ABSTRACT:Recently the concept of a poorly functional humoral immune response in the newborn was proposed. Data have been presented indicating that the impaired newborn B cell maturation, as shown in vitro in a pokeweed mitogen‐induced B cell maturation system, is due both to an immaturity of lymphocyte subsets and to an increased suppressive T activity. In the present work, we present evidence that there exists a predominance of a naturally occurring T lymphocyte suppressive activity in the cord blood in that the removal of the suppressive activity by irradiation allows a normal maturation of newborn B cells. Such normal maturation of newborn B cells can also be obtained using mixed cultures of adult T cells and newborn B cells. Newborn suppressor T cells belong to both EAγ(+) and EAγ(—) fractions, and it is not known whether these two groups do or do not belong to different subsets. The PGE2‐dependent monocyte suppressive activity does not play any role in the suppression observed in newborns since newborn monocytes are poorly suppressive and since they produce a smaller amount of PGE2than adult monocytes. Some observations suggest, on the contrary, that the suppressive T lymphocytes can regulate the level of the PGE2‐dependent monocyte suppressive activity.It should be noticed that similar observations about T lymphocyte and PGE2‐dependent monocyte suppressive activities have been made at the same time using mothers' cells. ABSTRACT:Using rabbit as a model, the roles of uteroglobin (UG) and transglutaminase (TG) in masking the antigenicity of early developing mammalian embryo have been investigated. Maternal lymphocytes in vitro, when mixed with mitomycin‐C inactivated blastomeres, incorporated H3‐thymidine, suggesting recognition of embryonic antigens by these cells. However, pretreatment of blastomeres with pregnant uterine fluid (PUF) or with UG alone or in combination with TG (coagulation factor XIIIa), resulted in a significant and dose‐dependent suppression of H3‐thymidine incorporation into these lymphocytes. A complete suppression was achieved at a concentration of 250 μg of UG/ml, in the absence of TG. However, in the presence of TG, only 1.0 μg of UG/ml was required for total suppression. Neither nonpregnant uterine fluid (NPUF), nor myoglobin, a nonspecific protein similar in molecular weight to UG, had any suppressive effect. Incubation of uteroglobin with anti‐UG or TG with its antiserum prior to the pretreatment of blastomeres eliminated the suppressive effect of these proteins. Inhibition of TG by neopentyl chloroethyl nitrosourea (NPCNU) also eliminated the suppressive effects of uteroglobin on H3‐thymidine incorporation into maternal lymphocytes. These results suggest that in the pregnant uterus UG in conjunction with TG may play a specific role in masking the antigenicity of developing embryos during

ABSTRACT:Physiologic changes occurring during the latent period of prematurely ruptured membranes (PRM) are poorly understood. Indicators predicting maternal and neonatal infectious morbidity do not correlate well with clinical outcomes. A previous report suggested that in vivo complement consumption occurred in response to the event of membrane rupture. In this prospective study, complement activity was measured serially throughout the latent period in cases of preterm PRM. In addition to total hemolytic complement activity (CH50), C3‐proactivator (C3PA), a primary component of the alternate complement pathway, was measured in maternal and cord sera. As with CH50, cord serum C3PA levels are significantly less than those in matched maternal samples. ABSTRACT:A common base population of White Leghorn was “synthesized” for a joint project on the genetics of egglaying, undertaken by animal breeding geneticists in 4 Scandinavian countries. After 6 to 7 generations of line selection for various egg‐laying parameters, MHC typing was undertaken of both the selection lines in Denmark, Norway, and Sweden and the respective control lines representing the common base population. ABSTRACT:The antigenic potency of porcine zona pellucida has been studied in the rabbit using low‐dose immunization (50, 100, 200, and 400 zonae per injection), and the subsequent level of anti‐zona antibodies was determined. The antibody titers as determined by the indirect immunofluorescence test (IF) and the zona precipitation reaction (PR) were 1:640 and 1:80, respectively, after immunization with 100 zonae. There was no difference in antibody titers between the immunization of 400 suspended zonae and 400 heat‐treated zonae. Injection of homogenized porcine ovary also resulted in titers of 1:640 and 1:80 (IF and PR, respectively). Immunization, however, with only 50 isolated zonae did not produce a measurable antibody titer. Immunization with 1200 intact or heat‐treated zonae resulted in transient infertility.The present study demonstrated that the limiting quantity of zonae required for detectable antibody production was between 1.65 μg and 3.30 μg zona protein as monitored by immunofluorescence and immunoprecipitation assays.
