A perianal bowenoid papulosis was examined for the presence of HPV types 2, 6, 11, 13, 16, 18, 32 and 33 by DNAin situhybridization. Positive signals were seen for HPV types 6/11, 13, 16 and 33. HPV type 13 is strongly related to oral focal epithelial hyperplasia and has not been reported outside the oral cavity before.

In an immunohistochemical study of 59 routinely processed tissue specimens from 48 adult testes with isolated carcinoma‐in‐situ (CIS) changes and of 66 specimens from adult testes without neoplasia, placental‐like alkaline phosphatase (PIAP) was shown to be a reliable marker of CIS cells preceding the development of a testicular tumour. Thus, a positive reaction was encountered in all 36 biopsies treated with formaldehyde, or Bouin's or Stieve's fluid. However, only 11 of 23 specimens fixed with Cleland's fluid were immunoreactive for PIAP. None of the non‐malignant components of seminiferous tubules, including the large abnormal spermatogonia, reacted with the antibody against PIAP. Besides the antibody against PIAP, monoclonal antibodies M2A and 43–9F were tested on CIS specimens fixed with the above‐mentioned fixatives. In the 17 specimens fixed with Stieve's or Bouin's fluid, a positive reaction was obtained in all sections with all three antibodies tested. However, for each antibody at least two specimens gave a weak staining reaction. When all three immunostainings were performed, in each case at least one of them gave a moderate or strong reaction, thus making CIS cells easily detectable. In the samples fixed with Cleland's fluid, a negative reaction was found in one to three specimens, depending on the antibody used. However, at least one of the three antibodies gave a positive reaction if all three immunostainings were applied. In only one of the formaldehyde‐fixed paraffin specimens did CIS cells react with the monoclonal antibody 43–9F, whereas M2A gave no positive reaction at all if this method of fixation was used. Thus, the sensitivity of the immunohistochemical staining procedure in the detection of CIS is dependent on the fixative used and increases when immunostainings with all three markers are performed

Contact hypersensitivity is characterized by an early and specific diapedesis of mononuclear cells into the site of antigenic challenge. In order to study the functional and ultrastructural properties of the endothelium involved in the recruitment of leukocytes, Sprague‐Dawley rats were skin sensitized to DNFB; and this was followed by challenge of the oral mucosa.In vitrobinding of peripheral blood mononuclear cells to high endothelial venules in lymph nodes was highly specific but no affinity of peripheral blood mononuclear cells to the vessels was observed in normal oral mucosa or in early contact hypersensitivity. However, 10 days after repeated DNFB challenge, occasional vessels bound overlaid peripheral blood mononuclear cells. Ultrasctructurally, we identified migration of mononuclear cells through small venules three h after challenge. The vessels involved, however, did not display morphological signs of activation reminiscent of high endothelial venules in lymph nodes. Using a whole bloodin vitromodel, we have investigated the effect ofEscherichia coli (E. coli), Streptococcus agalactiae(group B streptococci, GBS) and tumor necrosis factor alpha (TNF) on the generation of lactoferrin (LF), interleukin‐1 beta (IL‐1) and tissue thromboplastin (TPL) in healthy newborns at term and their mothers.E. coli(at a final concentration of 107/ml) significantly increased the release of LF in whole blood from newborns after 20 as well as 60 min stimulation, and in samples from their mothers after 60 min stimulation. A significant increase in the release of LF was observed in both newborns and their mothers after 20 and 60 min stimulation with TNF (at a final concentration of 1000 pg/ml). A combination of TNF/E. colior TNF/GBS never gave any significant additional stimulatory effect. After stimulation withE. colior GBS (both at a final concentration of 107/ml) for 60 min a significant increase in production of TNF and TPL was observed in newborns. In newborns a significant increase in production of TNF and TPL was observed also after 20 min stimulation withE. coli.TNF (at a final concentration of 1000 pg/ml) significantly increased the generation of TPL after 20 and 60 min stimulation in both groups. Salmonella entericaserovarbertawas isolated for the first time in Denmark in October 1984. This serovar has subsequently become endemic in the broiler industry. Plasmid profile analysis of 610 strains ofS. bertaisolated from broilers and broiler parent stock in the period October 1984 to July 1989 showed that plasmids were present in 85.7% of strains investigated, with a mean of 1.8 plasmids per strain. A significant increase in the mean number of plasmids per strain was seen during the observation period. Seventeen different plasmid sizes and 90 different plasmid profiles were identified. Plasmid profiling ofS. bertaappears to represent a promising tool for analysing the epidemiology ofS. bertainfections in broilers.

