The effect of a new analogue of the gonadotrophin-releasing hormone LH-RH, D-Ser(TBU)6-EA10-LH-RH, on the secretion of LH, FSH, as well as testosterone, oestrone, oestradiol, HGH, prolactin, TSH, and cortisol was studied in normal men. The same subjects were injected intravenously in 4-day intervals with 1.0, 2.5, 5.0, and 10.0 µg of this substance. A significant LH but no FSH release was seen after doses of 1.0 and 2.5 µg LH-RH analogue, while after 5.0 and 10.0 µg dose-dependent increases of LH and imposed elevations of FSH were observed. Peak levels of LH were reached after 30 min, those of FSH after intravenous injection after 120 min. LH and FSH remained elevated for 8–10 h. LH peak levels after 5 µg of LH-RH analogue were comparable to those seen after injection of 100 µg of the decapeptide LH-RH. Following the release of LH and FSH after doses of 5.0 and 10.0 µg LH-RH analogue, there was a late stimulating effect in testosterone and oestradiol secretion. 摘要:
The possible role of endogenous prolactin (hPRL) in the regulation of renal water, Na and K excretion during sleep was tested in a group of 10 healthy female volunteers. Plasma hPRL and total urinary Na and K excretion were measured at 2- and 4-hourly intervals, respectively, in the control period and after prolactin inhibition with bromocriptin. Despite adequate prolactin suppression, no significant changes were observed in the nyctohemeral excretion rhythms of water, Na and K, suggesting that endogenous prolactin is not instrumental in the control of these parameters.

Concentrations of testosterone, △4-androstenedione, dihydrotestosterone and Δ5-androstenediol were measured in 12 patients affected by hirsutism associated with menstrual disorders. Testosterone was elevated in 9 cases, in whom high levels of Δ4-androstenedione and dihydrotestosterone were found, and normal in 3 cases, 2 of which presented high values of dihydrotestosterone and one high levels of Δ5-androstenediol. In some hirsute females therefore, other androgens, such as dihydrotestosterone and Δ5-androstenediol, play a role in the pathogenesis of hirsutism. Three antisera specific to aldosterone and elicited with different aldosterone protein conjugates (aldosterone-3-oxime rabbit serum albumin and aldosterone-3-oxime bovine γ-globulin) were studied by radioimmunological methods at various times subsequent to first-immunization. A considerable variability of the parameters important in radioimmunoassay was observed over the whole experimental period. Electrochemical stimulation of the hypothalamus of 23-day-old female rats induced precocious puberty as judged by occurrence of vaginal opening, the degree of uterine hypertrophy, changes in ovarian steroid content and incidence of first ovulation. Three types of responses were observed: (I) pubertal ovulation within 96 h; (II) pubertal ovulation within 120 h, and (HI) vaginal opening at 120 h not followed by ovulation. All treated animals showed a sustained increase in the LH/FSH ratio in both pituitary and plasma. Plasma estrogen was also increased 1 h after stimulation. A preovulatory rise in plasma estrogen and gonadotrophins was noted in type I and type II animals. These data lend further support to the suggestion that brain stimulation causes a release of gonadotrophins which induce ovarian steroidogenesis leading to an ovulatory gonadotrophin surge via a positive feedback effect.

The mouse epididymis was studied to localize histochemically a number of hydroxysteroid dehydrogenases in the various zones. The epithelium of the posterior half of the initial segment (head) and the anterior half of the middle segment (body) shows a strong reaction for △5-3β-, 3α,5α-, 3α,5β-, 11β-, 16α-, 170β- 20α- and 20β-hydroxysteroid dehydrogenases. This activity attenuates posteriorly. Only the 11β-hydroxysteroid dehydrogenase is present throughout the length of the epididymis. The endometrial blood flow was measured in rats by injections of 85Kr saline into aorta and measurements of the clearance of the radioactivity by a Geiger-Müller probe situated in the uterine lumen. Estrus and diestrus were determined by vaginal smears. The endometrial blood flow was found to be 0.88 ± 0.08 (mean ± SEM) ml/g tissue/min in estrus and 1.60 ± 0.15 mg/g/min in diestrus. 摘要:
To investigate the sites of the negative feedback of corticoids in the regulation of ACTH secretion, the effect of 10 µg of dexamethasone on adrenocortical responses to ether stress was studied in intact and fornix-sectioned rats. Dexamethasone pretreatment in both groups depressed significantly both the basal and stress plasma corticosterone levels. However, in rats with fornix section the amount of depression of the adrenocortical response was much smaller than in intact animals, when compared to the non-treated group. These data would indicate that extrahypothalamic regions play a role in the action of glucocorticoids in the feedback control of pituitary-adrenal function and that the hippocampus participates in this mechanism
