摘要:

Spectral changes resulting from specific interaction between sterols and the polyene antibiotic filipin have been exploited in a simple sterol assay that is not subject to interference. This can be employed to measure the sterol content of brewing yeast prior to use thus permitting improved control of the fermentation process. It may also be applicable to quantification of fungal biomass.

Cells ofEscherichia coliandStaphylococcus aureuswere immobilized by centrifugation onto glass coverslips treated with 3‐aminopropyltriethoxysilane (APS‐centrifugation), labelled with fluorescein isothiocyanate (FITC), fixed with acetone and counterstained with propidium iodine (PI). Six strains ofEnterococcus faecaliswere tested for antimicrobial activity againstListeria monocytogenes.Using the agar spot test,E. faecalisNRIC 1143 and 1144 showed strong activity against all of five strains ofL. monocytogenestested. Antimicrobial activities of these strains were inhibited by some proteolytic enzymes.

Two methods for the direct extraction of DNA from soil were investigated using soil inoculated withStreptomyces violaceolatusISP5438 harbouring the multicopy plasmid pIJ673. Detection limits for plasmid DNA were determined by Southern blot technique. An SDS/heat lysis method gave approximately two orders of magnitude less sensitivity than lysis and extraction by bead‐beating soil inoculated with spores. The acridine orange direct count (AODC) technique was used to count bacterial numbers in beef mince. The ability of Alcalase 2·5L to degrade beef mince was compared with the previously used Alcalase 0·6L. Both methods were evaluated against the standard plate count. The small‐subunit rRNA sequences of 51 species ofBacilluswere determined by reverse transcription to elucidate the phylogenetic structure of the genus. Comparative analysis of the sequence data revealed five phylogenetically distinct clusters. Group 1 (Bacillus sensu stricto) includedB. subtilisthe type species of the genus and 27 other species. Group 2 consisted ofB. sphaericusand five other bacilli andSporosarcina ureaeclustered within the confines of this group. Group 3 consisted of a phylogenetically coherent group of 10 species whereas groups 4 and 5 comprised two and three species respectively. It is evident that the genusBacillusin genetically extremely heterogeneous and requires extensive taxonomic revision. Twelve strains of thermophilic campylobacters isolated in Valencia from faeces of infants and young children with diarrhoea, and from seawater, were biotyped and examined by analysis of chromosomal DNAHaeIII ribopatterns. The strains were identified asCampylobacter jejuniandC. coli, and all had different ribopatterns except for three identical faecal isolates ofC. jejunibiotype I. No correlation was found between Lior biotype andHaeIII ribopattern but several bands within ribopatterns were identified as possible species markers for use in diagnostic and epidemiological studies.

Three direct plating methods to enumerateEscherichia colifrom food in 24 h are described. Unlike the majority of enterics, 96% ofE. coliare able to cleaveβ‐glucuronic acid. This reaction can be observed by incorporating (1) 4‐methylumbelliferyl‐β‐D‐glucuronate, (2)para‐nitrophenyl‐β‐D‐glucuronate or (3) 5‐bromo‐4‐chloro‐3‐indolyl‐β‐D‐glucuronate (BCIG) into a peptone tergitol agar base.Escherichia coliproduce fluorescent, yellow or deep blue colonies from these three compounds respectively. BCIG agar proved the easiest to read and produced the least number of false positives and false negatives.
