Cyclic AMP generation was measured in isolated minces of the ipsilateral and contralateral cortex of young (3–4 months) and aged (28–29 months) Fischer-344 rats, 2 weeks after unilateral lesion of the nucleus locus ceruleus (LC). Measurements were conducted under basal conditions and in the presence of 0.1 mM isoproterenol. Despite marked norepinephrine (NE) depletion of about 90% in the ipsilateral cerebral cortex in both age-groups, cyclic AMP generation under basal conditions was unaffected. However, whereas isoproterenol-stimulated cyclic AMP generation was significantly higher in the NE-depleted ipsilateral cerebral cortex of young rats, such ‘denervation supersensitivity’ was not apparent in aged rats. Also, endogenous levels of NE were 31% higher, and isoproterenol-stimulated cyclic AMP generation was 33% lower in the contralateral ‘control’ cortex of aged rats than in young rats. These results suggest an age-related defect in the postsynaptic expression of noradrenergic mechanisms in the rat cerebral cortex. The influence of age on the number of receptors for insulin and glucocorticoids on human T cells was examined. The specific binding of 125I-insulin and 3H-dexamethasone to phytohemagglutinin-(PHA)-stimulated T cells was found to decrease with age. Populations of PHA-stimulated T cells, however, are heterogeneous with respect to cell cycle phases, and cells in different cell cycle phases have been shown to bear different numbers of receptors. Therefore, it was not clear whether the measured decrease in specific binding in cultures from aged donors was due to a decreased number of activated cells or to a decreased number of binding sites per cell. In parallel to measurements of receptors, performed at different times following stimulation (20 and 44 h), numbers of cells in the different early cell cycle phases G0, G1a and G1b were quantitated by flow cytometry. In this way the receptor number per cell in each phase of the cell cycle could be determined. Receptor numbers on resting cells and in the earliest phase of activation (G1a) were found not to be influenced by age. A decreased receptor density, however, was apparent on G1b cells from aged donors.

Ventilatory parameters such as tidal volume, minute ventilation (VE), and inspiratory flow rate decrease in 24- vs. 12-month-old male and female rats. Differences between male and female values are maintained with age. Ventilatory response male and female rats exhibit to hypercapnia is altered by a decrease of the intercept, but not the slope value. The pattern of breathing exhibited by young females and males in response to hypercapnia (i.e. an increase of VT and f) is different than that noted in old males and females (i.e. an increase in VT only). In contrast, the ventilatory response both of slope and intercept male rats show by 24 months is decreased compared to the 12-month value; but the slope value is actually increased in the older vs. younger female rats in response to hypoxia.

Aluminum concentrations in the whole organism increased during development and aging of Drosophila melanogaster. The amount of aluminum in the flies was also reflected by the dietary content of aluminum. Additional dietary aluminum, in the form of aluminum salts, decreased the life span by as much as 20%. Glucose phosphate isomerase (GPI) enzyme activity was measured in two strains of mice, A/HeNNia1 (Gpi-1a) and C57BL/6NNia1 (Gpi-1b). Seven solid tissues (muscle, brain, heart, kidney, liver, lung, spleen) and three cell types (peripheral blood lymphocytes, spleen lymphocytes, erythrocytes) were analyzed for GPI activity in mice from 6 to 36 months of age. No major changes in enzyme activity with age were found. The most striking observation was that A/HeNNia1 mice showed a significantly higher GPI activity than C57BL/6NNia1 mice in four of the solid tissues (muscle, heart, kidney, spleen) and two of the cell types (peripheral blood lymphocytes, erythrocytes) analyzed.

Four neocortical regions (frontal, parietal, temporal, occipital) of 44 cases were impregnated by a silver stain to show neuritic plaques and stained with Congo red to show amyloid plaques. In 11 cases with large numbers of argentophilic plaques these plaques decreased significantly with age while the reverse appeared to be true for the congophilic plaques. In the remaining 34 cases which had few argentophilic plaques the numbers of congophilic plaques increased significantly with age in the parietal and occipital regions. These results suggest that the neuritic component of senile plaques precedes the amyloid cores in their pathogenesis although a causative relationship is not implied.

The degree of multiple pathology in 184 consecutive patients admitted to a geriatric unit was recorded using the 13 commonest conditions present at the time of admission to provide a standard group of disorders for comparison. All of the conditions were chronic in nature, and 35% of the patients had a combination of four or more of these present. Multiple pathology was associated with poor prognosis in terms of mortality and the need for long-term institutional care. Families, including those who were elderly, continued to support most of the frail elderly people even in the presence of multiple pathology. This emphasizes the social implications of multiple pathology and the need for support and information in the management of long-term disability.
