The chronic treatment of female C3H/Sn mice with phenformin (2 mg/day) and diphenylhydantoin (2 mg/day) prolonged mean life span by 23 and 25%, respectively, and decreased spontaneous tumour incidence by 4.0 and 2.3 times, respectively. The chronic treatment of mice with L-dopa (2 mg/day) did not change these parameters and decreased the multiplicity of mammary tumours. The mechanisms of the drug action on mouse life span and tumour incidence are discussed.

The respiratory rate of liver homogenate in the male garden lizard showed a decline with advancing age. It was shown that kallikreinogen content was decreased in blood of old rabbits, kallikrein activity was somewhat increased, kininase activity was significantly decreased. In old animals, adenosine metabolism was activated, this being evident from the rise of 5′-nucleotidase and adenosine deaminase activity in blood and myocardium. Hypothalamic stimulation resulted in significant activation of the kallikrein-kinin system and sharp increase of kallikrein activity, the shifts being less marked in old animals than in adults. Cerebral blood flow, studied by means of a diffusible indicator, remains unchanged in the aging process (developmental changes). On the contrary, brain activity, evaluated by deoxyglucose uptake, is significantly reduced. Papaverine, which increases cerebral blood flow, and vincamine, slightly but significantly, act on this parameter in the older animals.

The effects of centrally and systemically administered L-leucine and L-tyrosine on ovarian function were studied in the old female rat. Systemically administered L-tyrosine reinstated regular vaginal cycling and ovulation, whereas systemically administered L-leucine did not. When administered centrally, through an indwelling cannula in the medial preoptic area, neither L-tyrosine nor L-leucine altered the ovarian or vaginal condition of the old female. When these females were subsequently treated with L-dopa, both systemic and central application were found to reinstate ovarian cycles. Potential mechanisms of action at the level of CNS monoaminer-gic neurotransmission are discussed.

Serum urate levels were marginally higher in 11 nonbacteriuric elderly men (5.1 ± 1.8 mg%) than in 24 nonbacteriuric women (3.9 ± 1.6 mg%) of the same age. Bacteriuric elderly subjects (6 men, 7 women) had lower levels, these being closely similar in the two sexes (3.4 and 3.6 mg%, respectively). Significant reductions of CIn, Ccreat, and TMPAH were observed for bacteriuric men, but not for bacteriuric women. Further, the CUrate: CIn ratio was higher in bacteriuric men and in women, bacteriuric or not, and was not depressed during the TmPAH periods. Increased daily intake of protein from 60 to 75 g was associated with higher serum urate levels in a larger group of subjects. We conclude that: (1) women, even at the age of 80 have a lower serum urate, related to a higher CUrate; (2) bacteriuria increases the ratio CUrate: CIn, particularly in men; (3) secretory mechanisms for urate differ from those of p-aminohippurate in the human, and (4) dietary protein overrides the effect of bacteriuria on serum urate without affecting the male:female rati

Urinary incontinence was studied in 11 elderly patients who were bedridden for more than 1 year in a home for the aged. The time when voiding occurred, the amount of urine at each voiding, the frequency and the interval between urinations were recorded for two periods of 3 days. Diurnal rhythm of water excretion disappeared or was reversed in most of the subjects. The amount of urine voided at one time varied considerably from time to time in the same subjects ranging from less than 50 to more than 200 ml, and this was observed during both the day and night. The average frequency of incontinent voiding was seven to eight times per day. Suggestions are made to apply the present results in the care of urinary incontinence in the aged.
