SummaryThe expression of the transferrin receptor (TfR) was studied in the acute leukaemias and lymphoproliferative disorders by means of indirect immunofluorescence and/or the enhanced alkaline phosphatase anti‐alkaline phosphatase (APAAP) techniques using monoclonal antibodies to the receptor. A total of 174 cases of acute leukaemia and lymphoproliferative disorder were studied. The results indicate that the receptor is expressed with increased density in the majority of positive cases of acute leukaemia. The lymphoproliferative disorders, however, only expressed the receptor in a minority of cases and did so with weak density. It is proposed that this weak expression i n the lymphoproliferative disorders may be of use as an indicator of an increase in cell activit

SummaryThe underestimation of WBC by the Ortho ELT800/WS cell analyzer in some cases of chronic lymphatic leukaemia has been investigated. Incorrect WBC was obtained on 44|X% of samples. SummaryIt is generally recognized that the standardized cyanmethaemoglobin (HiCN) method is the preferred method for the determination of haemoglobin in blood. In automated analysers, however, the reaction time available before the photometric measurement is too short. With the Ortho ELT800 we found that the conversion into HiCN took longer than 25 s. More important was the variation of results with the haemoglobin derivatives present in the blood sample. Our experiments suggest that Coulter counters and other automated analysers as well use denaturated globin haemichrome as the end product in their haemoglobin procedure although they profess to use HiCN by the addition of cyanide to the reagents. However, haemichrome is a suitable alternative for automated analysers as the conversion time is very short for all hacmoglobin derivatives including HbCO. In the Coulter S Plus 111 where white cell size distribution analysis and haemoglobin determination are performed in the same solution, the haemoglobin derivative formed shortly after mixing is not a stable one. White cell size analysis and haemoglobin determination should preferably be performed in separate channels with different lysing reagents giving optimal conditions for each determination. Evaluation of the analysers would be easier if the manufacturers would give correct information on the chemistry of the haemoglobin method and state the composition of the reagents used.

SummaryA practical instrument for routine haemoglobinometry (HemoCue) was compared with the reference haemiglobincyanide (methaemoglobincyanidc (HiCN)) method. The precision of the routine method was adequate, but a systematic error of about −3.5|X% was found. As this resulted from errors in the procedure for coupling the haemoglobinometer read‐out to the reference HiCN method, we drew up an improved calibration procedure for practical haemoglobinometers to ensure comparability of results with the reference method. After recalibration according to these rules, the HemoCue system proved to be accurate to within |Mp1.5

SummaryMacrophages obtained from human monocytes by monolayer culture were stimulated with recombinant immune interferon. They were compared with unstimulated macrophages and monocytes as indicator cells in the mononuclear phagocyte assay, using both IgG anti‐Rh(D)‐coated complement‐coated red cells. The presence of the interferon in the culture medium improved the adherence of macrophages in monolayers. The interferon.markedly augmented the number of IgG‐coated red cells which became attached to the macrophages and reduced the amount of antibody needed for red cell‐macrophage interaction. This stimulatory effect occurred regardless of the IgG subclass composition of the anti‐Rh(D) antibody. The activity of the stimulated macrophages to interact with IgG‐ or complement‐coated red cells was considerably greater than that of monocytes. The results imply that macrophages treated with recombinant immune interferon are more sensitive than monocytes as indicator cells in the mononuclear
