Reappraisal of Bromocriptine Treatment for Acromegaly |

摘要:
11 acromegalics were treated with bromocriptine for 2–18 months. Their hormonal response was assessed by an acute suppression test with bromocriptine (AST), an oral glucose tolerance test (GTT), and by measuring growth hormone (GH) concentrations during a day of hospital life. The GTTs and the 24-hour profiles were performed before and after bromocriptine. During the AST all patients showed a decrease of GH concentrations ranging from 33 to 86% of the basal. Following bromocriptine, the mean GH concentration was lowered in 7 out of 11 patients during the GTT, and in 8 out of 11 during the profile, but it was within the normal range in 4 patients only during the GTT, and in 1 during the profile. Bromocriptine normalises radioimmunoassayble GH levels in a percentage of patients (12%) which is less than those following conventional treatment of acromegaly, surgery (80%) and pituitary irradiation (70%). Clinically, however, bromocriptine was more effective than judged by the changes of GH levels. Subjective and objective symptoms of acromegaly, such as articular pain, excessive sweating, hypertension, amenorrhoea, urinary hydroxyproline excretion and heel pad thickness decreased in our patients after bromocriptine. A specific action of bromocriptine on the degradation rate of ‘little’ GH may result in a selective reduction of the bioactive monomeric component of GH and may explain the discrepancy between the clinical and the biochemical response to bromocriptine. The effects of prednisone therapy and the withdrawal of prednisone for 3 days on hormonal relationships was investigated in 6 adolescent patients, age 10–19 years with congenital adrenal hyperplasia (CAH) due to 21-hydroxylase deficiency. Prednisone continuously suppressed adrenal release of 17α-hydroxyprogesterone (17-OHP) in 4 patients and adequately controlled 17-oxosteroid (17-OS) excretion in 5. Serum dehydroepiandrosterone sulfate (DS) concentrations were not elevated during or 3 days after withdrawal of medication. Growth hormone (GH) secretion was not suppressed by prednisone. Withdrawal of treatment was associated with a decrease in the follicle-stimulating hormone (FSH) to luteinizing hormone (LH) ratio in 5 patients and a rise in serum testosterone in 4. The clinical courses of these patients emphasize the difficulty in achieving optimum control. 摘要:
The action of prolactin on RNA synthesis in cultured mouse mammary gland explants becomes manifest when the tissues are exposed only briefly (1 h or less) to prolactin. In contrast, the action of prolactin on casein synthesis only becomes apparent when the tissues are cultured for 6 h or more with prolactin. Once the actions of prolactin on RNA and casein synthesis are initiated, however, these effects persist for hours or days, even when the tissues are subsequently cultured in the absence of prolactin.

摘要:
Estradiol binding in preparations of female rat thymi is demonstrated by three different approaches (dextran-coated charcoal method, immunofluorescence technique and thin-layer gel filtration). Scatchard’s analysis of [3H]-estradiol proves consistent with the existence of a single class of receptor sites, having a dissociation constant (Kd) of about 2 X 10-10M. Steroid binding specificity in the thymus is similar to that found in the uterus. On thin-layer gel filtration the binding appears in the 8S region similar to that observed for the uterus. 摘要:
The endometrial blood flow in rats at the site of a progesterone-containing silastic IUD was compared to that from empty horns or horns containing an empty IUD. The blood flow was determined by injecting 85krypton saline into the aorta and measuring the clearance from the endometrium with two mini Geiger-Müller tubes inserted into the uterine lumen. The endometrial blood flow around the hormone-containing IUDs was 0.81 ml g-1 min-1. This flow was not different from that found in the two other experimental situations, indicating that blood flow changes is not a major mode of action of either progesterone-containing IUDs or empty silastic IUDs
