SummaryThe pneumonic lungs of 38 sheep and 62 goats were examined for Mycoplasma and Acholeplasma and they were isolated and their preliminary biochemical characterization determined from the pneumonic lungs of 13 of 38 sheep and 17 of 62 goats. Various species ofMycoplasmaandAcholeplasmawere finally identified from 18 of the 30 original cultures. They comprisedM. argininiin 13 cases (4 sheep and 9 goats),A. laidlawiiin 3 cases (2 sheep and 1 goat), andM. agalactiaeandM. capricolumin one goat each. All these species were isolated from pneumonic lungs for the first time in India andM. capricolumwas isolated from goat pneumonia for the first time anywhere. Zusammenfassung1. In einer Studie zur Parasitenfauna der Rhesusaffen (Macaca mulatta) wurden 50 Tiere untersucht. Insgesamt wurden 19 Parasitenarten vorgefunden. Diese sind nach der Häufigkeit ihres Vorkommens:Strongyloides fülleborni(76 %),Pedicinus obtusus(56 %),Oesophagostomum bifurcum(52 %),Pneumonyssus simicola(26 %),Oesophagostomum aculeatum(20 %),Trichuris trichiura(20 %),Streptophargus armatus(12 %),Trichostrongylus colubriformis(8 %),Bertiella studeri(6 %),Nochtia nochti(4 %),Gastrodiscoides hominis(4 %),Primatotrema macacae(4 %),Sarcocystis kortei(2 %),Sarcocystis nesbitti(2 %) undEnterobius bipapillatus(2 %). ZusammenfassungEs werden die wichtigsten Paradoxa der immunologischen Neutralisation beschrieben und erklärt. Der Danysz‐Effektist für das Ergebnis einer gebrochenen Neutralisation repräsentativ, bei der durch Affinitätsverlust ein finaler Überschuß an Antigen entsteht. Das Ehrlichsche Phänomendemonstriert, daß das Pufferungsmaximum eines Serums im Bereich partiell neutralisierten Antigens, d. h. in der titrimetrischen Zone relativen Antigenüberschusses gelegen ist. DieVariabilität des in vitro / in vivo‐Quotientenals drittes Paradox weist auf Störungen des Endpunktes schnellster Präzipitation durch inkomplette Antikörper hin. Die Paradoxa werden auf ihre theoretische und praktische Bedeutung untersucht, wobei auf Anomalitäten des Immundiffusionstestes eingegangen wird.SummaryParadoxes of neutralizationThe most important paradoxes of immunological neutralization are described and discussed. SummaryThe percentages of B and T lymphocytes in peripheral blood of normal foals were studied. B lymphocytes were identified by labelling of their surface immunoglobulins with fluorescein‐conjugated anti‐equine‐IgG antibody. T lymphocytes were enumerated by the use of the E‐rosette test with guinea pig erythrocytes. A few days old colostrum‐fed foals had only about 5 % B lymphocytes in blood. Beginning from 20 days of age the average percentage of B lymphocytes increased until 40 days of age when the values stabilised (approximately 12 % to 15 %) and seemed to remain constant until the end of the observation period (160 days). The average values of circulating T lymphocytes ranged from 5.2 % to 16.1 % and they seemed to be less dependent on age of the foals.ZusammenfassungZirkulierende B‐ und T‐Lymphozyten bei Fohlen während der ersten fünf LebensmonateDer Prozentsatz an B‐ und T‐Lymphozyten im peripheren Blut von gesunden Fohlen wurde untersucht. B‐Lymphozyten wurden durch Markierung ihrer Oberflächen‐Immunglobuline mit fluorescein‐konjugierten gegen Pferde‐IgG gerichteten Antikörpern identifiziert. Die Zählung der T‐Lymphozyten erfolgte im E‐Rosettentest unter Verwendung von Meerschweinchenerythrozyten. Ein wenige Tage altes mit Kolostrum gefüttertes Fohlen hatte nur etwa 5 % B‐Lymphozyten im Blut. Ab dem 20. Lebenstag stieg der durchschnittliche Prozentsatz an B‐Lymphozyten bis zum 40. Lebenstag und stabilisierte sich dann bei 12–15 %. Die Beobachtungen wurden nach 160 Lebenstagen abgebrochen. ZusammenfassungEs wurden die Auswirkungen einer Inokulation von Mykoplasma bovigenitalium in die Samenblase von Stieren auf klinischen Status und Spermiogramm