摘要:
Murine lymphocyte hybridomas which produce neutralizing or non-neutralizing monoclonal antibodies to different type 1, 2 or 3 poliovirus strains were isolated. The majority of these monoclonal antibodies reacted with antigenic determinants present on different polio-virus strains of the same type. However, hybridomas producing monoclonal antibodies specific for each of the three Sabin vaccine strains were also generated.

Equine infectious anemia virus (EIAV) was successfully inoculated onto cell cultures of canine and feline origin, resulting in chronic infections in these cultures. Infection of equine cell cultures, which were the previous sole in vitro source demonstrated for virus production, was also performed for comparative purposes. Determination of the nature of the virus produced in the heterologous as well as the equine cells was accomplished in several ways. SDS-PAGE of purified virus from the different cell lines indicated very similar protein composition. Immunological identity was observed in gel diffusion tests employing an antiserum to the major core protein (p24) of equine-derived EIAV. Competition radioimmunoassays also indicated similar antigenicity in the viruses derived from the several cell lines. Strong relatedness was further demonstrated by hybridization of viral RNAs to EIAV complementary DNA. These data indicate that EIAV has an amphotropic cell culture host range and that the viruses isolated from the permissive lines were similar.

L-Canavanine, a naturally occurring analog of arginine, effectively inhibited the morphogenesis of herpes simplex virus (HSV) and human cytomegalovirus (HCMV) when added at the time of infection, but allowed the expression of several cytopathic changes. Exposure to canavanine at progressively later times ultimately led to qualitatively normal virion maturation. Under no conditions were morphologically aberrant viral particles observed. However, HSV-infected cells treated at 3 h postinfection or later did contain distinctive cytoplasmic inclusions resembling HCMV dense bodies. Recovery experiments showed that HCMV-infected cells exposed to canavanine for 1–5 days could support normal viral morphogenesis when washed free of this agen

The role of human cytomegalovirus (CMV) was examined according to serological patterns in 37 patients with adenocarcinoma of the colon (ACC). The sera were examined for the presence of IgG antibodies by the immunoperoxidase antibody to membrane antigen (IPAMA) method and by the complement-fixation (CF) test. Antibody determinations were also performed by the IPAMA method for three other members of the herpesvirus group: Epstein-Barr virus (EBV), herpes simplex virus (HSV) and varicella-zoster virus (VZV). Comparison groups included normal subjects, ACC patients treated with chemotherapy, and patients operated on for benign diseases. No significant difference was found in the geometric mean titers (GMTs) for CMV and the other herpesviruses in the sera of nontreated ACC patients when compared with the control groups. However, a significantly elevated antibody titer to CMV was found in chemotherapy-treated ACC patients by both the IPAMA and CF methods. In this group, elevated titers were found by the IPAMA method for EBV and HSV, but not for VZV. The significance of serological studies in elucidating the role of CMV in ACC patients is discussed.

Interferon induction in BSC-1 cells by measles virus isolates previously found to be interferon inducing (IF+) or interferon noninducing (IF-) was studied in different cell types. Lack of induction by the IF– viruses was not due to an inhibitory factor since BSC-1 cells coin-fected with IF+ and IF– stocks produced interferon levels similar to those infected with IF+ stocks alone. Various tests indicated that the phenotypic expression of interferon induction by these isolates was dependent on an infectious virus genome, the cell type tested, and the passage history of the virus. In contrast to the in vitro results, the IF+ phenotype was stable in vivo, whereas passage of the IF+ virus resulted in selection against the IF+ phenotype.
