ABSTRACT:Serum from seven of 15 patients with non‐seminomatous testicular germ cell tumors impaired the transformation of lymphocytes from normal donors by phytohemagglutinin to less than 50% of that with addition to the cultures of normal AB serum. Similarly, serum from eight patients impaired the transformation by Staphylococcus aureus Cowan I. The impairment of the lymphocyte transformation did not correlate with the serum concentrations of alpha‐fetoprotein (S‐AFP) and human chorionic gonadotropin (S‐HCG). The patients with metastases and serum that impaired lymphocyte transformation to less than 50% of that With addition of serum from the controls and the other patients survived in the same way (0.50

ABSTRACT:Blood sera of 181 women with inflammatory conditions and with benign and malignant neoplasms of the ovary and of 36 healthy women (control group) were examined by determining the levels of defense proteins: C‐reactive protein (CRP), alpha1antitrypsin (A1AT), alpha2macroglobulin (A2M), components C3(beta1A‐globulin) and C4(beta1E‐globulin) of the complement, immunoglobulins M (IgM), A (IgA), and G (IgG), as well as the concentration of circulating immune complexes (IC). In women with ovarian pathology, as compared with the control group, a significant increase of all the defense proteins was found (p<0.05). The highest increase of the defense proteins, except in regard to A2M was observed in inflammation of the ovary. In malignant neoplasms of the ovary, the level of A2M was the highest and that of A1AT as high as in inflammatory conditions. There was also a high percentage of increased CRP values (57%). The concentration of the remaining proteins under study, although significantly higher than in the control group (p<0.05), was in this group the lowest in comparison with the groups of ovarian inflammation and benign neoplasms, especially with regard to component C3of the complement and to IgM and IgG (p<0.05). ABSTRACT:Binding sites for the Fc portion of immunoglobulin G (IgG) molecules (FcγBS) were identified on amnionic epithelial cells and chorionic trophoblast cells in human extraplacental membranes using direct immunofluorescence with labeled human IgG. The binding sites are similar to FcγBS on placental trophoblast in their specificity and high degree of relative binding affinity for IgG monomers. Neither immunoglobulin A (IgA) nor IgM bound to a significant degree to the membranes. Binding was completely inhibited with unlabeled IgG but not F(ab')2 fragments of IgG. Studies on isolated amnion cells demonstrated that the binding sites are expressed on the cell membrane. At the level of sensitivity of the immunohistologic methods used, aggregated IgG or antigen‐antibody complexes failed to bind amnionic epithelial cells or chorionic trophoblasts. ABSTRACT:The purpose of this study is to clarify the problem regarding individual specificity of maternal serum immunoglobulin G (IgG) and placental eluate IgG that block the unidirectional mixed lymphocyte reaction (MLR) in which maternal and cord blood lymphocytes (lys) were used as responder and stimulator, respectively. Both maternal serum IgG and placental eluate IgG were found to include blocking antibodies (BA) which showed similar blocking effect on MLR. But, in the case of serum IgG, individual specificity of BA on MLR was observed to be less great in MLR done using whole lys or T‐enriched lys of the cord blood as stimulator, whereas it became greatly significant when MLR was done using B‐enriched lys of the cord blood as stimulator. This was the case even in MLR done by using unrelated maternal lys as responder. On the other hand, in the case of placental eluate, it was found to be greater when compared to serum IgG even where MLR was done using not only B lys but also T lys as stimulator. ABSTRACT:The monoclonal antibodies (mAb) H315, H317, and OKT9 have been used in immunofluorescence to investigate the expression of fetal trophoblast membrane antigens by cells within human term amniochorionic membranes and the marginal area of term placental bed tissue. OKT9 reacted only with trophoblast of placental chorionic villi and did not react with any nonvillous cytotrophoblast population: this mAb is known to identify the cell surface receptor for transferrin. H315 identifies a trophoblast‐specific cell‐surface antigen and strongly stained both placental villous trophoblast and the cytotrophoblastic layer of amniochorion. This mAb also stained some extravillous cytotrophoblast in the term placental bed, notable interstitial cytotrophoblast within maternal decidua. ABSTRACT:We have tested peripheral mononuclear leukocytes (PML) from the cord blood of newborns, from sera of their mothers, and from sera of nonrelated nonpregnant adult women for sensitivity to suppressive exogenous prostaglandin E2(PGE2). Endogenous PG production was simultaneously inhibited by indomethacin 2.8 μM. The phytohemagglutinin‐stimulated (PHA‐stimulated) uptake of tritiated thymidine (3H‐TdR) by PML from the mothers and the nonpregnant women was suppressed by the exogenous PGE2at a concentration of 1.4 × 10−8M, 100 times less than the one required to suppress the PML from newborns (1.4 × 10−6M). In addition, 1.4 × 10−7M or less of PGE2reversed the suppression of neonatal PML to stimulation. The maternal PML were reversed into stimulation at 1.4 × 10−9of exogenous PGE2. The amount of endogenous PGE2synthesized by 1 × 106fresh, nonstimulated neonatal PML according to gas chromatography‐mass spectrometry assay was 5 ng (1.4 × 10−8M). The synthesis increased to 27 ng/106cells after 18 hours' incubation. These concentrations are similar to the ones of exogenous PGE2at which neonatal PML were slightly stimulated but the maternal cells were still suppressed.
