A colony hybridization method for visual detection of heat‐labile enterotoxin (LT)‐producingEscherichia colihas been developed. The interchangeability of probe DNA in the system offers the advantage of a more definitive indicator and detection procedure over current plating procedures. A hydrophobic interaction chromatography (HIC) procedure was used to compare the relative surface hydrophobicity of threeProvidencia stuartiistrains and wild type and envelope mutants ofEscherichia coliandPseudomonas aeruginosa. Providencia stuartiistrain Pv2 adsorbed to a greater extent to octyl‐ and phenyl‐Sepharose than did Pv67. The HIC technique showed a significant difference in surface hydrophobicity between wild type and envelope mutant strains, the latter being considerably more hydrophobic. Pre‐treatment of cell suspensions with chlorhexidine produced further changes in the hydrophobic nature of all the strains. A specific, high affinity monoclonal antibody to T‐2 toxin ofFusariumhas been produced. The monoclonal antibody was conjugated to horse‐radish peroxidase and employed to develop a direct competitive enzyme‐linked immunosorbent assay (ELISA) for the toxin. Cellulosic substrates were tested for their ability to bind the cellulases ofThermomonospora curvata. Protein‐extracted lucerne fibres had the highest adsorptive capacity while native cotton fibres had the least. Analysis of binding as a Langmuir adsorption isotherm yielded maximal binding capacities of 0.012 filter paper units and 0.51 endoglucanase units/mg lucerne fibre at enzyme saturation.
