SYNOPSISOn comparing the electron‐microscope appearance of, and the sites of tritiated thymidine uptake within, the nuclei of cartilage cells in the growing epiphysis, a corresponding pattern of distribution of electron‐dense material and isotope was found. Similarly, cells capable of tritiated thymidine uptake, and in which metaphase arrests can be produced with colchicine, were found to have electron‐dense nuclei on examination in the electron microscope. When the pattern of tritiated thymidine was changed by the prior administration of growth hormone to the animal, the distribution of electron‐dense nuclei showed the same changes.During the active phase, direct physical connections were observed to exist between the nuclei and surroundings of the cartilage cells. The effect of vitamin C deficiency was to inhibit the formation of endoplasmic reticulum, while leaving the connections between the nucleus and exterior of the cell intact.The main effect of colchicine was to remove the connections between the nucleus and its surroundings, while the nucleolus was frequently enlarged. (The central part of the nucleolus is normally connected directly with a large cytoplasmic inclusion.) SYNOPSISCoal, coke and some other carbonaceous materials present special difficulties in both thin sectioning and polishing, whether the sample is granular or in lump form. Improved techniques using polyester resin have been worked out, and have been found more reliable and convenient than other methods of thin sectioning and polishing. Large parallel‐sided thin sections, wholly enclosed in plastic, are useful for studying and demonstrating comparatively gross structures, but may also be used at high magnification to study fine detail. SYNOPSISA fibre microtome is described which combines the simplicity of the plate method of sectioning with the accuracy and efficiency of the Hardy fibre microtome. The instrument is cheap to make, uses only standard materials and may be constructed by anyone having access to a small lathe and simple hand tools.
