摘要:
In order to resolve a controversy concerning the relative effectiveness of adenosine and inosine for maintaining the high energy phosphates of incubating red blood cells, experiments were performed in which either of the nucleosides was added, sometimes at the beginning of incubation and sometimes later. The results indicated that the relative effectiveness varied according to the concentration and time of addition of the nucleoside. The variability of the results with time is attributed to the fact that adenosine and inosine affect different ATP synthetic reactions whose rates change during incubation. Experiments with adenine indicated that adenine moiety is important for ATP maintenance.

During storage of blood the percentage of viable erythrocytes decreases at such a rate that when these percentages are plotted against time of storage, they fall on an S‐shaped curve. This curve can be explained on the assumption that the resistance of red cells toin vitroaging is normally distributed,i.e., follows the normal probability curve. By a mathematical transformation used in other cases of biological standardization, the curve can be changed to a straight line, the slope of which, if accurately determined with a sufficient number of recipients, is the most accurate method of estimating the rate ofin vitroaging.To illustrate the method, the rate ofin vitroaging has been determined for the following storage procedures: (a) addition of ACD solution; (b) addition of two different solutions containing EDTA as anticoagulant; (c) prevention of coagulation by removal of calcium with an ion‐exchange resin and subsequent addition of glucose‐lactate or glucose acetate solution. Of these, the last had the fastest rate ofin vitroaging and the first the slowest. Possible reasons for this are discussed. In a series of red cell survival experiments, autotransfusions of erythrocytes drawn in CPD solution maintained better viability than those in ACD solution (NIH, Formula A), after 28 days continuous storage at 4 ± 1 C. Augmented erythrocyte preservation in CPD was accompanied by lesser amounts of hemoglobin in the plasma of the donor blood, and in the recipient's plasma at 15 minutes after the transfusion. A study was undertaken to establish whether the use of stored ACD blood, up to four days old and subsequently heparinized and recalcified, in the pump‐oxygenator for open‐heart surgery would have any detrimental effect upon postoperative coagulation and hemostasis. This paper encompasses a survey of the literature concerning the uses of dextrans in the U.S.S.R., China, Czechoslovakia and Poland. The data accumulated indicate that Communist dextran preparations are safe and effective plasma volume expanders and have properties and characteristics essentially similar to dextrans produced in the United States. In contrast to other dextran preparations, a hemostatic defect following intravenous infusions of the Soviet dextran, Polyglucin, has been reported not to occur. The Soviet claims that Polyglucin exerts an erythropoietic stimulatory effect and has hemostatic properties are at variance with Western reports and appear to be unsubstantiated by the published data. Dextran is currently being produced on a large scale in Communist countries, and its use in the treatment of hemorrhagic, traumatic, and operative shock apparently far exceeds that in the United States.

摘要:
In vivoandin vitroexperiments show that sickle trait blood can be safely used for blood transfusions. However, nowhere in the literature is there a reference as to the possible dangers of the use of such blood in exchange transfusions.We are reporting a case of a premature white infant with physiological jaundice who received an exchange transfusion of blood containing hemoglobin A and S and expired five days later. Autopsy revealed generalized intravascular aggregation of sickled erythrocytes similar to that found in patients dying in sickling crises.

A case is described in which a severe hemolytic and hemorrhagic reaction occurred as the result of incompatibility involving an anti‐Leaantibody of “immune” type. This antibody must have developed before the surgical procedure at which the reaction occurred, but was not detected because patient specimens for crossmatching had not been obtained frequently enough during multiple transfusions. The patient had a stormy postoperative course, mainly as a result of hematoma formation and renal shutdown, but recovered completely. Five months after the reaction, the antibody was detectable only after EDTA treatment of the serum or the addition of complement.
