Whilst plasma renin values are elevated in a minority of patients with essential hypertension, there is no indication that high renin hypertension is a distinct entity and even where renin levels are elevated, they are usually less than those observed in renovascular hypertension. It is still possible for the renin-angiotensin system to be important, if blood pressure is maintained by renin activity within the arterial or arteriolar walls. Accordingly, renin-like activity of rat aortic wall homogenate and plasma renin concentrations were measured simultaneously. Both plasma and aortic renin (measured with an incubation pH of 6.5) rose in salt-depleted and fell in salt-loaded rats. Both were elevated in Goldblatt 2-kidney hypertension. However, aortic renin fell gradually over 24 h after bilateral nephrectomy, whilst plasma renin had reached its nadir by 1 h. To assess the importance of arterial as opposed to plasma renin the renin-angiotensin system was blocked with the converting enzyme inhibitor Sq20881. A blood pressure fall attributable to renin-angiotensin blockade could be demonstrated for at least 6 h after bilateral nephrectomy. This supports the hypothesis that renin in the arterial and arteriolar wall, rather than plasma renin, is responsible for blood pressure maintenance. Nevertheless, in steady state conditions such as essential hypertension, plasma and arterial renin are closely related.

The assumption that abnormalities exist in both the pituitary and gonads was tested in 3 familial cases of Kallmann’s syndrome, 2 brothers, aged 16 and 17 years, and their cousin, aged 26 years. Serial serum and urinary LH and FSH titers were measured after subcutaneous administration of 100 µg LRH and in the 2 brothers after a second test using 400 µg LRH. Serum testosterone and estradiol were estimated before and 60 and 90 min after LRH and also before and on the 3rd and 4th day after daily administration of 5,000 IU of hCG for 3 days. Prolactin was also measured in blood samples of all LRH tests and found normal in 2 cases and increased in 1. Tomograms of the pituitary fossa were normal in this patient. In female Long-Evans rats, development of prolactin cell adenomas is preceded by hyperplasia of prolactin cells, suggesting that factors affecting the endocrine function of prolactin cells might have an important role in the genesis of prolactin-producing pituitary tumors.

The effect of pentylenetetrazol on adrenal cortex maturation was investigated. Two different groups of weanling Sprague-Dawley female rats were used. At the ages of 23 and 29 days, the animals were treated with pentylenetetrazol (10 mg/kg) for 3 days. Results indicated that pentylenetetrazol decreased plasma corticosterone levels of both age groups. This treatment abolished the corticosterone diurnal variation of the 23-day-old rats, but not that of the 29-day-old rats. An increase in uterine weight was also observed in the pentylenetetrazol-treated animals. Results from this study suggest that CNS stimulation with pentylenetetrazol can alter the adrenal cortex maturation.

The purpose of the present investigation was to study the mode of action of a crude aqueous pineal extract (CAPE) on corticosterone (B) production from ACTH-mediated isolated adrenal cortex cells. Corticosterone production from a heterogenous adrenal cortex cell population, isolated from 8 male Sprague-Dawley rats, was measured fluorimetrically. CAPE (25 µl) was tested in this system using ACTH (0–5,000 pg/ml) and dibutyryl-c-AMP (0–100 nA//ml) as stimuli for a period of 1 h. In a separate experiment, CAPE (25 µl) was administered to ACTH (50 pg/ml) stimulated adrenal cortex cells for 15, 30, 60, and 120 min incubation periods. CAPE significantly decreased B produced by adrenal cortex cells at all doses of ACTH administered. CAPE also decreased the B produced by adrenal cortex cells when dibutyryl-c-AMP was used as a stimulus. The inhibitory effect of CAPE was manifest at some point in time between 30 and 60 min. It was significant at 60 min and highly significant at 120 min. It is evident from these data that CAPE and ACTH are not competing for the same receptor

摘要:
Serial sections from 16-day mouse embryos from mothers injected with 3H-diethylstilbestrol were processed for autoradiography. The distribution, in the laryngeal region, of cells with nuclear concentrations of radioactivity was analyzed. Estrogen receptor cells were found in the mesenchyme of the vocal cords and lateral to the laryngeal aditus. The embryological and oncological implications of these findings are discussed.
