Exposure of a herpes simplex virus type 1 to UV light resulted in a multi-component survival curve. Plaques formed by the UV-irradiated virus were smaller than those induced by untreated virus. Maximum plaque counts in cultures inoculated with non-irradiated virus were obtained on the 4th day; when using irradiated virus, the plaque count continued to increase during the subsequent 4 days. These findings may reflect host cell repair. A non-linear relationship between plaque count and virus dilution, indicative of multiplicity reactivation, was observed in cultures infected with heavily irradiated virus.

Deoxythymidine kinase (TdR kinase) from rabbit kidney cells infected in vitro with herpes simplex virus type 1 (HSV-1) or type 2 (HSV-2) was further investigated. Enzyme activity induced by HSV-2 was more heat-labile and more sensitive to inhibition by deoxythymidine nucleotides than HSV-1 induced enzyme activity in both crude extracts and partially purified preparations. The thermostability of TdR kinase activity from cells coinfected with HSV-1 and HSV-2 was intermediate between enzymes induced separately. The difference in thermostability between the two enzyme activities was also observed in extracts from 1-β-D-arabinofuranosylcytosine (ara-C) treated infected cells. HSV-1 and HSV-2 induced enzymes also differed in their response to substrate concentration. Gel filtration with Sephadex G-100 revealed the presence of monomer, dimer and aggregated forms of HSV-1 induced enzyme. The molecular weights of the monomer and dimer were estimated to be 58,000 and 97,000, respectively. Such multiple forms were not observed with HSV-2; the molecular weight of the HSV-2 induced enzyme was estimated to be 45,000 to 60,000, depending upon the eluting buffers. HSV-1 induced TdR kinase activity showed a different electrophoretic activity pattern in polyacrylamide gel electrophoresis than did the enzyme induced by HSV-2

Eighteen temperature-sensitive (ts) mutants of herpes simplex virus type 1 were studied with regard to their ability to synthesize virus-induced thymidine kinase (TK) at permissive (34°) and nonpermissive (39°) temperatures. Five mutants were TK– at both temperatures (TK– mutants), 10 were TK+ at both temperatures (TK+ mutants) and 3 showed reduced TK activity at 39° but not at 34° (TK± mutants). Two of the 3 TK± mutants were ts in the production of TK at 39° while the third mutant synthesized a ts TK at both temperatures. Polyacrylamide gel electrophoresis of extracts of both uninfected and TK-mutant-infected cells exhibited one major and one minor peak of TK activity, whereas extracts of wild-type (WT) virus-infected cells revealed at least one additional major peak of TK activity. 摘要:
Chemical analysis of Semliki Forest virus grown in BHK21 cells revealed 6.3 % RNA, 12.2 % nucleocapsid protein, 44.4 % envelope polypeptides, 30.8 % lipid and 6.3 % proteinbound carbohydrate. The monosaccharides found in the virus include N-acetyl-glucosamine, N-acetyl-galactosamine, mannose, galactose, glucose, fucose and sialic acid. Analyses of the individual monosaccharides are presented. The number of molecules of each of the major constituents in an average virion was calculated.

摘要:
A radioimmunoassay based on the double-antibody technique was developed for the major group-specific (gs) antigen of Rous sarcoma virus. As little as 1 ng/ml of the gs protein could be measured. This test appears to provide a highly sensitive and specific assay for the expression of the RNA tumor virus genome in various biological samples.

Indirect immunofluorescence was applied to achieve the rapid diagnosis of enteroviral meningitis within hours of receipt of the specimen. Cerebrospinal fluid (CSF) leukocytes from 40 patients were studied by a method adapted to utilize combination antiserum pools in conjunction with indirect immunofluorescence, and results were compared to those of conventional isolation and serological techniques. When virus was isolated from the CSF (17 patients), there was general agreement with the virus identified by immunofluorescence staining of CSF leukocytes. The advantages of immunofluorescence were particularly striking in 19 patients from whom virus could not be isolated from the CSF and in 4 patients from whom virus could not even be isolated from the throat or rectum. By immunofluorescence, virus was detected in 20 of 21 of these patients. Overall, the rapid immunofluorescence findings were confirmed by the slower conventional methods in 32 of 33 cases. Economy of both time and materials was made practicable by use of the LBM combination antiserum pools, which allow 42 antigenic types to be identified in a single set of 8 simultaneous tests.

The appearance of simian virus 40 (SV40) after fusion of virus-transformed cells with susceptible cells was examined by electron autoradiography. The initial site of synthesis of virus particles was the transformed cell nuclei of the heterokaryon.
