ABSTRACT:Fresh E‐rosette positive lymphocytes from normal donors are not cytotoxic for the breast cancer‐derived target cell MDA‐157, although they do lyse other suspension and monolayer targets. E‐rosette positive cells from breast cancer patients in different stages of disease and from patients with benign breast disease also do not lyse MDA‐157. ABSTRACT:The cervical factor was evaluated in 52 infertile women with clinical signs of cervicitis. An abnormal postcoital test (PCT) was observed in 62% of the patients. Bacterial cultures from the cervix in these patients did not reveal any abnormal flora that could be held responsible for the abnormal PCT. The occurrence of Chlamydia trachomatis in four patients was comparable with the isolation rate in a general gynecologic out‐patient clientele. Increased levels of antichlamydial antibodies did not correlate with the result of PCT.On the other hand, 81% of the patients had colposcopically observed abnormalities, including an atypical transformation zone in 21%. Colposcopically directed biopsy specimens revealed severe inflammatory changes in 89% of the patients. Cervical intraepithelial neoplasia (CIN) grade 1 (mild dysplasia) was diagnosed in 11 (21%) patients. Treatment with cryocautery and tetracycline resulted in significant improvement in the result of PCT in 38% of the patients. The results indicate that cervical inflammatory disease in women with unexplained infertility must be carefully and extensively evaluated and treated, which often results in better outcome of the PC

ABSTRACT:Human spermatozoal polypeptides have been solubilized by incubation of spermatozoa in phosphate‐buffered saline. Inhibition of peptide release by the acrosin inhibitor N‐α‐tosyl‐L‐lysin‐chloromethyl suggests the direct involvement of the acrosomal enzyme in the liberation of the peptides. The peptides were coupled to activated Sepharose 4B gel and the peptide‐absorbed antibody of antispermatozoal antisera was detected with the aid of125I‐protein‐A tracer. The occurrence of three types of peptides became evident: one that binds to both sperm‐agglutinating and sperm‐immobilizing antibodies, one that binds to sperm‐immobilizing antibodies, and another that binds to sperm agglutinating antibodies only. ABSTRACT:Peptides representing the amino acid sequence of the carboxy‐terminal of the human chorionic gonadotropin (HCG) beta subunit were used in studies to determine whether an immunogen could be prepared that would be suitable for an HCG antifertility vaccine. Peptides of varying length were conjugated to several macro‐molecular carriers, in varying peptide‐carrier ratios, via an N‐ or C‐terminal group, with and without amino acid spacers. Antibody levels to peptides and intact HCG were used as the criteria for conjugate utility. Immunizations with a peptide of 37 amino acid residues elicited the highest antibody levels to HCG. No differences were found between N‐ or C‐terminal conjugates, but peptides with amino acid spacers elicited higher responses than peptides without spacer. Conjugates with a peptide:carrier ratio greater than 20 peptides/105daltons carrier were more immunogenic than those with a lower ratio. Diphtheria and tetanus toxoids were consistently the most effective carriers for enhancing antibody responses to peptides. It was concluded that conjugates of β‐HCG peptide 109–145 coupled to a toxoid carrier via its N‐terminus in a ratio of 20–30 peptides per 105daltons carrier was a suitable immunogen for further studies for the development of an

