SummaryBlood parasites of wildliving ruminants in AustriaIn a study on blood parasites of wildliving ruminants in Austria 10 fallow deer(Dama dama), 1 sika deer(Cervus nippon), 44 red deer(Cervus elaphus), 3 Père David's deer(Elaphurus davidianus), 105 roe deer(Capreolus capreolus), 12 chamois(Rupicapra rupicapra), and 14 moufflons(Ovis ammon musimon)were examined. Latent asymptomatic infections withTheileriaspecies andBabesiaspecies were recovered in red deer (68.2%) and roe deer (32.4%). Acute fatal cases of babesiosis were observed in roe deer and Père David's deer.Theileriaspecies inDama dama, Cervus elaphusandCapreolus capreolus, Babesiaspecies inCervus elaphusandTrypanosoma (Megatrypanum)species inCapreolus capreolusare recorded for the first time in Austria. SummaryLeptospirosis in West Germany Results of a survey of Leptospirosis in animals in the year 1984Following the 10th Conference of the Regional Commission of the O.I.E. for Europe held in London in 1982, the Government of the Federal Republic of Germany was requested to launch a survey regarding the incidence of leptospirosis (taking into account the serovars) and its geographical distribution in farm animals and dogs in the F. R. G.Altogether 31,000 animals were tested for leptospirosis by the microscopic agglutination test (MAT) using 10 serovars ofLeptospira interrogans.The total numbers (rounded‐off figures) of the species tested and the percentages of positive reactors were as follows: Bovines: 23,000 — 1.6%; pigs: 1,800 — 1.2%; sheep: 3,000 — 14.4%; goats: 700 — 0.3%; horses: 2,000 — 4.5%; dogs: 300 — 8.4%.The majority of positive reactors was detected in Southern Germany. Serovar hardjo was most frequently found in cattle and sheep, and serovar grippotyphosa was most frequent in horses. In pigs, serovars saxkoebing and grippotyphosa were the most prominent infective agents.ZusammenfassungSerumproben von 30963 Tieren wurden mit Hilfe der Mikroagglutinationsreaktion (MAR) unter Verwendung 10 verschiedener Serovare vonLeptospira interrogansuntersucht. Von 23093 Rindern reagierten 373 (1,6%) und von 1835 Schweinen 22 (1,2%) in der MAR positiv. Von 3040 Schafen zeigten 438 (14,4%) positive Reaktionen. Dagegen hatten von 694 Ziegen nur 2 (0,3%) Titer von ≥ 1:400. Von 2002 untersuchten Pferden waren 90 (4,5%) positiv bei einem Titer von ≥ 1:800. Bei 299 Hunden betrug der positive Anteil 25 Tiere (8,4%). Insgesamt waren von 30963 untersuchten Tieren 950 (3,1%) auf Leptospirose positiv.Beim Rind und Schaf warLeptospira interrogans serovar hardjodas in der MAR am häufigsten reagierende Antigen gefolgt von den Serovarengrippotyphosaundsaxkoebing.Bei Kreuzreaktionen wurde die Serovar angegeben, die den höchsten Titer aufwies. SummaryOn some problems in the demonstration ofHaemophilus somnus‐antibodiesAgglutination titres againstHaemophilus (H.) somnuswere found in 226 (67.1%) of 337 serum samples of cattle collected at random with titres ranging between 1:8 and 1:128. Comparative testing ofH. somnus‐antigens prepared from 25 field isolates and 2 references strains against a rabbit‐antiserum revealed titre differences from 1:20 to 1:320, suggesting antigenic varieties of the strains. Using an enzyme‐immuno‐assay similar differences could not be detected in 8H. somnus‐strains, even when the antigens had been extracted by heat, guanidinium chloride, sodium dodecylsulfate or phenol, respectively. Comparative investigations with antigenic preparations of other gram‐negative bacterial species(H. parasuis, H. pleuropneumoniae, H. equigenitalis, Pasteurella haemolytica)resulted in a significant cross reactivity of the various antigens. A better differentiation against the heterologous species could be obtained by phenol‐extracted antigens ofH. somnusimplicating that those antigen‐preparations, if further purified, may be useful for application in enzyme‐immuno‐test‐systems.ZusammenfassungBei serologischen Untersuchungen von 337 willkürlich ausgesuchten Rinderseren auf agglutinierende Antikörper gegenHaemophilus (H.) somnuswurden in 67,1% der Seren Agglutinationstiter zwischen 1:8 und 1:128 festgestellt. Das verwendete Antigen wurde aufgrund einer Vergleichsuntersuchung von Antigenpräparationen aus 25 verschiedenen Feldisolaten und 2 Referenzstämmen an einem Kaninchen‐Immunserum ausgewählt. Dabei waren bei einzelnen Antigenen Unterschiede in der Höhe der Agglutinationstiter zwischen 1:20 und 1:320 aufgetreten, was auf Antigen‐Variationen zwischenH. somnus‐Stämmen hindeutet. Derartige Stamm‐Unterschiede konnten allerdings im ELISA an dem gleichen Kaninchen‐Immunserum nicht beobachtet werden. Auch die Verwendung vonH. somnus‐Antigenen verschiedener Herstellungsart (Hitze‐, Guanidiniumchlorid‐, Natriumdodecylsulfat‐, Phenol‐Behandlung) ergab keine signifikanten Differenzen zwischen 8 untersuchten Stämmen. SummaryAn ELISA which detects bovine circulating IgG mycobacterial antibodies usingM. bovisPPD as antigen was assessed. PPDs prepared