA, fragmented to acid-insoluble RNA species during incubation in aqueous buffers at 60°. Incubation of RV14 genomic RNA in high concentrations of dimethyl sulfoxide at 60° did not result in fragmentation, suggesting that fragmentation was not a result of preexisting nicks. RV14 RNA that was treated with proteinase K prior to incubation at 60° did not undergo fragmentation. The association of protein(s) with RV14 virion RNA was demonstrated by radiolabeling with amino acids. 摘要:
Radial hemolysis-in-agarose gel (HIG) with members of the genera Aiphavirus and Flavivirus is described. The test can be reproduced with goose and sheep erythrocytes sensitized at the optimal pH for each virus. Human (O group), rooster, guinea pig, mouse and hamster erythrocytes were found to be unsuitable. The titer of immune ascitic fluids in HIG coincides with the titer in hemagglutination inhibition, investigation of convalescent sera after laboratory infection suggests the possibility of HIG application for serologic diagnosis of arbovirus infection.

A semiquantitative infectious bioassay, described by Sairenji and Hinuma for the measurement of Epstein-Barr virus (EBV) receptors, was adapted to study the adsorption of the lymphotropic New World herpesvirus ateles (HVA) to established lymphoid lines and lymphocytes of human versus marmoset origin. All marmoset T-cell lines [HVA- or HVS (herpesvirus saimiri)-carrying] had a high adsorbing capacity. EBV-carrying marmoset or human B-cell lines, one EBV-negative human B-lymphoma line, and one human myeloid leukemia-derived line failed to adsorb detectable virus. Two human T-cell-derived cell lines of acute lymphatic leukemia origin showed moderate but significant adsorption, and the same was true for un-fractionated marmoset lymphocytes. Human peripheral blood did not adsorb, before or after fractionation, whereas normal human thymocytes showed moderate but significant adsorption. It is concluded that HVA receptors are present on human T cells, probably belonging to certain subclasses.

A simple tissue microculture technique was developed for the titration of a cytoplasmic polyhedrosis virus (CPV) from Euxoa scandens. The procedure was similar to the 50% tissue culture infectious dose assay, but a single infected cell, detected by the presence of cytoplasmic polyhedra, was scored rather than the degeneration of cell monolayers. The filtration of CPV suspensions resulted in decreased virus titers under certain conditions. This microculture assay was used to determine the effect of cell disruption methods on virus yields. Sonication of infected cells was more efficient than freeze-thawing for the recovery of nonoccluded virus.

Six stomatitis papulosa and three Orf virus strains were compared by serology and by DNA restriction analysis. A neutralization kinetic study revealed extensive serological cross-reactivity between all strains, but did not allow their classification. Restriction analysis of viral DNAs revealed two distinct groups among the stomatitis papulosa strains while the Orf virus strains formed a third, more heterogeneous group. The large heterogeneity of restriction patterns of parapoxvirus DNAs as compared to those of orthopoxviruses is discussed.

Seedlings oï Kennedya rubicunda were manually inoculated with Kennedya yellow mosaic virus and planted along with comparable virus-free seedlings in a coastal bushland site where the plant and virus were common. Virus infection decreased the growth rate of the plants by about one-third; however, virus-free plants disappeared more than twice as quickly as infected plants as a result of grazing by herbivores. The distribution of related sequences between the genomes of pseudorabies virus (PRV) and herpes simplex virus, types 1 (HSV-1) and 2 (HSV-2), was determined. Approximately 7% of the sequences in PRV are shared by HSV-1 and HSV-2 DNAs. By means of the Southern blot technique, it was found that the homologous sequences are not sequestered in one region but are distributed throughout the PRV genome. HSV-1 and HSV-2 have the greatest homology with the long unique region of PRV DNA and the least with the inverted repeat regions of the molecule. HSV-1 DNA also has few sequences homologous to the short unique region of the PRV genome; HSV-2 DNA hybridizes well to this region. There was no homology of HSV-1 or HSV-2 DNAs with the extreme ends of the inverted repeat regions of PRV DNA.

Rat interferon (IF) induced a moderate and persistent elevation of adenosine 3’, 5’-cyclic monophosphate (cAMP) after 2 h of contact with rat embryo fibroblasts (REFs), whereas in rat glial tumoral C6 cells IF did not enhance cAMP. Noradrenaline potentiated the antiviral state of REFs treated for 2 h or more with IF and increased cAMP in these cells, but it did not potentiate the IF-induced antiviral state of C6 cells. A series of herpes simplex virus type 2 (HSV-2)-transformed hamster cells has been examined using the technique of lactoperoxidase-catalyzed cell surface protein iodination. The amount of cell surface fibronectin (LETS protein) was reduced after HSV-2 transformation, and this reduction correlated with increased tumor-forming ability.
