摘要:
Adsorption ofPseudomonas cepaciaonto the surface of glass containers enables cells to survive in the presence of user strength chlorhexidine (0.05% w/v). The organisms were found to be firmly attached to the glass and colonised glass slides could be used to infect uncontaminated chlorhexidine solutions. The efficacy of chlorhexidine againstPs. cepaciadecreased with decreasing pH. Storing infected solutions at low temperatures reduced the toxicity of chlorhexidine and promoted longer survival.

摘要:
Silver‐stained patterns of lipopolysaccharide on sodium dodecyl sulphate polyacrylamide gels and microbial antigenic profiles on nitrocellulose sheets produced by electroblotting and immunodetection were analysed with a laser scanning densitometer. Because the patterns fade rapidly it is usual to photograph them immediately to obtain optimum resolution. It has been shown that because of its high resolution the laser densitometer can be used to scan 35 mm photographic negatives without appreciable loss of detail. A wedge‐shaped, horizontal agarose gel gave better electrophoretic resolution of both large and small plasmids than conventional linear gels. The wedge‐gel increased mobility and separation of the larger plasmids whilst retarding the mobility of the smaller plasmids and tightening the bands. The linearity of the relationship between log10molecular size and log10relative mobility, for a range of plasmids from 2.1 to 221 kb, was increased. A comparison was made between a commercially available enzyme immunoassay (ELISA) and various cultural procedures for detecting salmonellas in minced meat contaminated with a standard inoculum. To detect salmonellas by the ELISA technique, it was necessary to modify the recommended baseline for spectrophotometric measurements to avoid false‐positive results.
