4 weeks after surgical ablation (MHA) of the medial basal hypothalamus (MBH) in adult male rats, serum levels of prolactin, LH, and LH-RH were determined by radioimmunoassay. Blood samples were obtained by jugular venipuncture under ether anesthesia (designated ‘stress’ samples) and 30 min after the ether stress by rapid decapitation (designated ‘resting’ samples). 30 min after ether, serum levels of prolactin, LH, and LH-RH in MHA rats were comparable to those of intact and sham-operated controls. Among intact and sham-operated rats, ether elicited an initial increase in prolactin but not in LH or LH-RH. In the MHA group, prolactin levels were also acutely increased, although the increment was not as great as in control groups. The data indicate that considerable basal prolactin and LH secretion persists after MHA, and that this continued secretion may be regulated by neurohormones such as LH-RH which arise from areas outside

In adult male rats, a pretreatment regimen of serial injections of dexamethasone (1 mg/kg), morphine (20 mg/kg) and pentobarbital (40 mg/kg) was evaluated for use in conjunction with studies on the effects of hypothalamic electrical stimulation on prolactin secretion. Serum prolactin levels were measured before and 15 min after electrical and sham stimulation of the hypothalamic arcuate nucleus in rats subjected to either the pharmacological regimen or to pentobarbital anesthesia alone. Pentobarbital alone caused a transient rise in serum prolactin levels, which obscured any effect of electrical or sham stimulation; this interference was not overcome by the addition of dexamethasone and morphine treatment. Thus, the results indicate that the acute stimulatory effect of pentobarbital anesthesia on prolactin release may interfere with further manipulation of prolactin-controlling mechanisms, by either pharmacological or surgical means. Furthermore, the dexamethasone-morphine-pentobarbital pretreated rat does not provide an adequate preparation for studying the effects of electrical stimulation on prolactin secretion.

Two 15-year-old boys with primary hypogonadism had evaluation of their hypothalamic-pituitary-gonadal axis. Testicular biopsies and chromosomal studies were also performed. Both patients presented with delayed puberty and short stature and had prepubertal LH, FSH and testosterone concentrations. Serial 24-hour frequent-interval blood studies over a 2-year period in one patient (R.F.) showed a gradual progression from a normal early pubertal LH secretory pattern to one characteristic of ‘primary’ testicular failure. The testicular biopsies showed prepubertal tests with no significant germinal cell maturation. Although both patients had some somatic stigmata of Noonan’s syndrome, they had different karyotypes (XY and XYq-). 摘要:
Testicular tissues obtained from 12 cases of testicular feminization syndrome were subjected to lipid analyses. The total lipids formed 2.75% of the total wet weight of the testicular tissue. Total cholesterol represented 30%, glycerides 33% and phospholipids 37% of the total lipids, respectively. Fractionation studies revealed that free cholesterol formed 66.7% and esterified cholesterol 33.3% of the total cholesterol. Triglycerides represented 89.89% and monoglycerides 10.11% of the total glycerides. Separation of phospholipids showed that phosphatidyl choline (46.54%) and phosphatidyl ethanolamine (24.89%) to be the major phospholipid classes.

Changes in the morphology and steroid content of ovaries were studied after 48 h of intravenous injection of 100 µg of cyproterone acetate or flutamide to diestrus or estrous rats. Treatment with cyproterone acetate at diestrus caused a decrease in the number of small follicles ( < 200 µm), freshly formed corpora lutea and the levels of estradiol-17β in the ovary, suggesting inhibition of ovulation. Following flutamide administration at diestrus, the number of follicles at all stages of development were reduced with a concomitant decrease in the ovarian levels of the hormones. Thus, flutamide suppressed the growth and maturation of follicles. On administration of these drugs at estrous, the steroid content of ovaries was more pari passu with the increase in the number of mature and medium follicles.
