SummaryHouse dust mites are widely recognized as major factors involved in the triggering of allergic diseases such as asthma. It is now apparent that the group III allergens of theDermatophagoidesmite species may play a significant role in a number of house dust mite allergic cases. NaturalDer pIII was isolated by gel filtration of salt precipitatedDermatophagoides pteronyssinusextract and as reported previously ran as a doublet of Mr 28 and 30 K on sodium dodecyl sulphate‐polyacrylamide gel electrophoresis (SDS‐PAGE). NaturalDer fIII was isolated by affinity purification with the 5A12 monoclonal antibody. Amino acid sequence data was generated for both these proteins which was used to construct DNA probes to screen aDermatophagoides pteronyssinuscDNA library by hybridization and resulted in the isolation of a recombinantDer pIII cDNA clone, P3WSI. The 1059 bp cDNA fragment included a 786 bp open reading frame which encodes a pre‐pro region of 29 amino acids and a mature protein of 232 amino acids with a calculated Mr 24985. A search of the BLAST protein database has confirmed that theDer pIII P3WS1 clone is approximately 50% homologous with other trypsin proteins. SummaryTo investigate the year‐to‐year variation of mite antigen density (Der p I, Der f1) in dust from mattresses and the relevance of residential factors for antigen load, information derived from an epidemiologic study including two surveys carried out in the households of a cohort of elementary school children (n= 1291) was analysed.When considering residences with measurements taken in both years in question (n= 1050), rank‐correlation indicated a predominance of stability for both antigens (Der pI: rs= 0‐82,p=0.0001;Der fI: rs=0.72,P= 0.0001), Using multiple regression analyses, significant associations between antigen concentrations and a variety of residential factors were found. Use of a blanket of animal hair, use of a cover or underblanket, wet spots in the bedroom, higher relative humidity and a low storey level were significantly associated with increased concentrations ofDer pI, whereas inverse relationships between this antigen and room temperature, number of persons per m2as well as use of underfloor heating were seen. RegardingDer fI, older mattresses, use of a cover or underblanket, higher weight of sampled dust, high educational level and higher ratio of inhabitants per m2were significantly associated with increased concentrations of the antigen. On the other hand, lowerDer fI concentrations were found when interior sprung mattresses were used and when the mattress was ‘treated regularly’.In conclusion, two measurements, 1 year apart from each other, show that stability of mite antigen concentrations predominated. SummaryThe kinetics of mast cell accumulation in the nasal mucosa during allergen exposure were investigated in animal model of experimentally induced allergic rhinitis. Guinea‐pigs were divided into an unsensitized control group and five sensitized groups each containing live animals. Five sensitized groups were immunized intraperitoneally with ovalbumin, followed by intranasal administration of ovalbumin in each of four groups for 1. 2, 3 and 4 weeks, respectively. There were significant increases in the number of mast cells in the epithelium after 2 and 3 weeks in the groups receiving intranasal ovalbumin. No significant changes were detected in the lamina propria and the total number of mast cells. In the lamina propria, the distance between a mast cell and the basement membrane was significantly decreased in the groups after 2, 3 and 4 weeks of intranasal administration. These findings suggest that an infiltration of mast cells from the lamina propria to the epithelium occurred after 2 weeks of intranasal administration of allergen. An electron microscopic study showed no mast cells at the luminal surface of the epithelium and the majority of mast cells lying around the basal cells. SummaryThere is uncertainly about the relative contributions of bronchial smooth muscle contract ion. mucosal oedema and mucus plugging lo airflow obstruct ion in the allergen induced late asthmatic response (LAR).We systematically studied the ability of the inhaled β2‐agonist salbutamol to reverse the LAR in eight subjects after allergen bronchoprovocation. Salbutamol reversed the LAR by restoring FEV1, to a level similar to the initial value measured at the same time of day (18.00 h)on the previous evening. For the eight subjects studied, this initial FEV1value, measured after abstaining from β‐agonists for 8 h, was a mean ±SEM 90.7 ± 5.6% of predicted, which suggests further bronchodilation may have been possible at this time. We then studied six of the eight subjects in an identical protocol with saline challenge substituted for allergen bronchoprovocation to answer the question whether further bronchodilation was possible at that time after salbutamol in the absence of an LAR. After salbutamol on the allergen challenge day the FEV1for the six subjects was 84.1 ± 7.0% of predicted, compared with 94.0 ± 3.7% of predicted at the same point on the saline challenge day (p<0.05).We conclude that, although the LAR may be effectively reversed by β2‐agonists, there is evidence for some residual airway narrowing, presumably related lo mucosal oedema, exudate and mu

SummaryThe major allergen of timothy grass pollen (Phleum pratense), designated asPhl pV, consists of isoallergenic components of 38 and 32 kDa with pi values of 5.2 7.5 and 4.8 5 9, respectively. The different‐sized proteins reveal similarities in IgE reactivity,N‐terminal sequence and protein staining. For epitope analysis of these allergens u combination of enzymatic cleavage of electrophoretically separated proteins and iminunoblotting techniques with subsequentN‐terminal sequencing was performed. After isolation of the components from two‐dimensional PAGE gels. proteins were enzymatically cleaved and separated by SDS‐PAGE. By endoproteinase Glu‐C cleavage six IgE‐reactive fragments of each 32 kDa protein and three of each 38 kDa allergen were obtained. Microsequencing of the fragments revealed internal sequences that did not show any similarities between the different‐sized allergens. SummaryThe effect of storage and high temperatures on the stability ofSaccharomyces cerevisiaeallergens was studied by immunoblotting.Saccharomyces cerevisiaeallergic serum pool and125I‐ and galactosidase‐labelled anti‐IgE were used in the assays. Freeze‐dried extracts were reconstituted with saline and with 50% glycerol and then stored at room (+ 20°C) and refrigerator temperature (+6°C) for different time periods.The stability was better in 50% glycerol at +6°C than at room temperature without glycerol. However, after 1 month, two of the most important allergens ofS. cerevisiae,the 48 and 32 kDa protein allergens. lost their IgE‐binding capacity even in the extracts stored with 50% glycerol al +6 C. The 45 kDa allergen was. on the other hand, quite stable after storage for 9 months at +6°C. Although the beneficial effect of 50% glycerol was clear, storage at +6°C, even with 50% glycerol should not exceed 1 month forS. cerevisiaeextracts.Two commercially availableS. cerevisiaeextracts in solution with valid expiry dates were also analysed. They had only little allergenic potency, while a freeze‐dried extract stored for 8 years showed good allergenic potency. SummaryIn the present study, allergens ofCladosporium herbarumwere analysed by SDS‐PAGE. two‐dimensional gel electrophoresis and immunoblot. The SDS‐PAGF under reduced conditions followed by western blotting analysis revealed 10 human IgE‐binding components with molecular masses ranging from 15 to 143 kDa. Within the same molecular mass range, five components were detected under non‐reduced conditions. Examination of the specificities of 10 individual sera revealed that two patients were sensitive to almost all the allergens, whereas the majority reacted with only some components. Two‐dimensional immunoblot was employed to further analyse the allergens in the extract.
