Leukocytes were removed from washed, freeze‐preserved red blood cells primarily by the clumping together of damaged cells. This agglomeration was associated with membrane and nuclear changes which were usually manifested after the removal of glycerol from the red blood cells. Washing glycerolized, freeze‐preserved leukocytes with solutions containing deoxyribonuclease prevented the agglomeration. Polymorphonuclear granulocytes were preferentially damaged and removed. Of the residual leukocytes that were isolated by the dextran sedimentation method, 70 per cent were lymphocytes. The dye exclusion test showed that the majority of the agglomerated leukocytes were nonviable, while the majority of the leukocytes isolated by dextran sedimentation were viable. The agglomerated material ranged in size from macroscopic aggregates to microscopic particles with a minimum diameter of 40‐50μ. In the study of 125 units of washed, previously frozen red blood tells approximately 96.6 per cent of the leukocytes and 98.9 per cent of the platelets were removed. Less than 0.5 mg of plasma protein remained per unit. The length of storage as whole blood at 4 C prior to glycerolization of the red blood cells influenced the number of leukocytes in the washed, previously frozen red blood cells. Red blood cells frozen on the day of collection had 7.2 per cent residual white cells, whereas those stored at 4 C for 10 days or longer prior to glycerolization, freezing, and washing had 1.1 per cent residual leukocytes. When red blood cells were stored at 4 C for longer than one week, glycerolized, and washed without freezing, they had about 2 per cent residual leukocytes and platelets. The washing procedure removed at least 95 per cent of the platelets from nonfrozen and previously frozen red blood cells. Overt hepatitis was found in 11.7 per cent of the hemophiliac population studied. In those who had received fractions prepared from pooled plasma, the incidence was 22 per cent. Concentrates of factor IX were associated with hepatitis about three times as often as were factor VIII fractions. Blood samples obtained during the acute phase of hepatitis were available from 12 of the 14 patients treated with factor VIII fractions and eight of the ten treated with factor IX concentrates. Two of the 12 samples from factor VIII deficient patients and seven of the eight from factor IX deficient patients were positive for HB Ag. Antibody (HB Ab) was found in six factor VIII deficient and one factor IX deficient patient with hepatitis. Severe diffuse intravascular coagulation (DIC) occurred in a factor IX deficient man who had been treated with factor IX fractions and who expired in hepatic coma.Multiple serum samples were available from 89 patients who had not had overt hepatitis but had received fraction therapy. Antigenemia was found in one hemophiliac and one afibrinogenemic. Antibody was found in 24 per cent of the factor VIII and 17 per cent of the factor IX deficient patients. Usually HB Ab appeared following therapy with plasma fractions and lessened or disappeared during quiescent periods only to recur after additional exposure to pooled plasma fractions. Remarkably, of the 113 patients who have received fraction therapy, 66 (58%) have not had hepatitis or developed circulating HB Ag or HB Ab.

A black woman in the second month of her fourth pregnancy was found to have moderately strong autosensitization of her own erythrocytes (RBC) by C3d (α2D); at the same time, anti‐Jkawas detected in her serum. At term she delivered a healthy Jka‐positive ABO‐compatible infant whose RBC were very strongly coated by C3d. Anti‐Jkawas demonstrable in maternal and cord serum at a titer of 1/64, and in an eluate from the newborn infant's washed RBC at a titer of 1/8. Despite very strong RBC C3d sensitization and the persistence of free anti‐Jkain the infant's circulation for at least six weeks postpartum, no evidence for hemolytic disease of the newborn developed. Three hundred thirteen women were typed for Gm and Inv factors and screened for the presence of antibodies to gamma globulin both before receiving Rh immune globulin and at six weeks, three, six, and nine months after injection. Thirty‐five of the 313 women received multiple doses of Rh immune globulin following one or more pregnancies. In this group of patients, the incidence of anti‐gamma globulins was no greater than in the women given a single 0.7 to 2.25 ml dose. Anti‐gamma globulins were found in only 17 women (5.4%). Eleven had antibody prior to treatment and in six it first appeared in post injection specimens. Five of the women had allotype specific Gm agglutinators. In three, these were found in the pretreatment specimens. No evidence of Rh sensitization was demonstrable in the eleven Rh immune globulin recipients who had circulating anti‐gamma globulins at the time of treatment. In this study, three cases of Hodgkin's disease with autoimmune hemolytic anemia are described. The IgG autoantibodies in each case were shown to have ITspecificity. There is only one other case of an IgG anti‐ITdescribed in the literature and that also was present in a patient with Hodgkin's disease. No other examples of anti‐ITwere found in studies which included 50 cases of Hodgkin's disease with negative direct antiglobulin tests, three cases with positive direct antiglobulin test but no hemolysis, and 70 cases of autoimmune hemolytic anemia, either idiopathic or secondary to other diseases of the reticuloendothelial system. Whole human blood was incubated for periods of up to 70 hours at 37 C under conditions that would cause minimal modifications in normal metabolic controls. Insofar as possible, the conditions utilized were similar to those foundin vivo. Control of pH within the limits of 7.2 to 7.5 was achieved by the addition of isotonic Tris buffer or isotonic TES buffer. Glucose, inorganic phosphate, adenine, and pyruvate were added as needed throughout the incubation period. The ADP/ATP ratio and levels of ATP, total adenine nucleotides, 2,3‐diphosphoglycerate and other metabolic intermediates were maintained near those of fresh erythrocytes. 摘要:
The vasodepressive activity contained in a 5 per cent heat‐treated human plasma protein solution has been studied and found to be a bradykinin‐like substance. When the solution was injected intravenously into a dog at a dose of 125 to 214 mg protein per kg body weight, at a rate of 18 to 30 ml per minute, a decrease in arterial pressure of 11.5 to 25.0 per cent was observed.The solution also showed a marked contractile effect on smooth muscle of the isolated rat uterus. The extent of contraction was nearly equal to that caused by a bradykinin solution at a concentration of 25 to 125 ng/ml. The molecular weight of the vasodepressive substance is in the range of 1,000 to 10,000 as demonstrated by ultrafiltration. The substance was thermostable, dialyzable, and inactivated by carboxypeptidase B. Separation of the vasodepressive substance from the heat‐treated plasma protein was achieved either by gel filtration on Sephadex G‐50 or by selective adsorption.
