Fibrinolytic Activity in Human Tumor Tissues |

The fibrinolytic activity of 156 malignant and 36 benign solid tumors from autopsy and biopsy specimens was studied by the fibrin slide technique. The inhibitory activity against fibrinolysis was graded according to the lysis time of vascular tissues within the tumor. The results show that all malignant solid tumors, with the exception of prostate carcinoma, demonstrated varying degrees of inhibition of fibrinolysis. Persistently high inhibitory activity was found in squamous cell carcinoma of the esophagus, the respiratory tract, cervix uteri, and skin; carcinoma of uterus; colorectal carcinoma; small cell anaplastic carcinoma of lung; neuroblastoma, carcinoma of bile duct, while malignant tumors of the kidney show a lesser degree of inhibition. In contrast, with the exception of the hydatidiform mole, benign solid tumors show little or no inhibition. A similar absence of fibrinolytic activity is seen in metastatic disease. Further studies of the role of the fibrinolytic system in tumors seems warranted.

A cytogenetically normal man with gynecomastia and a family history of diverse cancers developed adenocarcinoma of the breast 30 years following thymic irradiationIn vitroexperiments measuring colony-forming ability of cultured skin fibroblasts from family members implied that the patient had a small but significant increase in sensitivity to ionizing radiation, and a moderate increase in sensitivity to bleomycin, a radiomimetic drug. Enhanced radiosensitivity of fibroblasts from the patient's mother, and bleomycin sensitivity of fibroblasts from the sister suggested, but did not prove, that genetic susceptibility affected the risk of radiogenic cancer in this individualIn vitrostudies of cancer-prone kindreds are a useful research strategy in delineating mechanisms of carcinogenesis.

Nitrogen-13-L-glutamate, a labeled amino acid enzymatically synthesized from cyclotron-produced N-13 ammonia and alpha-ketoglutaric acid, was used as an imaging agent in 14 patients with soft-tissue sarcomas. Concentration of nitro-gen-13 label within the sarcoma was seen in 11 of the 14 patients; in some cases, clinically inapparent metastatic tumor lesions could also be detected. In eight patients, the tumor response to chemotherapy was evaluated by serial studies with this agent. In each case, the change in uptake of N-13 label by the sarcoma after therapy paralleled the clinical response to treatment. N-13-labeled L-glutamate may thus be of value as an imaging agent in the management of patients with soft-tissue sarcomas.

Tumor promotion research has accelerated at an explosive level during the past decade and continues to do so because of its importance to the understanding of the induction of human cancer. Furthermore, the promotion process, being mostly reversible allows one to find very effective ways to prevent cancer. The extensive data available as well as the multistage nature of tumor promotion suggests that this process, which is thought to occur in most tissues where cancer can be induced or occurs spontaneously, may involve the interaction of a number of environmental factors such as chemical, radiation, viruses and diet and nutrition, thus unifying all of the current areas of cancer research. In terms of human cancer, smoking, asbestos, radiation, alcohol and diet, and nutrition just to mention a few, are not thought to have more than a promotional influence in the carcinogenesis process. Here and in later papers we will discuss some important aspects of promotion as well as discuss the possible controversial nature of this process. In this review we present data which suggest that carcinogenesis can be operatively and mechanistically divided into various stages.

Progress in research usually follows rapidly after the development of new laboratory techniques. The editors ofCancer Investigationbelieve that it is important from time-to-time to include articles which will review“state-of-the-art”technical advances in methodology, instrumentation, and computer sciences. It is our hope to provide investigators with clear and useful information which they may wish to utilize in their own research programs. It is not our intent to provide step-by-step details in those articles, but rather information on what and how a technique is about: its uses, advantages, and limitations. The first in this series is devoted to high performance liquid chromatography, a technique which seems to have unlimited uses in studies of metabolites and drugs. We hope that our readers will enjoy this series. The Editors appreciate suggestions from the readers concerning topics for future issues.

This review of HPLC technology is presented in two parts. An introduction to chromatography, sample preparation, and basic information on chromatographic theory and terminology, modes, and instrumentation is presented here. A discussion of stationary and mobile phase parameters which can be varied to optimize separations is also presented here. The second part of the review, which will appear in the next issue, will cover peak identification, quantification, and selected applications. The applications include short discussions of applications of HPLC for metabolic profiling, for assays of adenosine and cyclic nucleotides in studies of disease processes, and for enzyme assays. The paper concludes with a short discussion of future trends for HPLC in biomedical research and clinical chemistry.
