A simultaneous radioimmunoassay for Δ4-androstenedione (Δ4), dehydroepiandrosterone (DHA) and 11 β-hydroxyandrostenedione (11βOHΔ4) in plasma is described. This involved preparing first an anti-11β-hydroxyandrostenedione-3-0-carboxymethyl oxime/BSA antiserum which binds both Δ4 and 11βOHΔ4, and an anti-dehydroepiandrosterone-7–0-carboxymethyl oxime/BSA antiserum. A chromatographic step using celite minicolumns separates these three steroids. The method was applied to the measurement of the plasma basal values of these three androgens in control subjects. Mean concentrations (ng/ml) of Δ4, DHA and 11βOHΔ4 were respectively 1.35, 6.63 and 3.13 in males; 1.35, 6.65 and 2.59 in premenopausal females; 0.46, 1.53 and 1.38 in post-menopausal females, and 0.39, 0.73 and 1.78 in children 1–6 years of age. Dynamic tests were also carried out: ACTH stimulation was found to increase Δ4, DHA and 11βOHΔ4. Dexamethasone had a reverse effect causing a 50% diminution in Δ4 levels, a marked decrease in DHA levels, and a 90% decrease in 11βOHΔ4 levels. Metyrapone test was found to produce a 223 % increase in Δ4 levels, a 196 % increase in DHA levels, and a decrease of more than 90% in the 11βOHΔ4 levels. Estroprogestative drug treatment was accompanied by a decrease of not only Δ4, but also of DHA and 11βOHΔ4. In 8 male subjects with normal testicular function, blood was drawn from the left spermatic vein and from the right cubital vein before and during an infusion of 4,000 IU human chorionic gonadotropin (hCG) (preceded by a bolus of 1,000 IU) or of 500 IU hCG plus human menopausal gonadotropin (hMG), respectively, within 30 min. Testosterone levels in the spermatic vein blood increased rapidly up to 518 or 215%, respectively. An increase of estradiol-17β and of 5α-dihydrotestosterone concentrations in spermatic vein blood was observed only in 1 and 2 subjects, respectively. It was shown that the human testicle releases also 5α-androstane-3α,17β-diol and testosterone-glucosiduronate. Both steroids did not respond to the short-term infusion of gonadotropins. The effect of hyperglycemia on growth hormone, thyroid-stimulating hormone, and prolactin response to oral diazepam (10 mg) was assessed in 7 normal subjects. A peak growth hormone response of 13.5 ± 0.3 ng/ml (mean ± SEM) significantly above the base line (p < 0.001) was achieved when diazepam was given alone. Hyperglycemia after glucose load abolished this response. Testosterone, oestradiol-17β, progesterone and prolactin were administered for 1 and 7 days to adult rats to study the short- and long-term effects of these hormones on testicular lipids. Testosterone administration induced a marked increase in testicular phospholipids and a significant fall in glycerides. Oestrogen, on the other hand, provoked a significant increase in total lipids and glycerides with no marked changes in testicular phospholipids. Progesterone elicited significant changes in cholesterol fractions; a marked increase in esterified cholesterol with a concurrent fall in free cholesterol was observed. Prolactin treatment resulted in a significant fall in total lipids and phospholipids with no marked change in tissue weight. It also induced a marked fall in total and free cholesterol with a significant increase in esterified cholesterol

摘要:
A reproducible animal model for studying pheochromocytoma is described. Rat pheochromocytoma cells (PC-12 line) maintained in tissue culture and injected subcutaneously into rats produce a functioning tumor at the site of inoculation. Animals harboring these tumors develop alterations characteristic of pheochromocytoma including weight loss, hypertension and a contracted blood volume. This model provides an easily reproducible system for studying the pathophysiologic features of pheochromocytoma.

Oestrone sulphatase activity was determined in the 12,500 g supernatant of rat uterus homogenates. The conditions used were 0.5 mM oestrone sulphate and 30 min incubation at pH 8.0 and 37 °C. The enzyme activity in uteri from normal animals was found to be 5.1 ± 0.7 nmol (mg protein)-1h-1. In ovariectomized and hypophysectomized animals the values were significantly higher, whereas subcutaneous administration of 17β-oestradiol to such rats resulted in significantly decreased oestrone sulphatase activities. Progesterone increased the enzyme activity of intact rats. It is concluded that the uterine tissue of rats contains oestrone sulphatase which may be of biological importance and which is inhibited by oestrog

Testicular response to human chorionic gonadotropin (hCG) was studied in male lambs. Adenosine 3’:5’-cyclic monophosphate (cAMP), testosterone (T), Δ4-androstenedione and 17α-hydroxyprogesterone content and cAMP and T production by dispersed interstitial cells were assessed in control and hCG-pretreated animals. Plasma T levels increased after hCG at 1, 4 and 8 weeks. Increments in the testicular content of cAMP, Δ4-androstenedione, and T were greater at 8 weeks and that of 17α-hydroxyprogesterone and 125I-hCG binding to dispersed interstitial cells were identical at all ages. cAMP and T production by dispersed interstitial cells from nonstimulated animals and the response to hCG and choleratoxin were similar in all lambs. In contrast, cAMP and T production were higher at 1 week only in animals pretreated with hCG in vivo. These data are compatible with hCG-induced desensitization at 4 and