im Vergleich zu Kontrolltieren, denen unbeimpfte Nährbouillon inokuliert wurde, untersucht.Charakteristische Veränderungen traten bei den mit Mykoplasma bovigenitalium infizierten Tieren in Form einer Vesiculitis seminalis einhergehend mit einem signifikanten Anstieg pathologischer Spermienformen auf. ZusammenfassungIm Darmligaturtest beim Ferkel wurde die Enterotoxinbildung von 35Pseudomonas aeruginosa‐Stämmen untersucht. Von 19Pseudomonas‐Stämmen, die von lebenden Tieren isoliert wurden, waren 16 und von 7 Stämmen, die aus Lebensmitteln stammten 7 positiv. Stämme aus pflanzlichen Futtermitteln reagierten dagegen zum überwiegenden Teil (89 %) negativ.Reproduzierbare positive Ergebnisse erreichte man nur in den ersten zwei Metern des Dünndarms und nach Inokulation von mindestens 5 × 109Keimen pro Segment.Die in vitro‐Enterotoxinbildung war relativ schwach. Die beste Toxinbildung entwickelte sich in belüfteten und gerührten Kulturen. Unterschiedliche Bebrütungstemperaturen und Nährböden hatten keinen Einfluß auf die in vitro‐Toxinbildung. Das in vitro gebildete Enterotoxin war sehr labil und ließ sich nicht mit den konventionellen Methoden konservieren.SummaryDemonstration of enterotoxin‐forming strains of Pseudomonas aeruginosa by the ligated ileal loop test in pigletsEnterotoxin formation by 35 strains ofPseudomonas aeruginosawas investigated by means of the ligated ileal loop test in piglets. Eighty‐four percent of the strains isolated from domestic animals were positive and from human foodstuff all of 7 isolated were positive. Strains isolated from animal feeds were predominately (89 %) negative. Reproducible results were attained only when the first two meters of the small intestine were used and when the inoculation consisted of at least 109bacterial cells per segment.The production of enterotoxin in vitro was relatively weak. The best titers of toxin were achieved in aerated and shaken cultures. Variations in growth temperature and composition of the growth medium had no influence on in vitro toxin formation. ZusammenfassungIn der vorliegenden Arbeit wurde die Art der bei experimenteller Aleutenkrankheit (A.K.) der Nerze vorkommenden Autoantikörper definiert und die Höhe der Antikörpertiter in verschiedenen Krankheitsphasen bestimmt. Die angewandte Methodik umfaßte den indirekten Immunfluorezenztest, die passive Hämagglutination, die Gegenstromelektrophorese und den Antiglobulintest (Coombs‐Test). Insgesamt 43 Seren von experimentell mit dem Virus der A.K. infizierten Nerzen sowie zwölf Kontrollseren wurden auf das Vorkommen antinukleärer Antikörper untersucht. Die benutzten Substrate waren Leber und Niere von Nerz und Ratte, Hühnererythrozyten, Schweineleukozyten und Kalbsthymozyten.Antinukleärer Faktor mit niederem Titer kam auch bei einzelnen nichtinfizierten Kontrollnerzen vor. Nach Infektion mit dem A.K.‐Virus war ein Anstieg des Antikörpertiters mit einem Höhepunkt 3 bis 4 Monate post infectionem zu beobachten. Versuche, Antikörper gegen cytoplasmatische Antigene in Tubulusepithelien der Niere und Gallengangsepithelien der Leber nachzuweisen, verliefen negativ. Antikörper gegen glatte Muskulatur waren bei zwei von vier untersuchten Kontrollseren und 16 von 17 getesteten Seren aleutenkranker Nerze mit Titern von 1: 4 bis 1: 256 vorhanden.Mittels passiver Hämagglutination ließ sich bei 20 von 36 untersuchten Seren (55,6%) Antiglobulinfaktor mit einem Titer von 1: 16 und höher nachweisen. Bei sechs von neun Kontrollseren lagen die Titer bei 1: 8. Zum Nachweis von DNS‐Antikörper wurden die Nerzseren in der Gegenstromelektrophorese gegen hitzedenaturierte DNS (ss‐DNS) getestet. Es zeigte sich mit dieser Methode, daß vier von zwölf untersuchten Kontrollseren DNS‐Antikörper mit niederem Titer enthielten. Bei aleutenkranken Nerzen lagen die Titer deutlich höher und erreichten Höhepunkte 2 bis 3 bzw. 5 bis 8 Monate post infect. Mit dem direkten bzw. indirekten Antiglobulintest (Coombs‐Test) konnten keine reproduzierbaren Ergebnisse erzielt werden.Die Ergebnisse dieser Untersuchung lassen folgende Schlußfolgerungen zu: Antinukleäre Antikörper stellen mit Wahrscheinlichkeit ein serologisches Epiphänomen dar und werden im Verlaufe der persistierenden A.K.