摘要:
The isoamylase activity ofPseudomonas amyloderamosaWU 5315 was stable over the pH range from 5.5 to 6.25 while only about 30% of the activity remained at pH 6.5. Low isoamylase activity (418 U ml‐1) was produced by the cells grown at high pH. Activity reached almost 3000 U ml‐1when pH was kept below 6.0 during the fermentation. With 1% glucose plus 2% maltose instead of 3% maltose as carbon source, however, no pH control was required and the isoamylase activity ofPs. amyloderamosaWU 5315 increased to 3400 U m

摘要:
A fast and simple enzymic method has been adapted to measure histidine decarboxylase activity in bacteria isolated from fish. It was possible to quantify the presence of histamine at levels higher than 1.03 times 10‐2μmol (3 ppm) per ml of culture medium. There was a good correlation (γ=0.99) between the histamine content and the increase of absorbance in the concentration range of 3 and 30 ppm. The method can be used for quantitation of the amount of histamine produced by a selected strain, and has the advantages of being low in cost, reliable, easy to use and fast to perform.Plesiomonas shigelloides, a bacterium frequently isolated from fish and aquatic environments, has been identified as a new histamine‐former in

Groundwater samples from two sites in Alabama, USA were plaque assayed for F‐specific RNA (FRNA) coliphages usingSalmonella typhimuriumWG49 as the host bacterium. While numerous plaques were detected with WG49 (a strain possessingEscherichia coliF pili), plaques were also observed with an F‐control strain ofSalm. typhimurium.Five isolates were plaque purified and examined by electron microscopy. All of the isolate particles were observed to be tailed, with five distinct particle types being differentiated. None of the isolate particles were consistent in morphology with the cubic FRNA coliphages. Host range evaluation supported classification of the isolates as salmonella phages. Somatic salmonella phages have previously been found to interfere with the assay of surface waters for FRNA coliphages. Their detection here demonstrates that such interference is also encountered in the assay of groundwat

Lysine mannitol glycerol agar (LMG) was compared to xylose lysine deoxycholate agar (XLD), bismuth sulphite agar (BS), andSalmonella‐Shigellaagar (SS) for the ability to detectSalmonellaspp. in clinical specimens, primarily faeces samples. During an 8‐month period, 15 salmonellae were isolated from 940 faeces on LMG, while 14 strains were obtained on XLD, 11 on SS and only 3 strains on BS.Salmonellatyphi was recovered from two blood cultures in 24 h on LMG, compared to 48 h on BS. LMG was augmented by addition of a sulphacetamide/mandelic acid (sulphamandelate) selective supplement (LMGS). During a 20‐month period, 43 salmonellae were isolated from 2622 faeces on LMG and LMGS. The selectivity of LMGS was superior to that of LMG with no decrease in sensitivity of detection; all salmonellae isolated on LMG were isolated on LMGS. Bioluminescence from developing L‐form colonies of the plant pathogen,Pseudomonas syringaepv.phaseolicola, was monitored using the enhanced light‐detecting capabilities of a charge‐coupled device. During L‐form induction, the bacteria entered a prolonged period during which the level of light output and hence metabolic activity, was very low. A relatively small number of highly bioluminescent L‐form colonies were then observed to develop against a background of non‐bioluminescent bacteria. The lipopolysaccharide (LPS) of 54 Australian isolates, nine isolates acquired or isolated overseas, and two reference strains ofSalmonella enteritidiswas studied to assess its relation to pathogenicity. LPS was extracted by proteinase K digestion of whole cells, and analysed by polyacrylamide gel electrophoresis. All isolates possessed an LPS structure identical to that of a reference strain ofSalm. enteritidisphage type 4. Representative strains of the clinically prevalent phage types 4, 14 and 26, which express long chain LPS, were assessed for their pathogenicity in mice.Salmonella enteritidisphage type 4 produced a lethal infection in BALB/c mice, but not in C3H/HeJ or Quackenbush (outbred) strains. Phage types 14 and 26 did not produce an obvious infection in any mice, suggesting Australian strains of phage type 4 are more virulent than phage types 14 and 26.
