Wall analyses on recently isolated strains of Willoughby's ‘spore‐dome’ actino‐mycetes grown in submerged culture would suggest that the peptidoglycan containedll‐diaminopimelic acid (ll‐DAP) ormeso‐DAP or even mixtures of the two isomers. This chemotaxonomic paradox was explained when it was shown that the vegetative mycelium had walls containingll‐DAP and glycine (wall chemotype I) and that spores contained onlymeso‐DAP as the diagnostic amino acid (wall chemotype III). A method previously used for the analysis of organic acids in silage has been applied to the detection and quantification of acidic fermentation products (C1to C6volatile fatty acids, lactic and succinic acid) of rumen bacteria and anaerobic fungi grown in pure culture. The acids were converted to tertiary butyldimethylsilyl derivatives prior to separation on a 30 m DB1 capillary gas chromatographic column. The quantitative recoveries of formic and succinic acids were found to be comparable to the recoveries of other acids reported in the original study. The quantitative recovery of lactic acid was found to be dependent on storage of the samples at ambient temperature for at least 24 h following derivatization. Data from analyses of the fermentation of cassava slurry were used to derive kinetic constants for the rate of hydrolysis of bound cyanide, formation of free cyanide, pH change and fermentation. The pseudo‐kinetic constant for the decay of bound cyanide was 1·6 times 10‐2/h and 2·5/h for free cyanide. Under the experimental conditions, the concentration of bound cyanide at any time during fermentation was 22·65e‐0·016tμg/g, wheretis time in h. The fermentation rate constant was determined as 2·2871 times 1019. Three close‐fitting lid fermenters were incubated at 27 ± 2°C for 120 h. ‘A’ served as a control and contained 100 ml raw goats' milk; ‘B’ contained 50 ml 48‐h‐old nono as starter culture plus 50 ml pasteurized milk; and ‘C’ contained 100 ml pasteurized milk only. Aureobasidium pullulansis a new source of enzyme beta‐galactosidase with optimum activity at pH 6·8 and a temperature of 45°C. Lactose induced enzyme synthesis, but low constitutive levels of enzyme were seen with other sugars. Addition of Mn2+and Ca2+increased enzyme production and activity. 摘要:
The efficacy of cinnamon oil, individually and in binary mixture with sodium chloride (NaCl), in preserving stored corn from fungal invasion and subsequent deterioration was investigated. It was observed thatAspergillus flavusandA. glaucus, the predominant storage fungi, invade corn during storage with the concomitant production of aflatoxin and the resultant loss of germinability in grains. The treatment with cinnamon oil at the non‐phytotoxic level of 5 μl/g corn in combination with 10 μl/g NaCl solution (5%) inhibits synergistically the fungal infection, growth and aflatoxin production together with the deterioration of grains (loss of germinability). However, the test oil, when applied alone, was found to be effective only at the phytotoxic level of 20 μl/g
