The intracellular site of virus-specific DNA synthesis was studied in mouse 3T6 cells early after efficient infection with the Harvey strain of murine sarcoma-leukemia virus. Treatment of cells with arabinosyl cytosine at different times after infection indicated that DNA synthesis at 1–4 h was essential for both the subsequent transcription of virus-specific RNA and the formation of progeny virus. Autoradiographic analysis of cells pulse-labeled with tritiated thymidine at different times after infection showed that DNA synthesis begins in the cytoplasm within 1 h after infection, reaches a maximal rate at 2 h, and decreases subsequently. Newly synthesized cytoplasmic DNA is rapidly transported to the nucleus, as indicated by autoradiographs of cells irradiated with ultraviolet light to block cell DNA synthesis, then infected with virus, pulse-labeled with tritiated thymidine and chased with unlabeled thymidine. The viral nature of newly synthesized cytoplasmic DNA was demonstrated by the isolation of labeled DNA from the cytoplasmic fraction at different times after infection that hybridized with viral 60–70S

摘要:
Effect of temperature on growth in monkey kidney (MK) cells of acute hemorrhagic conjunctivitis (AHC) virus was studied. The optimal growth temperature ranged from 32 to 34°, and 39° was nonpermissive. In one-step growth experiments at 33°, the standard strain J 670/71 of AHC virus grew exponentially after a latent period of 2 h, and reached a maximum titer within 6 h post-infection (p.i.). The temperature-shift experiment showed that incubation at 39° during the latent period of the growth cycle had little effect on the subsequent viral growth at 33°, and when the shift-down was carried out at 2 h p. i. or later, viral growth immediately started and reached a maximum. Conversely, viral growth at 33° was immediately stopped by shift-up to the nonpermissive temperature. Hemolytic plaque assays in gel were performed against burro red blood cells (BRBC) conjugated with hepatitis B surface antigen (HBS Ag). On indirect assay, spleen cells from HBS Ag-immunized mice gave 27–68 times more plaques than cells from unimmunized mice. No difference in numbers of plaques was seen between immunized and unimmunized mice when tested against either plain or control-conjugated BRBC. Hemolytic plaque assays can be used to study cells producing antibody to HBS Ag and should help to delineate aspects of the immune response to this viral antigen in chimpanzees and man during acute viral hepatitis, type B, and in the chronic HBS Ag carrier stat

Examination of renal glomeruli from two patients with African Burkitt’s lymphoma revealed host immunoglobulin G (IgG) and the third component of complement (C3), but not fibrinogen or albumin, along the glomerular basement membrane and mesangia in a granular, discontinuous pattern characteristic of immune complex deposits. Elution and subsequent immunologic assays indicated that the IgG bound to the glomeruli contained antibodies to viral capsid antigens (VCA) and early antigens (EA), but not detectable anti-body(s) to complement-fixing nuclear antigens (EBNA) or membrane antigens (MA) of Epstein-Barr virus (EBV). In contrast, serum contained antibodies to VCA, EA, EBNA and MA of EBV. Absorption of Ig obtained from the serum and renal eluate with EBV antigens revealed the presence of EBV antibodies in the renal glomeruli. The sera of 140 female subjects were investigated for precipitating antibody to type 1 and type 2 herpes simplex antigen. 73 of these sera formed a common precipitin line against type 1 and type 2 virus antigen which in every case gave a reaction of identity with type 1 ‘Band II’ antigen. Thus antibody to ‘Band II’, a major structural antigen, was represented in every serum which immunoprecipitated type 1 or type 2 virus
