AbstractExperimental conditions that best distinguish the abnormal susceptibility to SV40 found in skin fibroblasts from patients with hereditary adenomatosis of the colon and rectum (ACR) are reported. T-Antigen expression of these SV40-in-fected human cells was largely dependent on cell density and was optimal at a concentration of 1000-2000 cells/cm2. Significantly, the abnormal susceptibility of ACR cells to SV40 was greatest and most reproducible at this density. The transformation of these cells by SV40 was also dependent on the multiplicity of infection (MOI), represented by the linear portion of the curve (range about 100-1000 pfu) for both normal and ACR cells. Under these conditions, ACR cells were about sixfold more sensitive to SV40-induced T-antigen expression and transformation. These results provide additional support for the use of SV40 to identify humans at risk from ACR families as well as other dominantly transmitted forms of human cancer.

AbstractFibronectin (Fn) synergizes with macrophages (MϕS) in inducing cytostasis and cytotoxicity of neoplastic cells in culture. Since heparin enhances Fn's opsonic activity in many systems, we investigated its effect on Fn-macrophage synergy in cytostasis. MCG-T14 (a spontaneous mouse mammary adenocarcinoma) cells (4×104) were added to wells both with and without C. parvum activated MϕS monolayers. To these cultures were added increasing concentrations of Fn with or without heparin. Fn synergizes with both MϕSs and heparin in inhibiting tumor cell growth. The combined cytostatic effect of Fn, heparin and MϕSs is more than additive. In other experiments, MCG-T14 cells were pre-incubated for 2 hr with Fn, washed free of Fn, and treated as above. The results of these experiments were similar to coculture experiments, but the effect of heparin was even more pronounced. Dermatan sulfate and hyaluronic acid had a variable effect on Fn and Fn-macrophage induced cytostasis.

AbstractA case of benign fibrous histiocytoma in a patient with previous Hodgkin's disease is reported. Benign fibrous histiocytoma consists of a proliferation of benign histiocytes in addition to inflammatory cells and occasional Reed-Sternberg-like cells. Hodgkin's disease is considered a malignant disorder of the monocyte-histiocyte cell line but also includes benign inflammatory cells as part of its histological picture. The possibility that these two entities are biologically related is considered. The pathogenic relationship between both entities is discussed in the context of the immune dysregulation associated with Hodgkin's disease.

AbstractMalignant lymphomas disrupt the anatomical organization of lymphoid organs and are recognized by patterns of growth and the morphology of the abnormal cells. The phenotype of these cells can be determined with precision by the demonstration of cell surface receptors and antigenic determinants by standard immunologic procedures. Most B-cells are found within discrete follicles and display characteristic cytologic and immunologic features. Although malignant proliferations of B-cells do not follow normal structural boundaries.
