SYNOPSISCarmine particles and lipids are taken up by the digestive cells of the digestive gland ofHelix. These substances are taken up by phagocytosis, and the apical and “green” granule vacuoles (Sumner, 1965) of the digestive cells are phagocytosis vacuoles. Carmine eventually becomes incorporated into the small yellow granules showing that they are derived from the “green” granules. SYNOPSISClassical expositions of the theory of blood coagulation hold that consolidation of the fibrinous reticulum is due to the mechanical enmeshment of erythrocytes within it. Studies of coagulating blood by dark‐ground microscopy have revealed the attachment of submicroscopic threads to the erythrocyte walls at precise loci: it is postulated that the formation of captive cells (penderocytes) leads to haemostatic occlusion and the total retention of erythrocytes within the contracting coagulum. SYNOPSISNew observations are presented on the oxyntico‐peptic cells ofGallus domesticus. In the control animals (which were starved for 72 hours) acid phosphatase is present below the nucleus in the cytoplasm of the majority of the cells lying in the upper part of the alveoli. Following the administration of histamine, there is an increase of acid phosphatase. In cells situated in the upper part of the alveoli it is present above and alongside the nucleus. In many cells lying in the lower part of the alveoli it occurs chiefly in an infranuclear position, although in some it is above and alongside the nucleus. There is a progressive decrease in the number of pepsinogen granules, and in cells in many areas of the mucosa, this is preceded by a progressive decrease in the intensity of colouring of the granules. Evacuation of the granules commences in cells lying in the lower part of the alveoli and proceeds to those that are situated in the upper part of the alveoli. The larger type of granule is discharged first, and 2 to 3 hours after injection of the drug all the granules have been discharged from the cells. It seems probable that the pepsinogen granules reappear in cells found in the basal half of the alveoli about 6 hours after histamine injection. Following the injection of pilocarpine, very large spheroids which stain an intense blue after Bowie's method are found above the nucleus in cells lying in the upper part of the alveol

SYNOPSISA general problem encountered in quantitative histology is the error caused by the inclusion of fragmented cells in the observed count from histological sections. The result of including such fragments is an overestimate of the number of cells. With this problem in mind, a method is given for estimating cell numbers in systems where cells are mononuclear with single nucleoli. The method involves applying a correction to the difference in observed counts from adjacent histological sections of different thicknesses. The proposed method is based on the same formulation of this cell counting problem as is the method of Floderus, but a major distinction, however, is that the present method does not require the estimation of the mean particle diameter or vertical height of the smallest visible fragment and thus eliminates a potential source of error.The correction formula is derived by considering a simple model for this cell counting problem. A hypothetical cell population is studied to show the theoretical equivalence of the proposed method and that of Floderus and also to show the potential bias caused by using Abercrombie's formula which does not take into account the effect of invisible fragments. The new method is compared with other available techniques and is empirically evaluated by extensive counting in a human superior cervical sympathetic ganglion.