‐Virusinfektion geringgradig verstärkt gebildet. Dem nachgewiesenen DNS‐Antikörper kommt möglicherweise in der Pathogenese der A.K. eine gewisse Bedeutung zu, da bei diesem Autoantikörper die Titer deutlich höher als bei nichtinfizierten Kontrollen lagen.SummaryStudies on the pathogenesis of Aleutian Disease in mink IX. Occurrence of auto‐antibodiesThe type of autoantibodies that occurs in experimental Aleutian Disease (A. D.) in mink is defined and the antibody titres in different phases of the disease are recorded. The techniques include the immunofluorescence test, passive haemagglutination, counter‐current electrophoresis and the Coombs antiglobulin test. A total of 43 sera from mink experimentally infected with AD virus and 12 control sera were examined for anti‐nuclear antibodies. The substrates used were liver and kidney from mink and rats, fowl erythrocytes, pig leucocytes and calf thymocytes.Antinuclear factor at a lower titre also occurred in individual uninfected control mink. After infection with AD virus there was a rise in antibody titre with a peak 3–4 months after infection. Attemps to demonstrate antibodies against cytoplasmic antigen in the tubular epithelium of the kidney and the bile duct epithelium in the liver gave negative results. Antibodies against smooth muscle were found in titres from 1: 4 to 1: 256 in 2 of 4 control sera and in 16 of 17 sera from AD affected mink.Passive haemagglutination demonstrated antiglobulin factor in a titre of 1: 16 and higher in 20 of 36 sera (55.6 %). In 6 of 9 control sera the titre was 1: 8. To demonstrate DNA antibody the mink sera were tested by countercurrent electrophoresis against heat‐inactivated DNA. This technique showed that 4 of 12 control sera contained DNA antibodies in low titre. In mink with AD the titres were markedly higher and reached peaks 2–3 or 5–8 months after infection. No reproducible results were obtained with the Coombs test.The results of these experiments allow the following conclusions: Antinuclear antibodies probably represent a serological epiphenomenon and are only slightly increased in the course of persistent AD infection. DNA antibodies, as demonstrated, possibly have some significance in the pathogenesis of AD since the titre of these autoantibodies is markedly higher than in uninfected controls.RésuméRecherches sur la pathogénèse de la maladie aléoutienne du vison IX. Apparition d'autoanticorpsCe travail a cherché à définir le genre d'autoanticorps apparus chez des visons lors de maladie aléoutienne expérimentale et à déterminer la hauteur du titre des anticorps dans les différentes phases de la maladie. Les méthodes utilisées comprenaient le test d'immunofluorescence indirecte, l'hémagglutination passive, l'électrophorèse à contre‐courant et le test des anti‐globulines (test de Coombs). On a recherché l'apparition d'anticorps antinucléaires chez 23 sérums de visons infectés expérimentalement avec le virus de la maladie aléoutienne et chez 12 sérums de contrôle. Les substrats utilisés furent du foie et des reins de vison et de rat, des érythrocytes de poule, des leucocytes de porc et des thymocytes de veau.Un facteur antinucléaire avec un titre bas est aussi apparu chez quelques visons de contrôle non infectés. Une montée du titre des anticorps a été observée après l'infection avec le virus de la maladie aléoutienne, un pic étant atteint 3 à 4 mois après l'infection. Une tentative de mettre en évidence des anticorps contre les antigènes cytoplasmiques