Various antibodies and immune complexes have been demonstrated in patients with Guillain‐Barre syndrome (GBS). These factors may be part of the pathogenesis of the disease process through immune complex deposition and/or suppression of the immune system. Four patients with typical GBS were treated by plasma exchange. Aliquots from the first 100 ml of plasma removed from each patient were incubated with cultures of normal human peripheral lymphocytes that subsequently were exposed to mitogens, followed by incubation with tritiated thymidine. A decrease (p<0.001) in response to each of the mitogens was noted after incubation with patient plasma. Peripheral lymphocytes were taken from the patients before and after plasma exchange. An increase (p<0.01) in DNA synthesis following mitogen exposure was observed after plasma exchange in each patient. All patients exhibited subjective and objective clinical improvement. Plasma exchange for GBS appeared to remove a lymphocyte suppressor factor, which was characterized by column fractionation. The suppressor factor was determined to be a substance of approximately 10,000 to 12,000 daltons that reproducibly decreased the responsiveness of normal lymphocytes to mitogen

A technique for performing the enzyme phase of the antibody screen on red cells suspended in low‐ionic‐strength‐salt solution (LISS) is described. The reliability of this LISS spin‐enzyme (LSE) technique was compared with the two‐stage papain‐tile method, in the detection of 62 previously identified enzyme reacting antibodies. All 62 antibodies examined were detected by the LSE method, and no false‐positive reactions were found. Using LSE and papain‐tile methods in parallel, further assessment was obtained by screening 2000 sequential blood samples under routine service conditions. Fifty‐six blood samples contained alloantibodies, of which 43 reacted by both methods, eight by the LSE method only, and five by the papain‐tile method only. Whole blood and plasma drawn into plastic bags containing citrate‐phosphate‐dextrose (CPD) and stored at 4°C for various periods were studied for variations of coagulation factor and fibrinolytic activity. The blood was collected and processed in the conventional way. The most labile component, factor VIII coagulant activity, decreased to about 50 percent of its original value within the first 24 hours in whole blood, but thereafter, it decreased more slowly. Storage of whole blood for 6 hours at 4°C had an insignificant effect on VIII coagulant activity; an even slower decrease was found for factor VIII coagulant antigen. The major fall in VIII coagulant activity occurred between 6 and 24 hours of whole blood storage. Factor Vlll‐related antigen remained normal for about 1 week, but on further storage showed signs of proteolytic degradation.In plasma, there was a successive decrease in VIII coagulant activity with its minimum level (about 50% decrease as compared with the original level) after 7 to 14 days of storage. All other factor VIII activities in plasma remained unchanged throughout the study.Factor V retained its activity for about 1 week in the whole blood. Factors II, VII, IX, X, XII, XIII and fibrinogen did not fall below normal during a storage period of 35 days, nor was there any indication of increased fibrinolytic activity in either whole blood or plasma. Storage of whole blood and plasma at 4°C for 1 to 2 weeks seems to have relatively little effect on the levels and function of various coagulation components with the possible exception of factor VIII coagulant

The serum of a black woman has been found to contain a potent alloanti‐N that reacted in direct agglutination tests with the ‘N’ antigen carried on Ss‐active sialoglycoprotein. Human monocytes were isolated from the cellular residues remaining after plateletapheresis of donors using an automated blood cell processor. Mononuclear cells were obtained with density gradients and separated from lymphocytes by stepwise elutriation. The isolated cells were frozen using extracellular hydroxyethyl starch and intracellular dimethylsulfoxide.In three procedures, approximately 1 × 10° monocytes were obtained. Ninety‐nine percent of isolated monocytes were viable in a fluorescein diacetate (FDA)‐ethidium bromide (EB) test. Myeloperoxidase‐positive cells were 95 percent and 90 percent in the two chambers. Ninety‐four percent of monocytes ingested five or more opsonized polycyclic hydrocarbon particles and 95 percent ingested one or more ethidium‐treated zymosan particles.After storage in liquid nitrogen for up to 9 weeks, 99 percent of the cells were recovered after thawing.
