摘要:
The synthesis of a series of substituted bay-region bridged chrysene derivatives is described. The method involves the oxidative photocyclization of stilbene carboxylic acids followed by dehydration with hydrofluoric acid. The photocyclization of stilbene carboxylic acids lead to secondary photolactonizations and functionalization of a bay region carbon by a novel photolytic process. The mutagenic activity of the substituted chrysenes was determined and activity levels correlated with substituent effects.

摘要:
An efficient method for the synthesis of 1-hydroxy-2-nitropyrene from 1-hydroxypyrene is presented. The synthesis utilizes a recently described nitrating agent for aromatic compounds, 4-nitro-ethyl-2,3,5,6-tetrabromo-2,5-cyclohexadien-1-one. Application of this procedure for synthesizing nitro-derivatives from hydroxyarene precursors of fluoranthene, phenanthrene and chrysene is discussed.

Treatment of four PAH adsorbed on silica gel with a gas stream containing ozone and 2,3-dimethyl-2-butene leads to the formation of the K-region oxide in each case. A similar treatment using ozone alone did not give oxide formation. The oxide formation is thus due to one or more oxidants derived from the ozone-alkene reaction. We suggest that acetone oxide or dimethyldioxirane are good candidates for these oxidants. The results suggest that oxidants derived in this manner may be partially responsible for the observed production of mutagens and carcinogens in polluted atmospheres containing ozone, alkenes and particulate-adsorbed PAH.

As part of the Total Human Exposure to Environmental Substances (THEES) Study, weekly composites of in-home prepared meals from approximately ten individuals for each of three two-week “campaigns” were analyzed for benzo[a]pyrene content. In addition, daily composites for four individuals during the third “campaign” (THEES III) were analyzed. The analytical technique employs alcoholic KOH saponification of food samples, extraction, column chromatography on florisil and TLC chromatography on 20% acetylated cellulose and detection/quantitation using fluorescence. Limit of detection and sample recovery data are presented in addition to the meal data. B[a]P levels in food are highly variable but are frequently high enough to make contributions which exceed inhalation exposure.

Ames assays (TA98) were applied toward assessing the mutagenic activities of extract fractions and subfractions of inhalable airborne particulate matter obtained in the manner of the earlier ATEOS study. Specifically, sequential cyclohexane, dichloromethane and acetone extracts of 24-hour samples were composited on a quarterly basis (ca 15 samples/quarter) and then fractionated using thin layer chromatography. For all four quarters the highest specific activities were associated with dichloromethane extracts and, specifically, its most polar fractions. Hydroxynitro-PAH are likely components in the dichloromethane extract. The cyclohexane extracts, containing PAH and nitro-PAH, were much less active. The acetone extracts had significant activity possibly associated with azaarenes and phenols. While PAH levels decreased in summer, nitro-PAH levels rose. No simple trend was observed for the PAH-quinones. 2-Nitropyren-1-ol, formed readily via solution photoxidation of 1-nitropyrene, does not appear to be formed under ambient atmospheric conditions. Preliminary FTIR, HPTLC and GC/MS results are described.

Detailed identification of sulfur-containing polycyclic aromatic compounds (S-PAC) was accomplished for solvent extracts of five different coals. The S-PAC were isolated by ligand-exchange chromatography. Due to the great complexities of these samples, capillary gas chromatography (GC) was employed for their analysis. An element selective radio frequency plasma detector was used to provide sulfur selective detection. Gas chromatography-mass spectrometry was used to identify individual compounds in isolated sulfur fractions. Condensed thiophenic compounds were found to be the major constituents in all five S-PAC fractions. Diaryl sulfides were also detected in the high volatile bituminous Illinois #6 coal. The prevalence of various S-PAC in different coal extracts was observed as a function of their rank.
