A rapid method for estimating the number of cultivable bacteria in dental plaque samples was developed in which five fluorogenic substrates (4‐methylumbelliferyl (4‐MU)‐α‐glucoside, 4‐MU‐β‐glucoside, glycyl‐prolyl‐7‐amido‐4‐methyl‐coumarin (‐AMC), tyrosyl‐AMC and prolyl‐AMC) in 150 μl were incubated with 50 μl of dental plaque suspension. The increase in fluorescence measured at emission and excitation wavelengths of 380 and 460 nm correlated with the colony count per plaque sample. The development of standard suspension test methods for disinfectants and antiseptics for adoption in Europe is described. Evaluation of a range of disinfectant agents and products currently used in the UK under conditions as proposed for these tests indicates that the majority of products diluted in water of standard hardness showed satisfactory activity producing a 4·5–5 log reduction in viable count within 5 min againstStaphylococcus aureus, Pseudomonas aeruginosa, Enterococcus faecium, Proteus mirabilis and Candida albicansin the absence and presence of 1% albumin. The AMBIS is a system which can determine the relationships between microbial strains by comparing the profiles produced after their radiolabelled intracellular proteins are subjected to SDS PAGE. This system was used to compare the profiles of strains representing the five biovars ofPseudomonas fluorescens, a species implicated in food spoilage. The three strains of biovar I, three strains of biovar III and two strains of biovar IV segregated into three distinct clusters with correlation coefficients (cc) of 0·85, 0·85 and 0·87 respectively. Although two of the biovar II strains studied clustered together (cc = 0·74) one of the remaining biovar II strains linked (cc = 0·83) with the cluster of biovar IV strains while the other was linked with biovar I and V strains (cc = 0·68). Biovar V strains (three in total) also failed to form a single cluster which was expected since this biovar is known to be heterogeneous.
