AbstractPeripheral blood lymphocyte counts were determined in 57 breast clinic patients. These patients were grouped into four clinical/pathological groups: 18 had benign breast disease, 13 had a history of breast cancer but were free of disease at the time of the study, 7 had a history of breast cancer and were free of disease at the time of the study but were on adjuvant chemotherapy, and 19 had active metastatic breast cancer. Two parameters were investigated in a double blind study: 1) the absolute lymphocyte counts and 2) the percentages of lymphocytes that bound different bacteria as markers of lymphocyte subpopulations in conventionally stained blood smears. A significant reduction in mean lymphocyte counts was demonstrated in patients with advanced disesase. A significant increase in these counts was found in patients who were free of disease following surgical treatment alone. The T1T2cells (our denomination), the subpopulation of cells responsible for specific killing in vitro and for suppression of natural cytotoxic cells, was significantly reduced in patients with advanced disease under treatment. The T/B cell ratio in all groups was the same and within normal range. These observations suggest that the observed decrease in the total lymphocytes is due not only to an overall decrease in all classes of lymphocytes but especially in those responsible for specific cell‐mediated reactions. AbstractA retrospective analysis of 80 cases of seminomas treated at the University of Louisville Radiation Center from 1958 to 1979 has been done. Eight cases have been treated with a diagnosis of anaplastic seminoma. This study is a review of the eight cases, along with a review of literature, regarding the prognosis and management of anaplastic seminoma.

AbstractA case of synovial sarcoma in the abdominal wall of a 50‐year‐old Iraqi female is reported. The ultrastructure of the tumor has been studied. Eight other cases of synovial sarcoma in the abdominal wall which were reported in the English literature up to 1978 have been reviewed. Synovial sarcomas in the abdominal wall tend to occur with a much greater frequency in females in contrast to such tumors in the extremities or the neck which tend to occur with a much greater frequency in males. No ultrastructure studies of abdominal wall synovial sarcomas could be found in the English literature. Ultrastructure characteristics of this tumor were similar to those arising in the usual sites and in the neck. AbstractSixty‐four adult patients with soft‐tissue sarcomas received adjuvant chemotherapy with a six‐drug combination regimen after surgery. Seventy percent remain free of disease with a median follow‐up of 50 months. Only seven patients have died of their disease, all within the first 24 months after surgery. Most patients experienced severe nausea and vomiting secondary to the actinomycin D and dacarbazine parts of the protocol, and three patients experienced frank Adriamycin cardiomyopathy. Toxicity from this combination was otherwise mild. The 58% recurrence rate of 12 patients who discontinued therapy early because of nausea was significantly greater than the 23% rate for those who completed or relapsed on therapy (P = 0.01). Adjuvant chemotherapy should be considered after surgery for patients with soft‐part sarcomas, especially those with high‐grade tumors that have a considerable risk of

AbstractOne hundred and fourteen cases of inflammatory breast cancer were reviewed. Actuarial survival curves, independent of the stage and type of treatment, showed a median survival of less than 15 months. As regards the three different subgroups of 80, 14, and 20 women, N0–1–2M0, N3M0, and NxM1respectively, the actuarial survival curves showed figures of median survival that decreased from 14 to 4 months, according to the extent of the disease. Survival rates for the subgroups of patients treated by radical mastectomy, radical mastectomy followed by radiotherapy, and radiotherapy alone were not significantly different. Neither the more sophisticated therapeutic approaches intended to modify the hormonal medium of the patients, nor the combined chemotherapy plus radiotherapy, proved to be successful. Inflammatory breast cancer should be considered a systemic disease; consequently, in spite of discouraging results, only a systemic therapeutic approach, consisting of aggressive combination chemotherapy and perhaps immunostimulation, seems worthwh

AbstractEffect of cryoinjury to normal rat kidney was studied at the light and electron microscope level, and characterization of the time course of pathologic changes in kidney tissue after cryosurgery was described from 1 hr up to 14 days after production of a standard cryolesion. Cryoinjury to the kidney resulted in sharply demarcated lesions with minimal inflammatory response during the period of cell destruction and healing by fibrosis. Cellular protein was completely coagulated within 1 hr after cryoinjury, with crystallization of protein and disruption of membranes. Cellular disruption was rapid, and the clearance of cellular debris from the area of injury by macrophage infiltration was accomplished through a quite mild inflammatory reaction. Supporting stromal elements of renal parenchyma remained intact, including collagen tubular framework and basement membranes, and formed the lattice over which fibrosis developed.

