A study was undertaken to determine the effectiveness of trisodium phosphate (TSP) in wash water in the inactivation ofSalmonella montevideoon the surface and in core tissue of unwashed, mature‐green tomatoes. Complete inactivation on the tomato surface was achieved by dipping tomatoes in a 15% TSP solution for 15s. Significant (P<0.05) reductions were obtained by dipping tomatoes in a 1% solution for 15s. Populations were significantly reduced in core tissue of tomatoes dipped in 4–15% TSP. However, even at 15%, only about 2 log10reduction was achieved. Upon ripening, the hue and chroma of tomatoes, indices of colour and brightness, respectively, were unaffected by treatment of TSP. S. SANDHYA. 1996.Nocardioidessp., isolated for organic sulphur degradation, harbours catabolic plasmid pSB1 of molecular weight 34.2 kb. A correlation was noticed between the plasmid content and degradation of the organic sulphur compound dibenzothiophene (DBT). The maximum oxidation of DBT was only after 64 h, during which the content of plasmid DNA was also optimum.

Outbreaks of serious mortality among cultured kuruma prawns (Penaeus japonicus) with white spotted syndrome in the carapace occurred in the summer of 1993 in I‐Lan, Taiwan. A swarming bacterium, strain Swy, was isolated from the hepatopancreas of the moribund prawns using tryptic soy agar supplemented with 1% NaCl and/or thiosulphate citrate bile salt sucrose agar. This strain was characterized and identified asVibrio alginolyticuson the basis of a number of biochemical tests. M. PAREKH, H.L. DRAKE AND S.L. DANIEL 1996.Desulfovibrio desulfuricansATCC 27774 was screened for reactivity against aromatic compounds during lactate‐dependent, nitrate‐dissimilating growth. Only aromatic aldehydes (benzaldehyde, 2‐hydroxybenzaldehyde, 3‐hydroxybenzaldehyde, 4‐hydroxybenzaldehyde, vanillin,iso‐vanillin ando‐vanillin) were reactive and, with the exception of 2‐hydroxybenzaldehyde, were stimulatory to lactate‐dependent growth. Aromatic aldehydes were transformed to their corresponding benzoate and benzyl alcohol derivatives, with the ratio of benzoate‐to‐benzyl alcohol derivatives being dependent upon lactate availability. In presence of lactate, aromatic aldehydes were primarily reduced to their corresponding benzyl alcohol derivatives; in the absence of lactate, aromatic aldehydes were mainly oxidized to their corresponding benzoate derivatives. In the absence of nitrate, 3‐hydroxybenzaldehyde was neither reduced nor oxidized. These results indicate thatD. desulfuricansis competent in the bidirectional transformation of aromatic aldehydes under nitrate‐dissimilating conditions and that the direction of transformation (i.e. reduction or oxidation) is regulated

The effect of growth temperature and growth rate on the susceptibility to heat and pH stress were investigated inListeria monocytogenesgrown in continuous culture where these two growth variables could be varied independently of each other, and in batch culture. After growth at 30°C or 10°C at constant growth rate, or at 30°C at different growth rates, cells did not differ in their resistance to heat at 55°C. Cells grown at 30°C were more resistant to acid stress at pH 2.5 than cells grown at the same growth rates at 10°C. Cells grown at low growth rate at 30°C gave greater resistance to acid stress than those grown at high growth rate. Growth temperature and growth rate had independent effects on the susceptibility ofL. monocytogenesto acid stress conditions. This may have implications for the survival ofL. monocytogenesin acidic

The genetic similarity of nineteen isolates ofCandida albicansfrom four patients were compared by restriction fragment length polymorphism (RFLP) usingEcoRI orHinfI, which both detected five types, and by random amplification of polymorphic DNA (RAPD), which detected three types. Phenotypically unusual isolates also produced distinct patterns with both typing systems demonstrating the carriage of two groups ofC. albicansas well as the presence of more than one type in some subjects. Methods of DNA preparation were compared for the production of reproducible patterns; including using the supernatant fluid of boiled intact or spheroplasted cells for RAPD, and DNA precipitated from chloroform extracted cell lysate for RFLP and RAPD. Consistent patterns were produced from the DNA precipitate by RAPD and after an additional precipitation by RFLP, thus removing the necessity for lengthy extraction procedures or the use of toxic chemicals for purification.

M.W. SCHOEMAN AND D.J. DICKINSON. 1996. Ferulic acid acted as an adequate sole carbon source forAureobasidium pullulansisolate 105 in liquid culture. A corresponding depletion of ferulate, indicated by depression of the culture filtrate's absorbance at 280 nm, was observed over 4 weeks of incubation. Growth of isolate 105 and six other isolates ofA. pullulanswas also observed on three other aromatic compounds, catechol, protocatechuic acid and vanillic acid. However, none of the isolates grew on dioxane lignin. These results may contribute to a better understanding of the way in whichA. pullulanscolonizes painted wooden surfaces.
