ABSTRACT:Previous studies in mice and humans have shown that the first IgA+and IgG+B cells appearing during ontogeny also bear IgM on the surface. The acquisition of IgD seems to occur at a later stage in differentiation. In this study we have combined autoradiography with two‐color immunofluorescence to directly detect human B cells expressing three surface isotypes. We report that the vast majority of IgA+cells in the neonate also bear IgM and IgD; the same holds true for IgG+cells. This phenotype is peculiar of newborns, while in the adult the majority of IgA+and IgG+cells are single. We discuss the genetic implications of such a finding for heavy chain switchin

ABSTRACT:Isoelectric focusing (IEF) on a thin layer of polyacrylamide gel revealed that strain A/J produces antibodies with pI values distributed over the whole assayed pH range (pH 5.05–7.75), while strain B10 produces antibodies of a limited heterogeneity with pI values distributed between pH 6.3 and 7.5. The analysis of sera of congenic resistant (CR) and recombinant strains showed that haplotype H‐2ais associated with wide‐heterogeneity antibodies, while the presence of haplotype H‐2bis associated with the occurrence of limited‐heterogeneity antibodies. Thus a gene located within the I region of the major histocompatibility complex (MHC) may affect the degree of heterogeneity of the anti‐sheep red blood cell (SRBC) response. The gene is transmitted to the F1generation as a dominant trait.After administration of endotoxin (LPS LT2, 10 μg/mouse) to the B10 mice, antibodies with pI values below pH 6.3 and above 7.5 appear in the serum; the heterogeneity of these antibodies resembles that of the A/J strain

ABSTRACT:The analysis of anti‐sheep red blood cell (SRBC) antibody production in the congenic resistant (CR) mouse strains A/J and A.BY, B10 and B10.A showed that the level of IgG antibodies after immunization with SRBC is controlled by a gene(s) localized in the H‐2 complex. The use of the h2, h4, and i5 H‐2 recombinant haplotypes allowed us to map this gene into a region proximally defined by subregion I‐J and distally by region H‐2G. The IgG antibody level is simultaneously under the influence of non‐H‐2 genes, of which those of the A/J strain origin determine the high IgG level and those of the B10 strain origin determine the low IgG level.Weights of spleens of the A/J and B10 mice before and after primary and secondary immunization with SRBC were compared. Before immunization, spleens of the A/J mice were approximately 10% lighter than those of the B10 mice (0.08 g vs 0.09 g). After the first immunization the spleen weights equalized (θ = 0.12 g). Four days after the second immunization the weight of the spleens of A/J mice increased by 312% (relative to the nonimmune state) while that of the B10 mice increased by only 74%. ABSTRACT:Employing the techniques of in vitro lymphocyte transformation (LTF) and complement fixation, cell‐mediated immunity (CMI) and antibody to cytomegalovirus (CMV) were studied in pregnant and nonpregnant women. The LTF activity was determined by the whole blood microassay using four strains of CMV (AD‐169 and its early antigen [EA], Davis, Veca, and Towne strains), and phytohemagglutinin (PHA).Lymphocyte transformation response to specific CMV antigens at 11–30 weeks of gestation and to nonspecific mitogen (PHA) in all pregnant and postpartum women were found to be significanty depressed compared with the nonpregnant women. The lower LTF responses to CMV antigen and PHA were found in specimens taken from pregnant women at 21–30 weeks of gestation. There were no significant differences in the mean complement‐fixing (CF) antibody titers and the percentage of E‐rosette‐forming T lymphocytes between subjects in various stages of pregnancy.In addition, concanavalin A (Con A)‐generated suppressor T cell activity was evaluated in pregnant and nonpregnant women. The suppressor effect of Con A‐activated lymphocytes in pregnant women was somewhat higher than in nonpregnant women. These observations suggest that CMV‐specific suppression of cellular immunity may play an important role in reactivation

ABSTRACT:The conceptus is tolerated in spite of the antigenic dissimilarities between mother and fetus. A new hypothesis to account for this phenomenon is put forward. It is suggested that the vessels within the trophoblastic villi—analogous to the vessels of malignant tumors—are permeable to humoral substances but impervious to cellular elements. This property allows for the exchange of nutrients and metabolites but, at the same time, precludes the passage of leukocytes. The block to diapedesis is believed to be due to the deposition of an occlusive layer of alpha2‐macroglobulin on vascular endothelium. The source of this protide is the fetal liver. ABSTRACT:In this study, betamethasone was found to significantly inhibit the binding of immunoglobulin‐coated O Rh‐positive red cells to fetal macrophages. ABSTRACT:Experiments were performed to determine whether uterine secretory proteins (PAP) of the pig affect T cell or B cell functions in vivo. T cell functions examined were E‐rosette formation (ERF) and cutaneous reactivity (to phytohemagglutinin) in mature BALB/c females. Two ERF experiments of identical design were performed using five groups of ten mice each. Each mouse was presensitized with sheep erythrocytes (SRBC) designated as day 0; and the various groups were treated with saline, pig serum (PS), or PAP at different times. For cutaneous reactivity, the area of erythema at the sensitization site (pinnea of ear) was measured, and three groups of 11 mice were used. These groups also received either PS or PAP. B cell function was examined by measurements of hemagglutination titers to SRBC sensitization in BALB/c females treated in two experiments similar to the ERF. T cell functions examined were highly suppressed (p<0.01) by PAP given hours, not days, prior to the measurement. The B cell function examined was not affected by PA