AbstractWe report here the lack of any relationship between immunogenicity and the potential to metastasize among four B16 melanoma variants. Groups of C57BL/6J mice were immunized with one of four B16 variant lines (maintained by serial IM passage) that were shown in separate experiments to produce the following incidences of pulmonary metastases four to six weeks after isograft excision: M1, 10%–15%; M2, 10%–20%; M3, 75%–80%; and M4, 75%–90%. After immunization, mice were challenged with one of the four variant lines in a crisscross fashion. Two challenge doses for each line were chosen on the basis of earlier experiments showing 100% and 50% challenge tumor take in nonimmunized animals. Despite earlier work published from this laboratory showing that splenocytes harvested from immunized mice could distinguish antigenic differences among the B16 variants, we could find no significant difference in immunogenicity or cross‐protection among the lines regardless of their metastatic potential. We conclude that no correlation exists between the antigenic differences shown by in vitro splenocyte‐mediated cytotoxicity assays among the B16 variants and in vivo immunogenicity or cross‐protection experiments. AbstractThe technique of flow cytometric DNA histogram analysis (FCM) shows there to be two distinct cell populations (diploid vs aneuploid) in the poorly differentiated R3327‐G rat prostatic adenocarcinoma. The following study compares tumor weight measurements with several FCM computer‐based methods designed to determine rapidly the proliferative status of tumors. Hypophysectomy, bilateral adrenalectomy, orchiectomy, sham operations, or diethyl‐stilbestrol treatments were initiated when the tumors were palpable (day 21) and continued until the tumors were excised (day 52). Hypophysectomy, orchiectomy, adrenalectomy, and diethylstilbestrol treatments all resulted in significant inhibition by tumor weight. Quantitation of the percentage of mid‐S phase aneuploid cells by summation gave the best correlation with tumor weight. Tumors grown in hypophysectomized, orchiectomized, adrenalectomized, or diethylstilbestrol‐treated animals showed a significant reduction in the proportion of mid‐S phase cells as compared with controls. The calculation of the percentage of all aneuploid cells was significantly reduced in hypophysectomy, orchiectomy, and diethylstilbestrol‐treated animals. However, tumors grown in adrenalectomized animals were not significantly different from controls by this method. Adrenalectomy was found to be the least effective form of therapy, and this was reflected in all of the parameters measured. AbstractFifty patients with gastric leiomyosarcoma seen at M.D. Anderson Hospital between 1957 and 1978 were reviewed. Symptoms included weakness, gastrointestinal bleeding, and epigastric pain. An upper abdominal mass or tenderness was the most frequent physical finding. Laparotomy was performed in all patients, with gastric resection in 86%, and resection for cure in 68%. Survival after distal subtotal gastric resection (mean: 62 months) was longer than after proximal subtotal (30 months) or wedge resection (46 months). The five‐year survival was 19% for all patients, and 32% for those operated on for cure. A favorable prognosis was predicted by mild atypia or few mitoses (grade 1). No differences in survival were demonstrated between patients with tumors of different cell types (epithelioid, spindle, or pleomorphic). No lymph node in any patient contained metastatic tumor, suggesting a lymph node dissection need not be performed. Commonly, tumor spread was to liver or lungs or by direct invasion of contiguous tissue or organs. Radiotherapy and chemotherapy are of limited value in treating unresectable disease. AbstractThe chloride salts of lithium (Li+) and cesium (Cs+) were evaluated for their ability to influence the growth of Sarcoma I implants in A/J mice. The administration of daily doses of either 1 or 3 mEq/kg CsCl to these mice reduced the incidence and size of tumor implants. This effect was not apparent in animals receiving a smaller dose (0.5 mEq/kg) of the same drug. At the time of sacrifice the serum level of Cs+in this latter group was approximately half that recorded in animals receiving the higher doses of CsCl. No effect on tumor incidence or rate of growth was observed in animals receiving different doses of LiCl. Because of the similarities that existed between cesium and potassium, it was postulated that the effect of cesium was due to alterations in the intracellular composition of the tumor cells. Also, the possible role of cytotoxic agents in potentiating the inhibitory effect of cesium on tumors was discussed.
