A new medium has been developed for the isolation ofListeria monocytogenesfrom clinical specimens with a mixed flora. Almost complete inhibition of unwanted organisms was achieved and recognition of colonies ofListeriaspp. was usually possible after 24 h using the aesculin‐ferric ammonium citrate indicator system. The conditions for optimum protoplast formation fromPenicillium nalgiovensewere determined. Best results were achieved using Novozyme 234 in combination with β‐glucuronidase. Buffered solutions of chlorine‐releasing agents, sodium hypochlorite (NaOCl), sodium dichloroisocyanurate (NaDCC), and chloramine‐T, showed similar activity against vegetative cells ofBacillus subtilisbut there was considerable variation in activity against spores, NaOCl showing higher activity than NaDCC, which in turn was more active than chloramine‐T. The effect of coat and cortex extraction on sporicidal activity was determined. It was concluded that although spore coats play a role, they do not totally account for chlorine resistance and that the cortex is also involved, probably through its function in maintaining a low water level in the enclosed core. Sediment and the overlying water samples were collected from 67 fresh and 38 salt water sites in the UK. Only 13% (five) of the samples taken from salt water sites were negative for magnetotactic bacteria compared with 47·7% (32) of the fresh water ones. Two hundred and twelve strains representing 11 species of fluorescent pseudomonads along with an additional 198 strains isolated from the leaf and root surfaces of plants were tested for rutin glycosidase, on a mineral base medium supplemented with rutin and glucose. AH of the arginine dihydrolase negative phytopathogens (182 strains) with the exception of 12 of 12 strains ofPseudomonas syringaepv.glycinaand one of 28 strains ofP. syringaepv.phaseoticolashowed rutin glycosidase activity whereas few of the arginine dihydrolase positive strains (22 of 228) were positive.
