Endogenous Glyceride Synthesis Rate in Baboons Fed Diets Containing Sucrose or Glucose |

Triton WR 1339 has been used to block lipid removal from the serum of baboons. When Triton WR 1339 is given intravenously to baboons, there is a linear increase in the serum lipid concentrations over a 3-hour period. The rise in serum glyceride concentration is very marked, while cholesterol and phospholipid concentrations increase more slowly. The rates of secretion of glyceride, cholesterol and phospholipids into the serum have been calculated for baboons in the fasting state. The mean values for glyceride, cholesterol and phospholipid secretions in the male and female animals are not statistically different. In both male and female baboons, the rates of secretion of glyceride into the serum are about six times greater than the rates of secretion of cholesterol and phospholipid as determined in these experiments. Consumption of a diet containing 75% by weight of sucrose for a period of 4 weeks is associated with a rise in fasting serum glyceride concentration in male and female baboons. In those animals consuming 75% by weight of glucose, no significant changes occur in the fasting serum glyceride concentrations. Ingestion of a high sucrose diet produces no significant change in the fasting serum cholesterol and phospholipid concentrations, while in those baboons consuming the high glucose diet there is a significant fall in the serum concentrations of these lipid fractions. There is no measureable increase in the rate of secretion of glycerides, cholesterol or phospholipids into the serum in the fasting state after 4 weeks of consumption of high glucose or sucrose diets as compared with the control period.

A 3-week-old septicemic full-term infant developed severe metabolic acidosis (total CO2 < 5 mEq/l) and underwent intravenous antibiotic and bicarbonate therapy with good response. Attempts to introduce milk formulae triggered recurrence of metabolic acidosis (total CO2 < 11 mEq/l) which was spontaneously corrected without bicarbonate therapy during periods when only glucose and water or PM 60/40 were given. One month after apparent recovery from the septicemia, the technique of net acid balance was applied to study the effects of Nutramigen on each component of endogenous net acid production and net acid excretion. Comparison was made with similar balance periods on glucose-water and Similac formulae. When metabolic acidosis developed during feedings on Nutramigen and Similac, the acid balance was positive. Spontaneous recovery occurred when glucosewater was substituted and acid balance reverted to zero.The data indicated that the acidogenic properties of Nutramigen and Similac played an important part in the development of acidosis; whereas, glucose-water was shown to be compatible with zero to negative hydrogen ion balance and recovery from systemic acidosis. It was postulated that as a sequelae of the septicemia, the renal acidification mechanism was temporary impaired, resulting in the development of metabolic acidosis when the milk formulae with greater hydrogen ion loads were used. It was recommended that, besides correction with sodium bicarbonate, substitution of milk formula with glucose-water for a few days should be considered in infants who develop systemic acidosis without apparent cause.

The nutritional effects of feeding either sucrose or starch as components of a high carbohydrate low protein diet were studied and the effects of a combined oral contraceptive observed. Animals receiving the contraceptive had a greater weight loss. An inverse relationship between serum albumin and γ-globulin was demonstrated, but did not hold for rats on the sucrose diet receiving the ‘pill’. Five days fasting in 12 non-obese, healthy volunteers caused a reduced capacity to excrete a single water load and to form maximally hypotonic urine. The minimum value of osmolarity of urine after fasting was on average 113mOsm/l, while it was 63 mOsm/l under control conditions. This abnormality in the formation of hypotonic urine could not be explained as the consequence of reduced glomerular filtration or enhanced solute excretion. The reduced diluting capacity is associated with an inadequate reduction of tubular water reabsorption.

To study the effect of protein deficiency on collagen cross-Unking in albino-rat skins and granulomas, two groups of weanling albino rats were raised for 3 weeks with normal and low protein diets by paired-feeding technique. The analysis of skins showed α1and α2-chain subunits of neutral salt-soluble collagen appreciably increased, β11- and β12-chains and the aldehyde content significantly decreased whereas the solubility of insoluble collagen in KCNS, urea or pronase increased greatly in protein-deficient animals compared to controls. The gel reversibility of neutral salt-soluble collagen increased in both skins and granulomas of protein-deficient animals compared to controls. Three dietary levels of essential fatty acids (EFA) 3.0, 0.75, and 0.07 cal% were studied for their influence on the concentration of lipids, linoleic and arachidonic acids, water, nitrogen, potassium, and ash of the whole body of rats. The animals were sacrificed and examined at 60, 90, and 120 days of age. They were fed the experimental diets and kept under standardized conditions for two generations or more before the actual experiment. The total body concentration of lipids increased and that of water decreased with increasing age independently of the dietary EFA level. In the rats fed 0.75 cal% EFA, the lipid concentration was higher than in those fed 3.0 cal% EFA, while in rats fed 0.07 cal% EFA the lipid concentration was lower than in those fed 0.75 cal% EFA. The concentration of water varied inversely with that of lipids. The differences were most pronounced at the highest age (120 days), and larger in the female rats than in the males. Nitrogen, potassium, and ash did not differ between the groups at any age. The role of EFA for the amount of the adipose tissue is discussed.
