Lysozyme was demonstrated to increase in content in the human intervertebral disc in a significant age-dependent manner. In young discs ( 65 years) the concentration was 3,460 U/g wet weight. These findings contrasted with the hexuronate level (as a marker of proteoglycans) and neutral sugar content of the tissues which decreased with age. The level of lysozyme determined in the disc tissue also correlated with the morphological grade of disc degeneration.

Since very little is known about the impact of aging on adrenocortical function in female rats, it was of interest to compare the age changes in the circulating levels of adrenocorticotropin (ACTH) and corticosterone in male and female Sprague-Dawley rats, and correlate these changes with those in plasma prolactin (Prl), which is known to stimulate adrenal steroid secretion. Hormones were measured in young (3–4 months) and old (24–26 months) male rats as well as in young (3–4 months), old (25 months) and senescent (33–35 months) female rats. Sequential plasma samples were obtained from chronically cannulated animals every 30 min from 11:30 a.m. to 4:30 p.m. ACTH was measured in trunk serum – obtained between 11:30 a.m. and 1:30 p.m. – whereas corticosterone and prolactin were measured in plasma using specific RIAs for the corresponding hormones. No age changes were detected in serum ACTH in either sex. The integrated values of plasma corticosterone did not change with age in males but decreased significantly (p < 0.05) in old and senescent as compared to young females. Plasma corticosterone was higher in young females than in young males but this sex-related difference disappeared with age. Plasma Prl increased significantly with age in both male (p < 0.05) and female (p < 0.001) rats but showed no significant correlation with corticosterone levels. The present results suggest that ACTH secretion does not show major age- or sex-related changes. The median and maximum life spans of Zaprionus paravittiger are 43 and 79 days, respectively, for males and 52 and 91 days for females at 24 ± 2°C. Sodium selenite (SS), an antioxidant, feeding prolongs the median as well as maximum life span of both the sexes. Antioxidative enzymes, catalase and peroxidase showed greater activity during the developmental stages. Females, at most of the age intervals, exhibited greater enzymatic activities as compared to their age-matched males. Both the sexes showed a statistically significant increase in the baseline activity of these enzymes as consequent upon SS feeding. The relation between carotid blood flow measured by Doppler spectrum analysis and brain atrophy on computed tomography (CT) was studied in 22 subjects with multiple lacunar cerebral infarctions. The subjects were divided into two groups, 7 patients with multi-infarct dementia (MID) (mean age 73 years) and 15 nondemented lacunar stroke subjects (NDLS) (mean age 66 years), according to DSM III criteria. All subjects had a score of 7 points or more on Hachinski’s ischemic score and showed no carotid artery stenosis. Systolic peak frequency of the common carotid artery (CCA) was measured by Doppler spectrum analysis (Angioscan II). Brain atrophy was measured quantitatively on CT images by two-dimensional measurement using a digitizer. Peak frequencies were lower in MID than in NDLS. Brain atrophy was more severe in MID than in NDLS. There was a significant correlation between peak frequencies and brain atrophy in all subjects. These results indicate that CCA blood flow may reflect brain function in patients with multiple lacunar infarction

The serum concentration of 25-hydroxyvitamin D level and plasma albumin-adjusted calcium, phosphate, and alkaline phosphatase levels were studied in 200 patients with hip fracture (age range 49–93 years) and 427 elderly subjects living in the community (age range 60–90 years). The mean serum 25-hydroxyvitamin D levels in controls were higher than in temperate countries, but the 25-hydroxyvitamin D concentration was significantly lower in the patients than the controls for all sex and age groups. There was little difference in albumin-adjusted calcium and alkaline phosphatase levels, but the phosphate level was higher in the patients than in the controls. None of the patients with a low 25-hydroxyvitamin D level had a blood picture suggestive of osteopathy resulting from vitamin D deficiency or frank osteomalacia. Hip fracture patients with a low 25-hydroxyvitamin D level were much less ambulant and went outdoors much less frequently than hip fracture patients with a normal vitamin D level. We have shown that the majority of elderly female patients in continuing care are in a hyperosmolar state. In a cross-sectional analysis of 2,301 ambulatory elderly subjects, the mean resting heart rate for women (66.2/min) was significantly higher than for men (63.6/min), p < 0.0001, and heart rate was positively correlated with age for both men and women. Resting heart rate was positively correlated with the number of drugs used (p < 0.05), number of symptoms reported (p < 0.05), systolic (p < 0.01), and diastolic (p < 0.0001) blood pressure. Of 975 participants who were followed longitudinally over an 8-year period there was a significant decrease in resting heart rate of approximately 2.75/min, p < 0.0001.

The activity of total and heat-stable glutamate dehydrogenase (GDH) in lymphocytes increased with aging in 40 control subjects (p < 0.001). The total GDH activity was lower in patients with olivopontocerebellar atrophy (OPCA), motor neuron disease, Parkinson’s disease or Alzheimer’s disease than in age-matched control subjects, but not in patients with nondegenerative neurological diseases. The decrease in heat-stable GDH activity was observed in patients with OPCA, Parkinson’s disease or Alzheimer’s disease, but heat-labile GDH was decreased only in patients with OPCA.

The investigation of thyroid disease in the elderly is difficult. Conventional tests are influenced by ill health and abnormal concentrations of thyroid binding proteins. To screen for thyroid disease thyroxine (T4), then tri-iodothyronine (T3) or TSH and an index for binding proteins are normally measured, methods which are expensive and time-consuming. TSH immunoradiometric assays (IRMA) offer the potential of a single test assessment of thyroid function in the elderly. This was assessed by comparing the measurement of IRMA-TSH with T4, T3 and the free thyroxine indices in 290 elderly subjects. Concordant results were obtained in 236 subjects. Non-concordant results were seen in 54 subjects. The commonest cause was sick euthyroid syndrome but 4 cases of high T4 and normal TSH were seen with amiodarone therapy.