ABSTRACT:Peptides representing sequences of the beta subunit of HCG (Cys‐(Pro)6‐111–145 and 109–145) were coupled to tetanus toxoid (TT) and the resultant conjugates used to test the ability of eight experimental adjuvants and six different vehicles for enhancing antibody responses to the peptides in inbred strains of mice (C3H/He, C57BL/6, DBA/1, and SJL) and outbred rabbits. Antibody levels reactive to both125I‐peptides and125I‐HCG were used as criteria of efficacy of these adjuvants and vehicles. In most experiments, groups of animals were immunized with conjugates in complete Freund's adjuvant (CFA) for comparison. Two hydrophilic adjuvants‐NAc‐nor‐Mur‐L.ala‐D.isoGln, CGP 11,637 and NGlycol‐nor‐Mur‐L.α‐abu‐D.isoGln, DT‐1‐were consistently the most effective in stimulating antibody production to both peptides and HCG. An emulsion containing squalene‐water stabilized with arlacel A proved to be superior to other vehicles tested for administering conjugates and adjuvants to animals. Antibody levels produced by animals with different genetic backgrounds were shown to be polymorphic with respect to experimental adjuvants, but responses among groups using the same adjuvant and different vehicles were more consistent. ABSTRACT:Data are presented that suggest that the activation of cytochrome c‐specific T lymphocytes is dependent on the integrity of the three‐dimensional structure of the protein antigen. ABSTRACT:Female rats of the PVG/c strain were found to be approximately four times more susceptible than males to the induction of autoimmune thyroiditis by thymectomy and irradiation (Tx‐X) (female vs male incidence, 60%:14%). Similarly, the severity of the lesion was generally greater in females than males. Female Tx‐X rats also had a significantly higher incidence of antibodies to thyro‐globulin than males (incidence, 67%:24%, respectively) and developed higher titers.Prepubertal orchiectomy had a profound influence on both the severity and incidence of thyroiditis. Orchiectomized Tx‐X males were found to have changes approximating those of Tx‐X females (72%:60%, respectively). A corresponding influence was also noted on the incidence and titer of antibodies to thyroglobulin (incidence, 78%:67%, respectively). Postpubertal orchiectomy similarly intensified the incidence and severity of the disease.Ovariectomy also caused a slight but significant increase in autoimmune thyroiditis as compared to entire Tx‐X females (78%:60%, respectively).Sham‐gonadectomized control rats did not differ in overall susceptibility from entire Tx‐X rats.These observations indicate that the incidence and extent of autoimmunity induced in this particular model are modulated by sex steroid hormone production rather than by sex‐linked gene(s) directly determining susceptibility. ABSTRACT:The expression of transplantation antigens by cells of the placenta was examined by immunohistological and immunoprecipitation procedures with defined conventional and monoclonal antisera to beta2‐microglobulin, DR and DC gene products, and H‐Y antigen. Cells of the mesenchymal stroma within chorionic villi were positive by immunofluorescence for major histocompatibility complex antigens, and in male pregnancies for H‐Y antigen, but the trophoblast was consistently negative for all antigen systems examined. Immunohistological examination of viable suspensions of cultured diploid trophoblast and of isolated membranes also gave negative results, and after radioiodination and solubilization of membranes, no detectable radioactive material was immunoprecipitated. These results provide further evidence that transplantation antigens are not expressed by human trophoblast. Since this is a fetal structure exposed directly to immunologically competent cells of the mother in the intervillous spaces, this observation may be relevant to the apparent lack of damaging maternal immune responses directed against the fetal homo

ABSTRACT:The antigens recognized by heteroantisera raised to human trophoblast membrane were studied on a variety of normal tissues and certain neoplastic cell lines, both by immunohistological and immunoprecipitation techniques. Native antisera reacted with all tissues examined, but after absorption with normal human serum and lyophilized normal liver, reactivity with normal tissues was restricted to the trophoblast membrane, endocervix, and mitogen‐ and antigen‐stimulated lymphocytes. Several membrane components were precipitated from trophoblast and activated lymphocytes by native antisera, whereas after absorption a single radioactive peak of MW 62,000 was obtained. HeLa, Chang, AV3, and HEp‐2 cell lines were also positive by immunofluorescence with the absorbed antisera, and a single molecular species was again precipitated. Additional enzymatic studies both of immunoprecipitated material and of the tissue of origin provided evidence that this species was the placental isoenzyme of alkaline phosphatase. These results indicate that the major specificity in absorbed trophoblast antisera is the heat‐stable placental isoenzyme of alkaline phosphatase. Furthermore, since similar material appears to be present on other normal and transformed tissues, this isoenzyme may not therefore, despite previous claims, be truly trophoblast
