The main cyclo-oxygenase-dependent arachidonic acid (AA) derivatives, i.e. prostaglandin E2 (PGE2) and thromboxane A2 (TXA2), have been measured by radioimmunoassay in platelet-free cultures of human monocytes from young and old subjects, in presence and in absence of activating substances (10% fetal calf serum). No difference was found between cells from the two groups as far as the production of PGE2 and TXB2 (stable metabolite of TXA2) was concerned, at variance with reported data in young and old experimental animals. The addition to the cultures of exogenous AA caused a reorientation of cyclic endoperoxide metabolism resulting in a consistent decrease of the ratio TXB2/PGE2, but only in monocytes from young subjects. The data are discussed with respect to the claimed role of prostaglandins in the age-related immune derangement which is present in aged humans.

Using several models of multivariate analysis, it is found that age per se is a specific determinant on the in vitro lymphocyte response to phytohemagglutinin A in a random sample of 30 diseased patients. Depending upon the used model, sex and lymphocyte subsets appear as other significant variables. The multiple regression coefficients reach 0.85. A 2% fall per decade is expected for phytohemagglutinin A responsiveness expressed in In cpm.

Continuous monitoring of emitted carbon dioxide (VCO2) was taken as an index of respiratory exchanges in male Sprague-Dawley rats of different ages (31–814 days) submitted to a light (100 lx)-dark (LD) 06.00–18.00 h alternation at a temperature of 20 °C and with food and water ad libitum. In grouped 82- and 426-day-old rats harmonic and spectral analyses show that aging decreases the amplitudes of the circadian (τ = 24 h) respiratory rhythms, decreases the amplitudes and modifies the phases of the ultradian respiratory rhythms of mean and great (10 min < τ < 24 h) periods. Male rats aged 6, 19, or 33 months were trained successively in one- and two-way avoidance tasks. The one-way avoidance test consisted of up to 30 trials given in a single session with the conditional stimulus (CS; 14-kHz tone) presented for either 3 s or for 10 s in separate groups. Senescent rats performed poorest, middle-aged rats intermediately, and young adult rats best. Failure of the longer CS to yield better acquisition than the short CS in the senescent group suggested that the age-related deficit probably did not result from slower responding. In subsequent shuttle box training there was no appreciable age difference in achieving the learning criterion. A compound visual-auditory CS was used, and in further evaluation of well-trained rats it was found that the auditory component was much more effective than the visual component in eliciting avoidance. However, this differential effect of the two stimuli was much weaker in the senescent group than in the young adult group. Nonetheless, these same senescent and adult rats readily learned to make avoidance responses using only the auditory CS, demonstrating that this was an effective stimulus for all age groups.

The effect of ageing on the contractile properties of the first dorsal interosseous muscle was investigated. A group of 42 healthy male subjects between the ages of 10 and 80 years took part in the study. The time to peak tension was significantly prolonged with increasing age but no evidence was found of an extension of the half relaxation time. When left and right hands were compared no significant differences were found. The results are discussed in relation to the changes previously observed in ageing muscle and may indicate a general age change of the muscle or the loss of faster twitch fibres.

The range of plasma concentrations in 177 and erythrocyte magnesium concentrations in 104 elderly patients was found to be similar to that for healthy adults. No significant difference was observed between the sexes or between patients taking diuretics and those not receiving diuretic therapy. The latter finding is contrary to some previously reported studies.

The biological rhythms of rectal temperature were documented in young (circadian variations) and elderly (circadian and seasonal variations) human subjects either in apparent good health or suffering from senile dementia of Alzheimer type (SDAT). All the subjects were synchronized. Data obtained showed a decrease of the body core temperature rhythm amplitude in the healthy elderly for each documented season but not in patients with SDAT. Seasonal variations in these rhythms were observed in these elderly groups of persons.

Thirty-five percent of a sample of 100 elderly patients suffering from a degree of cognitive impairment were found to have urinary incontinence. They were significantly older, had lower memory and information scores and were more disorientated, hyperactive, aphasic, apraxic and socially incompetent than their continent counterparts.
