ABSTRACT:To investigate the role of systemic suppression during allopregnancy, CBA/J female mice were immunised against H‐2dprior to mating. Cytotoxic T lymphocyte (CTL) activity was tested at days 14–16 of pregnancy. A reduction of CTL activity was observed only in multiparous animals. Although a nonspecific suppression was detected in isopregnancy, suppression was more marked in allopregnancy. Conversely, the CTL activity observed in the spleen during the first pregnancy (iso or allo) was always significant in mice presensitized with 2 or 3 alloimmunizations prior to pregnancy. Such animals have in vivo effector cells, since allografts of Sarcoma Sal were rejected in secondary fashion in alloimmunized mice, while the fetus remained unharmed. These observations demonstrate that allospecific anti‐MHC CTLs are specifically impaired in multiple allopregnancy, but also tend to rule out theories postulating that systemic suppression of CTL generation and function is required for successful allopreg

ABSTRACT:Guinea pigs were vaccinated intramammarly (IMM) with attenuated Mycobacterium bovis (BCG) and challenged intramammarly and intradermally with tuberculin. A significant (p<0.05) milk leukocytosis, consisting primarily of polymorphonuclear leukocytes, occurred from 6–30 hr after challenge with tuberculin. Intradermal challenge with tuberculin produced typical delayed‐type hypersensitivity cutaneous reaction in these animals. Normal guinea pigs were adoptively sensitized with lymphoid cells from the inguinal lymph nodes or spleen of BCG‐vaccinated animals and subsequently challenged intramammarly and intradermally with tuberculin. The mammary and dermal responses of the lymphoid cell recipients were similar to, but less pronounced, than those in the actively immunized animals. The responses in the recipients of lymphocytes from donors injected with tuberculin 18 hr prior to cell collection were greater than those from injected donors. Guinea pigs that were injected intraperitoneally with serum from BCG‐vaccinated donors did not express significant dermal or mammary responses to subsequent challenge with tuberculin. ABSTRACT:One‐way wife‐husband and wife‐donor mixed lymphocyte reactions (MLR) were performed in ten patients with hydatidiform moles and ten age‐ and maturity‐matched pregnant controls. ABSTRACT:Human seminal plasma has an anticomplement effect that can be measured by a standard immune hemolytic assay. We found that about 20% of samples lack complement‐inhibiting activity. To determine the anatomical origin of a complement inhibitor, inhibition of hemolysis was measured in seminal plasma samples that were primarily of prostatic, vesicular, or epididymal/testicular origin, as well as in seminal plasma from vasectomized men. All samples contained complement‐inhibiting activity, although epididymal fluid showed nearly twice the amount. Therefore, the factor (or factors) is ubiquitous in secretions of the male reproductive tract. Complement‐inhibiting activity is eluted in two high‐molecular‐weight peaks upon gel filtration. We found evidence that the inhibitory factor is not lactoferrin, a proteinase, or one of the proteinase inhibitors known to be present in human seminal plasma. ABSTRACT:General and specific immune function were examined in women with endometriosis. Nonspecific parameters included total leukocyte and differential counts, quantitative immunoglobulin (IgG, IgA, and IgM) determinations, total hemolytic complement, C3, C4, mitogen‐induced lymphocyte stimulation, and human leukocyte antigen (HLA) typing; no differences were observed when data were compared to age‐matched controls without endometriosis. In contrast, the specific immune response T‐lymphocyte‐mediated cytotoxicity to autologous endometrial cells was significantly reduced (P<0.01) in women with endometriosis. When results from patients were analyzed according to clinical severity of endometriosis, even more striking immunologic alterations were delineated. In addition, lymphocyte stimulation responses to autologous endometrial antigen were lower in patients with severe or moderate disease, approaching statistical significance (P = 0.18 and 0.12, respectively). ABSTRACT:Female mice were isoimmunized with homologous spermatozoa of the same strain. Hybrid cells that secrete monoclonal antibodies to mouse sperm isoantigens were generated by modified hybridoma techniques using a semi‐solid Iscove's modified Dulbecco's medium containing methylcellulose for the initial cloning. Out of more than 1,000 colonies that were initially recovered for subculture, 246 were shown to produce antibodies reacting with various cytological regions of mouse spermatozoa, when methanol‐fixed sperm were employed in an indirect immunofluorescent assay. More than 75% of the generated monoclonal isoantibodies were shown to bind the acrosomal regions of mouse spermatozoa. Some were found to cross‐react with spermatozoa from other mammalian species including those of human, rabbit, rat, and guinea pig. However, none were shown to cross‐react with mouse lymphocytes. Two‐thirds of the generated monoclonal antibodies can also bind live mouse spermatozoa. By an immunohistochemical technique using testicular sections, some of these monoclonal antibodies were shown to react with specific antigens expressed during different stages of spermatogenesis. It is concluded that these mouse sperm isoantigens are sperm‐specific and appear uniquely during spermatogenesis.
