ABSTRACT:The necessity of T lymphocytes in local adoptive transfer of experimental allergic orchitis (EAO) is an established fact. The present study was undertaken to elucidate the participation of B lymphocytes in the local transfer system of EAO. The capacity of purified lymph node T and B cells to transfer EAO was compared. Donor strain 13 guinea pigs were sensitized with testicular antigen and complete Freund's adjuvant. We then injected 1 × 108cells just under the tunica albuginea of the testis of a syngeneic recipient. The recipients were killed 5 days after cell transfer. Nylon‐wool nonadherent T cells alone produced only minimal lesions, but could induce more severe mononuclear lesions with the addition of syngeneic, normal macrophages. In contrast, B‐cell enriched populations either alone or combined with a small number of macrophages, were able to transfer extensive lesions. In the combined transfer of immune T and immune B cells the severity of transferred lesions was dependent upon the number of B cells. ABSTRACT:We demonstrated anticomplementary activity on once‐washed human sperm, and in normal and vasectomized seminal plasmas. It was demonstrated to be a normal component of human semen. The origin of the activity is proposed to be the seminal plasma with sperm adsorption of activity. The properties of the seminal anticomplementary factor were characterized further, and the molecular size was shown to be less than 3500 daltons. Reduced anticomplementary activity was found to be associated significantly with abnormal semen profiles and infertility in males. The activity in seminal plasma was shown to have no effect on complement‐dependent sperm immobilizing antibodies in the serum of an infertile woman, implicating an effect on the post‐C3components of the complement pathway. ABSTRACT:The experimental immune oophoritis (EIO) model was used to study the functional relationship between the ovary and organs of the immune system. EIO was developed in Balb/c mice by twofold subcutaneous injection of saline ovarian extract in complete Freund's adjuvant. The ovary, thymus, and spleen were investigated histologically, histoautoradiographically, and electromicroscopically on days 7, 14, 21, and 28 after immunization. EIO was manifested by massive atresia of follicles and perivascular mononuclear cell infiltration. The appearance of the germinal center like structures in the thymus and increased T cell mitotic activity and B cell stimulation in the spleen pointed out that EIO is mediated both by T and B cells.

ABSTRACT:Cells from mice alloimmunized in the presence of placental extract were coinjected with cells from mice conventionally alloimmunized in the footpad of virgin female recipients. The contralateral footpad received a control twice the dose of cells from alloimmunized mice. Cells from the popliteal lymph nodes were harvested on day 3, and pulsed in vitro for measurement of proliferative capacity with3H thymidine. The response of lymph nodes was compared (homo‐ vs contralateral). The cells from mice alloimmunized with placental extract in conjunction with alloantigens displayed a marked suppressive capacity of the local graft versus host (GVH) potential of cells from mice conventionally alloimmunized. ABSTRACT:Antibodies were raised in rabbits to purified human term placental villous plasma membrane. These were cytotoxic to human peripheral blood lymphocytes and manifested cross‐reactivity to kidney and liver. After absorption with these tissues, reactivity was retained with placental villous plasma membrane. ABSTRACT:Placental extracts obtained from CBA (H‐2k) mice during syngeneic (CBA 9 × CBAcr)pregnancy were evaluated for their capacity to deviate the immune reaction of CBA mice toward A/J(H‐2a) immunizing spleen cells, as tested by alloantibody subclass formation and Sa 1 allograft accelerated rejection—or enhancement. The immunomodulatory activity appeared to be located in the soluble and in the insoluble extracts of placenta. The sodium deoxycholate (SI)O) solubilized proteinic material, fractionated on Concan‐avalin A [Con A] and injected to CBA mice simultaneously with A/J spleen cells caused a suppression of the hemagglutinating titer and C‐mediated cytotoxicity (IgG2). However, it also favoured antibody‐mediated mast cell degranulation (IgGl) and Sa 1 allograft enhancement. The presence and role of IgGl antibodies in both tumor enhancement and gestation has been described. It is concluded that placenta contains Con A‐binding glycoproteins which are located in the membranes and released in soluble form. ABSTRACT:Neutrophils isolated from cord blood of healthy newborns (33 blacks and 21 whites) were investigated by EDTA‐microagglutination for their expression of neutrophil specific antigens that have been associated with isoimmune neonatal or autoimmune neutropenia. Equal volumes of various neutrophil anti‐sera (2 μ) and cord neutrophils (3–5 × 106/ml) were mixed in tissue typing microplates under oil and were incubated at room temperature for 6–8 hr, following which the degree of agglutination was noted. Our data revealed that all the currently recognized neutrophil antigens are readily demonstrable by antineutrophil antibodies in cord blood, (NA1, 52–54%; NA2, 81–85%; NB1, 95–96%; NC1, 90%; 9A, 29–30%) suggesting that neutrophil antigens are fully e

ABSTRACT:Evidence of transplacental immunization in rhesus monkeys was obtained by testing postpartum sera from 252 females for antierythrocyte agglutinins. One‐third of the sera contained antibodies reactive with the mates' or the newborns' cells. Fetal erythrocytes were detected in the maternal circulation as early as 8 weeks after conception and as late as 12 days postpartum. Maternal antibodies were detected as early as 2 months after conception and persisted for more than 3 weeks postpartum. It was concluded that the fetal cells stimulated antibody production. Several features of transplacental immunization differ between rhesus monkeys and humans. A parity effect was not observed in rhesus. In fact, 33% of primiparous rhesus females produced antibodies. Also, several of the different allogeneic blood group factors appeared to be immunogenic but differed in immunopotency. Finally, direct antiglobulin tests indicated that erythrocytes of 11% of newborns were coated with maternal antibodies. Nevertheless, in contrast to humans, hemolytic disease was not observe
