The early kinetics of mitogen-stimulated spleen cells from young adult and aged NMRI mice were investigated. Using RNA synthesis as measured by cytofluorometry and 3H-leucine and 3H-thymidine incorporation as parameters, it was apparent that age-related changes occur at different levels during mitogen stimulation. First, in aged mice fewer cells per 106 spleen cells are being activated by mitogen, and, second, fewer of the activated lymphocytes will proliferate. However, the cells which initiate synthesis of RNA, protein, and DNA apparently behave in identical manners, chronologically, as cells from young adult mice.

The studies concerned age-dependent peculiarities of the vasopressin effect on the hemodynamics and tone of the coronary vessels in dogs, the contraction of the isolated vascular strip in rats, and the hemodynamic and ECG indices in rabbits and in rats. The data obtained indicate the great sensitivity of old vessels to vasopressin. In aging, both humans and animals show a rise of vasopressin concentration in the blood. Age-dependent differences of the vasopressin effect on the kallikrein-kinin system, adenosine metabolism, the contents of prostaglandins and cyclic AMP have been established. High sensitivity to vasopressin in combination with its increased concentration in the blood is an important factor that contributes to the development of arterial hypertension and ischemic heart disease.

Aging mice showed a retrograde amnesia for passive avoidance behavior when a single electroconvulsive shock (ECS) was given at post-training intervals up to 1 h, whereas adult mice showed no significant amnesia when ECS was given 10 min or more after training. Catecholamines (norepinephrine and dopamine), but not serotonin (5-HT) changed with aging in the several brain regions, but only 5-HT was significantly changed (elevated) by ECS. Protein synthesis was significantly inhibited by ECS, notably in limbic structures. The increased susceptibility of aging mice to the amnesic effect of ECS could be related to aging-related changes in the rate of memory consolidation or to the duration of neural chemical changes that disrupt processes such as consolidation, storage, and/or retrieval of the memory trace.

A clinicophysiological investigation of motor control was carried out in 199 apparently healthy, socially active elderly (aged 60–69 years) and long-living (90 years and over) subjects in order to establish the peculiarities of the motor sphere specific to age-related changes of the nervous system. Analyzing the experimentally induced state of readiness (intention) before a spontaneous movement, we found an increase with age in the latent period of the muscle intentional activity (IA) parallel to an increase in the latent period of the spontaneous movement, a decrease in IA amplitude with more frequent structural deviations of the EMG in the prestarting period and a reduction of the required IA selectiveness. The described changes in the organization of readiness for a spontaneous movement seemed to be related with age impairment of supraspinal (mainly corticospinal) influences and may be used for an explanation of a number of clinical peculiarities of human motor control in late ontogenesi

Theophylline kinetics were evaluated in 6 elderly (age 65–75 years) and 5 young (age 22–28 years), nonsmoking males following the oral administration of 200 mg of Elixo-phylline®. Theophylline serum concentration in the postabsorptive phase declined in a monoexponential manner. Total body clearance, elimination half-life, volume of distribution and protein binding were determined. Medication histories were obtained from 3,192 ambulatory elderly persons participating in a health screening program. A total of 2,009 (63%) women and 1,183 (37%) men were included in the drug use study. Flurazepam, pentobarbital and the combination of amo-barbital and secobarbital were used by 4.5, 0.6 and 0.4% of participants, respectively, and these three agents accounted for 87.6% of all prescribed hypnotic drugs. Diazepam, mepro-bamate and chlordiazepoxide were the most commonly used minor tranquilizers. Barbiturates were contained in nearly one fourth of minor tranquilizers used by these elderly subjects. Less than 1% (0.5%) of the participants in this program reported the use of a neuroleptic. Approximately 3% of the participants reported the use of an antidepressant drug. Nearly 90% of participants who reported the daily use of a hypnotic agent stated they had been using the product on a daily basis for longer than 1 year.
