摘要:
The peroxidase‐anti‐peroxidase immunoenzyme method was applied to red blood cell ghosts for the detection of ABO mixed‐red blood cell populations. Red blood cell ghosts produced by acid‐glycine lysis eliminated previous problems due to distorted red blood cell and hemoglobin‐associated peroxidase activity. Anti‐A,B was used to discriminate between various mixtures of test cells in group O cells. The positive‐stained minor population of test cells stained dark brown following application of the peroxidase‐anti‐peroxidase method and the nonreactive group O cells were counterstained light blue with Coomassie brilliant blue which facilitated quantitation. The expected and observed numbers of positive‐stained cells per 1000 cells were not significantly different as computed by X2. Erythrocytes, serum, and blood monocytes from patients with erythrocyte autoantibodies were examined byin vitroserologic and cell culture procedures in order to evaluate factors modulating patient mononuclear phagocyte activity. Monocyte recognition of autoantibody‐coated erythrocytes was influenced both by antibody titer score and by the monocyte source. The results suggest that patients with low serum autoantibody titer scores and normal monocyte behavior are less likely to destroy autologous or normal donor erythrocytes than are patients with high‐serum antibody titer scores or enhanced monocyte activity. We conclude that patient, rather than normal donor monocytes, must be used in cellular assays designed to predict the fate of erythrocytes transfused into patients with erythrocyte autoantibod

摘要:
Antibody elution by chloroform and trichloroethylene was compared with the Rubin ether method modified by Hughes‐Jones for IgG antibodies and the Landsteiner and Miller heat method for IgM antibodies. After washing the cells, the chloroform and trichloroethylene method was done at 37° C for 10 minutes. With this technique the eluate remained on the upper layer above the red blood cell stroma layer, while with Rubin's method the eluate remained below the cell stroma. Approximately 25 percent of black blood donors have an elevated red blood cell (RBC) sodium (Nai) level compared with white donors. This elevation results in a significant increase in the mean Nai from black (9.00 ± 2.96 mmoles/l RBC) as compared to white blood donors (7.04 ± 1.48 mmoles/l RBC, p less than 0.001). Red blood cells from four black donors with mean Nai levels of 15 ± 2.8 mmoles/l RBC were stored for 35 days in citrate‐phosphate‐dextrose‐adenine and compared to that of four donors with normal levels of Nai. Serial measurements of red blood cell adenosine triphosphate, diphosphoglycerate, glucose‐6‐phosphate dehydrogenase, pyruvic kinase, lactate production rates, and intracellular cations showed no differences between the two donor groups. Furthermore, the mean 24‐hour posttransfusion survival was not significantly different for the high Nai group (83.2 ± 5.6%) as compared with the control group (82.3 ± 6.9%). Previous studies suggested that a general metabolic defect might be responsible for impaired chemotaxis of stored granulocytes (PMN). We studied PMN energy metabolism in fresh and stored PMN.Adenosine triphosphate (ATP) decreased slightly in unstimulated PMN after 48 hours of storage at room temperature. Exposure of fresh PMN to opsonized zymosan resulted in a 12 percent decrease in ATP. In contrast, PMN stored for 24 or 48 hours at room temperature and then exposed to opsonized zymosan, showed significant decreases in ATP. Incubation of fresh or stored, stimulated (opsonized zymosan) or resting PMN with potassium cyanide resulted in no change in the pattern of ATP decrements during storage. Likewise, incubation of fresh or stored, stimulated or resting PMN in the presence or absence of extracellular glucose, resulted in no change in the pattern of ATP decrements during storage. Preincubation of PMN with 2‐deoxy‐glucose obscured the differences in ATP maintenance between fresh and stored PMN. ATP decrements were also observed in unstimulated PMN which had been stored at high cell concentrations. Decrements in ATP correlated stronglywith decreased glucose (r = .82) and pH (r = .89) in the storage medium.These decrements were partially reversed by incubation of PMN in fresh buffer. The ATP decrements in stored, concentrated granulocyte preparations thus were prevented by maintenance of pH. ATP decrements were not prevented by additional glucose. Thus, in PMN stored for 24 or 48 hours at room temperature, there was a defect in ATP maintenance which was occult in unstimulated cells, but was evoked by the energy‐dependent process, phagocytosis. These findings are compatible with a major role for glycolysis and glycogenolysis in maintaining ATP in fresh and stored PMN. The effect of white cell alloimmunization on patient outcome during gram‐negative sepsis treated with granulocyte transfusions was studied. Twenty‐five episodes of sepsis were observed; 19 were associated with resolution of sepsis and six with continuing sepsis and death. Compatibility testing included the granulocyte indirect immunofluorescence test and the lymphocytotoxicity assay. The number of compatible and incompatible granulocyte transfusions determined by the indirect immunofluorescence test compared with patient outcome was significant (X2= 44, p less than 0.001). The same comparison with the lymphocytotoxicity assay was not significant (X2= 3, p greater than 0.05). The duration of the granulocytopenia after the first positive blood culture was 10 days or longer in 13 patients, and 12 of the 13 survived. The duration of the granulocytopenia was less than 10 days in 12 patients, and five of the six deaths occurred in this group. These five patients died without evidence of bone marrow recovery and with persistent gram‐negative sepsis. Granulocytes were isolated b overlaying leukocyte‐rich plasma discontinuous density gradients. The cell suspensions were 95 percent granulocytes. Sera of 383 polytransfused patients and 521 multiparous women were examined for lymphocytotoxins, granulocytotoxins, and granulocytoagglutinins. On the basis of these tests, sera were identified which contained only granulocytotoxins. These were further identified with more precision as to their titers and antibody absorption; reactivity with granulocytes, B, and T lymphocytes; and K 562 cell line.
