The literature on bioassay methods for mycotoxin detection has been reviewed. An outline of the range of bioassay methods is given and the role of cytotoxicity tests in particular has been emphasized.

After incubation with 20 different mycotoxin standards and extracts from fungi and feed stuffs, fluorescent flow cytometry was used to measure viability of NS.1 cells and compared to microscopic assessment of cytotoxicity on stained HEp‐11 monolayers. Both methods gave essentially the same results but the cytometric analysis offered a more quantitative approach and was particularly appropriate in the screening of extracts containing fats. A monoclonal antibody (mAb) has been produced to aflatoxin B, (AF B,) after successful immunization of mice and fusion of sensitized spleen cells with myeloma cancer cells. The mice were immunized with AF B, +xime‐protein conjugate. Positive mAbs were screened using an indirect ELISA specific for AF B1. The selected mAb was then developed in direct competitive ELISA and immunoaffinity column chromatography methods for aflatoxin detection in foods and feeds. Both assays are rapid, sensitive, specific and require only the minimum of sample preparation. Both immunological assays have now been commercialized and are produced in convenient ready‐made kit form

An improved procedure for the preparation of CVP medium is described on which pectolytic erwinia rapidly develop deep, localised pits. The medium can be prepared quickly and easily and forms a smooth, firm gel suitable for streaking.

Acid and alkaline phosphatase activities in culture liquid and mycelial extract during autolysis were studied in seven fungi of the general Ascomycotina, Basidiomycotina and Zygomycotina. High activities of extracellular and mycelial extract acid phosphatase and lower activities of alkaline phosphatase were found in Ascomycotina, and acid phosphatase was present in Basidiomycotina. In Zygomycotina only mycelial extract alkaline phosphatase activity was detected. A correlation between degree of autolysis, pH and acid phosphatase activity was demonstrated.

Electroporation was used to facilitate transformation of Listeria species with plasmid DNA. Optimal conditions for transformation ofL. monocytogeneswere a field strength of 8·5 kV/cm, 200 Ohms resistance, 25 μF capacitor with a time constant of 5 ms. Industrial fish peptone was an excellent substrate for biomass production in solid and submerged fermentations. The maximal growth rates of several microorganisms were two to three times higher than those grown on beef (bacto)peptones and the final biomass concentrations were almost twice as great as those grown on beef peptones. Groundnut cake (‘kulikuli’) purchased from four major markets in Ibadan, Oyo State, Nigeria were analysed for aflatoxin B, and associated rnycoflora. In all but two of the samples aflatoxin B, concentrations were between 20 pg/kg and 455 pg/ kg. Mould counts were low (1·0 × 1024·40 × 102colonies/g). Eight mould species were isolated. Of these Aspergillus niger, Paecilomyces oarioti,Aspergillus flavrs and Fusarium moniliformedominated in decreasing sequential order.
