SYNOPSISThe exocrine pancreatic cell of the mouse was the test object for the investigation. Tissue fixed with a 0.5 p.c. aqueous solution of chromium trioxide was embedded in Araldite and sections were examined with the electron microscope.Chromium trioxide does not convert the cytoplasm into a coagulum. What appears to be a coagulum in optical microscopy is in fact a three‐dimensional network of ergastoplasm (endoplasmic reticulum), the cisterna of which retain their identity (though the ribosomes are not preserved).The mitochondria are not destroyed, but no internal structure is seen. No vestige of the Golgi element remains.The inner nuclear membrane is preserved, the outer only here and there. Nothing that could be interpreted as a thread‐like structure is seen in the nucleus. SYNOPSISThe preservation and identification of free lipids is most difficult because they are very readily dissolved by the usual embedding media. At present only unsaturated triglycerides fixed by postchromation and lipofuscins can survive this extraction. There is no general fixative for lipids. A few unusual embedding media will preserve small droplets of lipids, but they are technically far from satisfactory. Included in this group is a new medium—urea‐formaldehyde. SYNOPSISThe 1–2 μ protein fibres forming an internal lining to the fragile arenaceous shell ofHaliphysemaare made of 0.2 μ diameter, cross‐striated fibres, which have an axial period of approximately 670 A and six distinct intraperiod bands. It is suggested that the fibres may be collagen, a fibrous protein generally considered to occur in all animal phyla except the Protozoa. SYNOPSISA technique is described for the storage of biological materials in air tight sachets made from a new aluminium‐polythene laminate. Tissues containing highly mobile soluble radioactive material have been stored for periods up to six months in these sachets which are stored in a liquid nitrogen refrigerator.