from unheated cultures of twoM. bovisstrains were compared with autoclaved bovine PPD; the latter was found to be a more reliable antigen.Ninety per cent of bacteriologically confirmed tuberculous cattle were detected by this method (18/20), whereas 89.8% of negative reactions (44/49) were observed in healthy cattle from a tuberculosis‐free area. Antibody levels of tuberculin negative cattle from an endemic area did not differ significantly from antibody levels of cattle from a tuberculosis‐free area.An almost complete qualitative correlation was observed between instrumental and visual readings.In the present trial, the sensitivity and specificity of the ELISA were similar to those of the tuberculin skin test at its best. In addition, operative advantages and low cost of the ELISA make of it a valuable tool for the diagnosis of bovine tuberculosis.ZusammenfassungBestimmung der Sensitivität und Spezifität eines enzymgebundenen Immunadsorptionstests (ELISA) zum Nachweis von mykobakteriellen Antikörpern bei boviner TuberkuloseEin ELISA zum Nachweis von zirkulierenden, bovinen IgG‐Antikörpern mitM. bovisals Antigen wurde überprüft. Autoklaviertes bovines PPD erwies sich, verglichen mit PPDs aus nicht erhitzten Kulturen zweierM. bovis‐Stämme, als geeignetes Antigen.Mit Hilfe dieser Methode konnten bei Rindern mit bakteriologisch abgesicherter Tuberkulose in 90% der Fälle (18/20) Antikörper nachgewiesen werden. Negative Befunde wurden dagegen zu 89,9% bei gesunden Tieren aus einem tuberkulosefreiem Gebiet ermittelt. Zwischen den Antikörpergehalten tuberkulinnegativer Rinder aus einem endemischen Gebiet und Tieren aus dem tuberkulosefreien Gebiet konnte keine signifikante Abweichung festgestellt werden. Eine nahezu vollständige Korrelation bestand zwischen instrumenteller und visueller Ablesung.In der vorliegenden Untersuchung waren die Ergebnisse des ELISA bezüglich Sensitivität und Spezifität ähnlich denen des Tuberkulin‐Hauttestes. SummaryBroncho‐alveolar clearance after inhalation ofPasteurella multocidaaerosol in young pigsAs invivoproof of broncho‐alveolar clearance ofPasteurella multocidaaerosol, lung washings (lavage) of narcotised young pigs (15–20 kg body wt.) were examined. For 2P. multocidastrains (capsular types A and D), clearance was studied over a period of 6 days. In the first few hours the A strain was cleared more slowly than the B strain. The rate of clearance after 24 hours was for both strains higher than 99.9%. The behaviour of the broncho‐alveolar cells after the aerosol uptake as well as the involvement of alveolar macrophages and neutrophil granulocytes in the clearance reactio are described.ZusammenfassungAlsin vivo‐Prüfverfahren der bronchoalveolären Clearance nach Verabreichung vonPasteurella (P.)‐multocida‐Aerosolen hat sich die Lungenspülung (Lavage) am narkotisierten Läuferschwein bewährt. Für zweiP.‐multocida‐Stämme (Kapseltyp A und D) wurde die Clearance über einen Zeitraum von 6 Tagen bestimmt. SummaryEighteen strains ofActinobacillus seminisisolated from ovine semen samples and epididymitis were characterized according to their cultural and biochemical behaviour. The semen samples were collected from young, unmated and mature breeding rams suffering from epididymitis or subclinical genital infection.AllA. seminisstrains were non‐motile, non spore‐forming, uncapsulated, Gram‐negative pleomorphic rods, approximately 1–8 μm × 0.8‐1 μm. On bovine blood agar they formed low convex, greyish‐white, non‐adherent and non‐haemolytic colonies, which grew under aerobic conditions, but more luxuriantly when cultures were incubated in an atmosphere containing 10% CO2at 37°C.All strains were catalase, oxidase, nitrate and ornithine decarboxylase positive, but indole, urease, phosphatase and beta‐galactosidase (ONPG) negative. They were fermentative in the O/F test and usually produced acid within 1 to 3 days from glucose, arabinose, fructose, maltose and xylose, but not from lactose, mannose and sucrose.In the agar gel diffusion test, antigenic differences were demonstrated among the isolates. SummaryThree different species ofYersinia(5Yersinia enterocolitica, 3Yersinia intermediaand 7Yersinia frederiksenii, were isolated from the content of the posterior intestine of 15 out of 148 specimens ofCyprinus carpio(carp) captured in the river Valdivia (southern Chile).All the strains ofY. enterocoliticacorresponded to biotype 1 and phagetype Xz; the serovar from two of them was 0:7,8 and the remaining three were 0:4, 0:6 and 0:16.The threeY. intermediastrains were different (1/4/Xz; 1/non‐agglutinable/Xo and 5/non‐agglutinable/Xz). SummaryThe effect of feeding bacitracin methylene disalicylate (BMD) on the tetracycline resistance status of faecalEscherichia coliwas investigated in commercially reared pork pigs. The trial continued for almost three months. During this period the magnitude of tetracycline resistance was determined inE. colifrom both treated and non‐treated pigs.
