Six monoclonal antibodies (MAbs) againstMoraxella bovisGF 9 were used to quantify haemagglutinating adhesins of 16 strains of this organism. The amount of each MAb necessary to inhibit one haemagglutinating unit of each strain varied between 4 and 0·007 times that required by strain GF 9. Total direct and direct viable counts of fresh and injured cultures ofEscherichia coliwere determined by image analysis in preparations stained with acridine orange, ethidium bromide and 4′,6‐diamidino‐2‐phenyl indole (DAPI). Cells stained with DAPI were not detected by image analysis. Fresh cultures stained with acridine orange or ethidium bromide gave comparable counts. InjuredE. colistained with ethidium bromide gave higher counts that with acridine orange. The use of flow cytometry to detectCryptosporidiumoocysts in water was investigated using a Skatron Argos 100–5 instrument. Raw and treated drinking water samples seeded with oocysts and treated sewage samples known to contain oocysts were analysed by flow cytometry and by fluorescent microscopy. Oocysts could be rapidly detected in the treated sewage in raw and treated drinking water when the latter were seeded with levels of 1000 oocysts/1 or higher. DNA‐DNA hydridization has been used to detect homologies between the TOL plasmid‐encoded gene clusterxylXYZLand the chromosomally‐locatedbenABCDgenes inPseudomonas putida;both sets of genes code for isofunctional enzymes converting benzoate and toluates into catechol and its derivatives. Growth ofBacillus cereusNCIB 8579 was studied on four varieties of rice with and without tapé fermentation. Fermented and unfermented rice supported growth ofB. cereusto 107–109cfu/g. Biodegradation of softwood (Pinus radiata) by the ascomyceteChrysonilia sitophilawas dependent on the nitrogen and glucose concentrations in the culture medium. Optimization studies of the delignification process, in which the nitrogen (0–50mmol/l NH+4) and glucose (0–2%) concentrations were varied, showed a maximal value of 17·8% for sawdust degradation in cultures containing 10 mmol/l NH4+and 1·0% glucose. Solubility of the decayed sawdust in 1% NaOH at maximal delignification conditions showed a threefold increase and changes in the thermogravimetric pattern were also observed.
