摘要:
ABSTRACT:A case of congenital listeriosis presenting with premature labor at 24 weeks' gestation is described. Listeria monocytogenes were isolated from the amniotic fluid, which also had an increased concentration of IgA. The significance of amniotic fluid Ig levels and their potential value in the diagnosis of intrauterine infections as an adjunct to the isolation of the infectious agent is discussed.

ABSTRACT:In order to further examine the inhibition of lymphocyte activation reported with conventional antisera to human trophoblast membranes, we studied their effects on cells stimulated by antigen in mixed lymphocyte culture (MLC) and by the mitogen phytohemagglutinin. The results were compared with the effects of antisera known to recognize intrinsic membrane determinants on activated T cells (DR antigens and placental alkaline phosphatase) and with those of antisera to normal human serum components such as transferrin which may bind to the activated lymphocyte membrane. The results indicated that antibodies to placental alkaline phosphatase, which constitutes the predominant specificity of conventional trophoblast membrane antisera, caused inhibition both of MLC response and, to a lesser extent, of activation induced by mitogen. Similar inhibition was obtained with antisera to human DR antigens, while antisera to normal human serum and transferrin were not suppressive. These findings, together with time course studies and immunocytochemical studies of the homologous antigens, suggest that these antisera mediate their inhibitory effects in part through binding to antigens which appear on cells as they undergo activation.

ABSTRACT:In view of the protective and enhancing effect of blocking antibodies (BA) on the survival of trophoblastic cells, the presence or absence of individually specific BA against paternal HLA antigens in trophoblast‐bound immunoglobulin G (IgG) of molar as well as normal term placentas were investigated using a mixed lymphocyte culture reaction (MLR) blocking assay and complement‐dependent lymphocytotoxicity test.It was found in this study that IgG could be eluted from molar trophoblasts. Further, it was strongly suggested that each molar and normal placental eluate‐IgG had individual, specific cytotoxic and blocking effects against paternal lymphocytes and on MLR between spouses, respectively.In conclusion, in molar as well as in normal term placenta‐bound IgG, BA that are heterogeneous and partly contain IgG specific to the paternal HLA antigens, including HLA‐D/DR antigens, are likely to exist. ABSTRACT:Trophoblast cells of the placenta represent the frontier of the conceptus at the feto‐maternal interface, exposed to the maternal immune system. The presence or the absence of major histocompatibility (MHC) antigens on trophoblast cells, thus, is a key determinant of immune interactions between the mother and the allogeneic fetus during pregnancy. This article critically reviews the studies of the alloantigenic status of murine and human trophoblast cells, including the published data in the mouse and unpublished data in the human in the authors' own laboratory. It is shown that, in both these species, a significant proportion of trophoblast cells express Class 1 but not Class 2 MHC antigens in a manner directly accessible to the maternal immune system. However, the lack of immunogenicity of semiallogeneic trophoblast cells can not solely be explained on the basis of the absence of Class 2 molecules, despite the presence of Class 1 molecules. It is proposed either that the Class 1 MHC antigens on the trophoblast cells are presented in a nonimmunogenic manner to the maternal T cells or else that certain cells at the feto‐maternal interface (eg, fetally derived trophoblast cells, maternally derived lymphoid cells, or decidual cells) elaborate potent immunosuppressor molecules capable of abrogating T cell alloreactiv

ABSTRACT:Fetal suppressor cells are potentially important for maintenance of pregnancy and for the immune regulation of the identification of self. We have identified and partially characterized a population of murine fetal liver cells which are active suppressors of a variety of immune responses, including mixed lymphocyte responses (MLR), responses to mitogens plaque‐forming cell responses to heterologous erythrocytes. The suppression is non‐H‐2‐restricted and can act via a factor. The cell is preactivated in fetal liver and may represent a T‐cell precursor. ABSTRACT:The presence of fetal antigens on a wide variety of tumors has been well‐documented it has been attributed, for the most part, to de‐repression of genes normally operative in the early stages of embryonic development. Controversy still exists, however, about the role of fetal antigens in antitumor immunity. It is not clear whether the tumor inhibition shown in some systems is due to an immune response. In the present study, conducted in a weakly immunogenic Lewis T241 fibrosarcoma, syngeneic for C57B1/ 6J mice, immunization of normal mice with syneneic fetal cells resulted in striking inhibition of growth and metastases of tumors. On the other hand, immunization with syngeneic normal, adult spleen cells or with allogeneic A/J fetal cells did not inhibit tumor growth or metastases. Mice that had gone through single pregnancy also showed inhibition in tumor growth and metastases. Immunization of female mice with syngeneic tumor cells or fetal cells, prior to pregnancy, resulted in a high incidence of fetal death in these mice. Further studies showed that tumor inhibitory response evoked by fetal cell immunization was due to fetal antigens the male‐specific HY antigen was not responsible for antitumor response. When tumor cells were mixed with spleen cells from fetal antigen‐immunized mice and then injected into normal mice (Winn neutralization assay), significant inhibition of tumor growth and metastases was observed. In this assay, mixing tumor cells with syngeneic fetal cells, normal adult spleen cells, or spleen cells from C57 mice immunized with allogeneic fetal cells did not inhibit the tumor growth or metastases. These results show that inhibition in tumor growth and metastases, following fetal cell immunization, was due to a specific immune response to the oncofetal antigens involved. ABSTRACT:The beta‐human chorionic gonadotropin (HCG) radioimmunoassay was used to determine the presence of HCG immunoreactivity in serum of patients (n = 71) with diagnosis of cancer. Of patients with active neoplasia, 60.5% showed HCG immunoactivity above controls (>>5 mlU/ml). An apparent degree of correlation was observed with tumor activity in that a case with widespread metastases due to a colonic carcinoma exhibited the highest HCG levels while, in one patient, the level of HCG decreased progressively according to therapeutic response. A high frequency of immunoactive HCG was found in patients with carcinomas of the cervix, breast, gonad digestive system and in patients with melanoma. Trophoblastic cells were not evident in the tumors biopsied. Immunologic similarity of HCG secreted by tumors and that contained in serum of pregnant women, of patients with hydatidiform mole of males injected with exogenous HCG was shown by parallel inhibition curves in the radioimmunoassay.
