SummaryIn vitrochallenge tests of leucocytes from patients with intrinsic asthma have shown increased amounts of histamine release on challenge with anti‐IgE in 60% and anti‐IgG in 30%. These results suggest that the leucocytes of patients with intrinsic asthma are coated with increased amounts of IgE and to a lesser extent IgG in the same proportion and amount as is found in patients with extrinsic asthma. However, in contrast to extrinsic asthma, total serum IgE levels were normal in nineteen out of twenty cases. There were no clinical differences detected between those groups of patients with and without excessive histamine release in the reversed leucocyte anaphylaxis, except that all of the small number of sera in which autoantibodies of various types were detected showed absence of excessive response in this test. SummarySera of 50% of persons extremely sensitive and of 30% of persons moderately sensitive to milk had heat‐labile reaginic antibodies conferring passive cutaneous anaphylactic sensitivity on monkeys for 24 hr. More persons in each group had IgE antibodies that bound radio‐isotope labelled β‐lactoglobulin and passively sensitized monkey basophils for anaphylactic degranulation.These tests, however, are not certain evidence of clinical allergy, because of the sera of nineteen topic children, three mediated monkey PCA tests and five had γE anti‐$bt‐lactoglobulin antibodies detected by antigen binding or basophil degranulation, though the serum donors were not clinically allergic to milk. Similarly few normal, milk‐tolerant adults also had γE anti‐β‐lactoglobulin antibodies.Sera of nine children with milk‐induced gastroenteritis occurring 12–36 hr after exposure had no anti‐milk reagins, and only one had γE binding β‐lactoglobulin. This allergy may be mediated by heat‐stable IgG antibodies, because three sera conferred milk sensitivity on monkey skin for 4 hr and two others for 2 hr. The three sera had high titres of IgG agglutinins, one to β‐lactoglobulin, one to α‐lactalbumin and one to casein. All three fixed complement in the presence of milk, and one had precipitins. They did not passively sensitize human mast cells or monkey basophils for anaphylactic degranulation, but they prepared human lung for anaphylactic release of hist‐amine, though in small amounts compared to that released from lung prepared with reagins.The sera of two of three cases of cot death had

SummaryThe ability of a series of mercaptans, i.e. 2‐mercaptoethanol, 1‐cysteine, C‐acetyl‐cysteine, N‐acetyl‐cysteine, β‐β‐dimethyl‐cysteine and cysteamine, to inactivate skin sensitizing antibodies in the serum of ragweed allergic individuals was studied. Followingin vitroreductions and alkylations, 2‐mercaptoethanol was shown to be the only mercaptan capable of destroying completely reaginic activity at 0.08–0.1 M. The other mercaptans, under similar conditions, were able to reduce only partially the skin‐sensitizing activity. Some reaginic activity was restored if reoxidation of the reduced allergic serum was not prevented. However, it was shown that mixtures of allergic serum and mercaptan, when injected into monkey skin, did not regain skin sensitizing activity. SummaryTwo aspects of the role of infection in asthmatic attacks in children have been investigated. Virus isolations from children admitted to a paediatric department during 1968 showed close correlation with the incidence of admissions with upper respiratory tract infection and with lower respiratory tract infection. The peaks of virus isolation corresponded to the peaks of respiratory infection admissions. There was a clear dissociation between virus isolations and admissions with acute asthma. Fifty‐one children admitted with clinical acute attacks of asthma were investigated more thoroughly. Only three viruses were isolated from these case? compared to eleven viruses isolated from a control group of children with respiratory infections. The incidence of bacterial pathogens was similar in both groups. Serological evidence of virus infection was obtained in two out of twenty‐six children with asthma and in four out often children with respiratory infections. SummarySkin, nasal and bronchial tests with extracts ofAspergillus fumigatusand in particular with its protein fraction elicit dual. Type I plus Type III, allergic reactions in all three organs in patients suffering from allergic broncho‐pulmonary aspergillosis.The immediate asthmatic reaction is readily reversible and less severe than the late asthmatic reaction which lasts for many hours and is only temporarily reversed by the inhalation of isoprenaline. SummaryThe sera of persons with cutaneous vasculitis following bacterial infection were examined for antibacterial anaphylactic antibodies that could have predisposed tissues to deposition of harmful antigen‐antibody complexes.Sera of persons with or without cutaneous vasculitis after bacterial infection had no reagins to streptococcal or staphylococcal cellular or extracellular antigens detectable by monkey passive cutaneous anaphylaxis or histamine release from monkey lung, and no IgE antibodies to the extracellular antigens were found by radio‐immunodiffusion precipitation gel tests. One person with, and one without, vasculitis after streptococcal tonsilitis had heat‐stable short‐term passive sensitizing antibodies mediating increased vascular permeability in monkey skin, and one person had reagins toCandida albicanscellular antigens.The sera of three persons sensitive to tetanus toxoid were used to control the reliability of the above tests. One serum mediated monkey PC A and histamine release from monkey lung, and all three were found to contain IgE anti‐tetanus toxoid antibodies.It is concluded that either small amounts of IgE reagin to lipopolysaccharides are not detected due to technical difficulties, or anaphylactic sensitivity to polysaccharides may sometimes be mediated by other classes of