ManyEscherichia coliof serotypes commonly found in the normal intestine and in extraintestinal diseases, and having capsular antigens of the low molecular group called group II, will, in simple saline extracts, produce complexes between some or all of the lipopolysaccharide molecules and some of the polysaccharide K molecules. Non‐complex‐forming and complex‐forming strains with the same O and K can be found. The complexes are thermostable but are disrupted by some detergents. O‐K complex formation may lead to misinterpretation of immunoprecipitation results; one example is the counter current technique used for K determination ofE. coli.In this technique O antigen lipopolysaccharide may, when complexed to K polysaccharide, mimic a K antigen. Three different methods were used for detecting and isolating microorganisms with high radiation resistance from the microbial contamination on infusion sets prior to sterilization. By all three methods, microorganisms with a radiation resistance high enough to be a critical factor in a sterilization process (D‐6 value ≥ 30 kGy) were found with a frequency of approximately two colony forming units (cfu) per 100 product items, even though the product items in two of the series of analyses were irradiated with doses of 3–6 kGy. The frequency of occurrence of isolates with D‐6 values>30 kGy was 0.45 per 1000 cfu of the total aerobic count. Eight different isolates of microorganisms had D‐6 values>40 kGy when irradiated in dried laboratory preparation. Semiquantitative cultures were compared with blood cultures during one year in order to see if the routine use of a semiquantitative catheter culture method (SQC) in unselected patients can detect or predict infection associated with central venous catheters. Catheter infection, i.e. ≥ 15 colony forming units (cfu) per plate, occurred in 137 of 542 catheter tips (25%), mainly with coagulase‐negative staphylococci. Catheter‐associated bacteremia occurred in 17 of 93 cases (18%) where blood cultures had been drawn. In 15 of these, the catheter tip grew ≥ 15 cfu. The predictive value for bacteremia of a positive SQC was only 21%. SQC is not a suitable method for the detection of catheter‐associated bacteremia, but may be an indicator of the standard of cental venous cathete

Eight antibodies (UCHL1 (CD45RO), MT1 (CD43), MT2 (CD45R), 4KB5 (CD45R), MB1 (CD45R), MB2, L26 (CD20) and LN1 (CDw75)) have been examined for reactivity with routine specimens of normal and hyperplastic lymphoid organs (n = 6), non‐Hodgkin's lymphomas (n = 62), Hodgkin's disease (n = 27) and non‐lymphoid malignancies (n = 9). In normal and hyperplastic lymphoid organs, UCHL1 and MT1 stained predominantly T cells; 4KB5, MB1, MB2, L26 and LN1 stained predominantly B cells; and MT2 reacted with a subset of B and T cells. The lineage of the neoplastic cells was correctly identified in 24 of 28 (86%) peripheral T‐cell lymphomas; and in 31 of 35 (88%) B‐cell malignancies. In two cases of lymphocyte‐predominant Hodgkin's disease, the Hodgkin's and Reed‐Sternberg (H&RS) cells were 4KB5+, L26+ and/or LN1+. The H&Rs cells in nodular sclerosis and mixed cellularity Hodgkin's disease were positive with 4KB5 in 17 of 25 cases. Antibodies UCHL1, MT1, MB1, MB2, L26 and LN1 also labelled some H&RS cells, but in a much smaller proportion of the cases. In three of nine non‐lymphoid neoplasms, UCHL1 and MB2 showed a staining of the neoplastic cells, but the staining was cytoplasmic rather than membrane‐associated. The remaining antibodies were unreactive with the non‐lymphoid malignancies. It is concluded that many non‐Hodgkin's lymphomas can be typed in routine specimens, and that antibodies UCHL1, MT1, L26 and LN1 are especially useful in this respect. The vasculature of 25 lymph nodes of patients with human immunodeficiency virus‐related lymphadenopathy was investigated morphometrically. The number of small vessels, the morphological features of high endothelial venules and the migratory index of the lymphocytes passing through the high endothelial venules, as well as the stage‐dependent change of each parameter, were analysed. Twenty reactive lymph nodes served as controls. The total number of vessels in the HIV‐infected lymph nodes was relatively stable. However, the small vessels with flat endothelium increased in number, while the number of high endothelial venules decreased as the disorder progressed, and the decrease in the lymphocyte migration seemed to precede the change in the morphology of high endothelial venules.