dans les épithéliums tubulaires des reins et des canaux biliaires du foie est restée négative. Il y a eu des titres d'anticorps de 1: 4 à 1: 256 contre la musculature lisse chez 2 des 4 sérums de contrôle examinés et chez 16 sur 17 sérums testés de visons atteints de maladie aléoutienne.On a mis en évidence un facteur antiglobulinique avec un titre de 1: 16 et plus chez 20 des 36 sérums testés (55,6 %) avec l'hémagglutination passive Le titre fut de 1: 8 chez 6 des 9 sérums de contrôle testés. Pour la mise en évidence d'anticorps‐ADN, on a testé les sérums de vison en électrophorèse à contre‐courant vis‐à‐vis d'ADN dénaturé à la chaleur (SS‐ADN). Cette Méthode a montré que 4 des 12 sérums de contrôle examinés possédaient des anticorps‐ADN avec un titre bas. Le titre était nettement plus élevé chez les visons atteints de maladie aoéoutienne pour atteindre les valeurs les plus élevées 2 à 3 et 5 à 8 mois après l'infection. On n'a pas obtenu de résultats reproductibles avec le test des antiglobulines (test de Coombs) direct et indirect. Les résultats permettent de tirer les conclusions suivantes:Les anticorps antinucléaires représentent vraisemblablement un épiphénomène sérologique et ne sont que faiblement renforcés au cours d'une infection persistante de maladie aléoutienne. Les anticorps‐ADN mis en évidence pourraient avoir une signification dans la pathogénèse de la maladie aléoutienne étant donné que le titre des autoanticorps était plus élevé chez les animaux infectés que chez les animaux de contrôle.ResumenEstudios sobre la patogenia de la plasmacitosis del visón europeo IX. Presencia de autoanticuerposSe definió en el trabajo presente la especie de los autoanticuerpos que se presentan en la enfermedad aleutiana (EA) experimental de los visones europeos y se valoró el nivel de los títulos de anticuerpos en las distintas fases nosológicas. Las técnicas empleadas abarcaban la prueba indirecta de inmunofluorescencia, hemoaglutinación pasiva, electroforesis de contracorriente y prueba de antiglobulina (prueba de Coombs). Se examinaron en total 43 sueros sanguíneos de visones europeos infectados experimentalmente con el virus EA así como doce sueros sanguíneos testigos en cuanto a la presencia de anticuerpos antinucleares. Los substratos utilizados eran hígado y ricción con el virus EA se observaba un aumento en el título de anticuerpos con una cima a los 3 o 4 mesespost infectionem.Resultaron negativos los ensayos para identificar anticuerpos contra los antígenos citoplasmáticos en epitelios tubulares del riñón y epitelios de los conductos biliares del hígado. Se hallaban presentes anticuerpos frente a la musculatura lisa en dos entre cuatro sueros testigos examinados y en 16 entre 17 sueros contrastados de visones con plasmacitosis con títulos desde 1: 4 hasta 1: 256.Mediante hemoaglutinación pasiva, se pudo identificar en 20 entre 36 sueros (55,6 %) examinados el factor antiglobulínico con un título de 1: 16 y mayor. En seis de nueve sueros testigos eran los títulos del orden de 1: 8. Para identificar los anticuerpos ADN se contrastaron los sueros de visón en la electroforesis de contracorriente frente a ADN desnaturalizado por medio de calor (ss‐ADN). Se pudo comprobar con este método que cuatro de doce sueros testigos examinados contenían anticuerpos ADN con título bajo. En los visones con la enfermedad aleutiana se encontraban títulos mucho más elevados, alcanzando cimas 2 hasta 3 resp. 5 hasta 8 mesespost infectionem.Con la prueba directa resp. indirecta de la antiglobulina (prueba de Coombs) no se pudieron lograr resultados reproducibles.Las resultancias de este estudio permiten sacar las conclusiones siguientes: Los anticuerpos antinucleares constituyen con toda probabilidad un epifenómeno serológico y se forman de manera ligeramente acrecentada en el curso de la infección viral EA persistente.
